Robust transcriptional indicators of immune cell death revealed by spatiotemporal transcriptome analyses

Recognition of a pathogen by the plant immune system often triggers a form of regulated cell death traditionally known as the hypersensitive response(HR).This type of cell death occurs precisely at the site of pathogen recognition,and it is restricted to a few cells.Extensive research has shed light on how plant immune receptors are mechanistically activated.However,two central key questions remain largely unresolved:how does cell death zonation take place,and what are the mechanisms that underpin this phenomenon?Consequently,bona fide transcriptional indicators of HR are lacking,which prevents deeper insight into its mechanisms before cell death becomes macroscopic and precludes early or live observation.In this study,to identify the transcriptional indicators of HR we used the paradigmatic Arabidopsis thaliana–Pseudomonas syringae pathosystem and performed a spatiotemporally resolved gene expression analysis that compared infected cells that will undergo HR upon pathogen recognition with bystander cells that will stay alive and activate immunity.Our data revealed unique and time-dependent differences in the repertoire of differentially expressed genes,expression profiles,and biological processes derived from tissue undergoing HR and that of its surroundings.Furthermore,we generated a pipeline based on concatenated pairwise comparisons between time,zone,and treatment that enabled us to define 13 robust transcriptional HR markers.Among these genes,the promoter of an uncharacterized AAA-ATPase was used to obtain a fluorescent reporter transgenic line that displays a strong spatiotemporally resolved signal specifically in cells that will later undergo pathogen-triggered cell death.This valuable set of genes can be used to define cells that are destined to die upon infection with HR-triggering bacteria,opening new avenues for specific and/or high-throughput techniques to study HR processes at a single-cell level.

BACKGROUND The impact of social determinants of health in allogeneic transplant recipients in low-and middle-income countries is poorly described.This observational study analyzes the impact of place of residence,referring institution,and transplant cost coverage(out-of-pocket vs government-funded vs private insurance)on outcomes after allogeneic hematopoietic stem cell transplantation(alloHSCT)in two of Mexico's largest public and private institutions.AIM To evaluate the impact of social determinants of health and their relationship with outcomes among allogeneic transplant recipients in Mexico.METHODS In this retrospective cohort study,we included adolescents and adults≥16 years who received a matched sibling or haploidentical transplant from 2015-2022.Participants were selected without regard to their diagnosis and were sourced from both a private clinic and a public University Hospital in Mexico.Three payment groups were compared:Out-of-pocket(OOP),private insurance,and a federal Universal healthcare program“Seguro Popular”.Outcomes were compared between referred and institution-diagnosed patients,and between residents of Nuevo Leon and out-of-state.Primary outcomes included overall survival(OS),categorized by residence,referral,and payment source.Secondary outcomes encompassed early mortality,event-free-survival,graft-versus-host-relapse-free survival,and non-relapse-mortality(NRM).Statistical analyses employed appropriate tests,Kaplan-Meier method,and Cox proportional hazard regression modeling.Statistical software included SPSS and R with tidycmprsk library.RESULTS Our primary outcome was overall survival.We included 287 patients,n=164 who lived out of state(57.1%),and n=129 referred from another institution(44.9%).The most frequent payment source was OOP(n=139,48.4%),followed by private insurance(n=75,26.1%)and universal coverage(n=73,25.4%).No differences in OS,event-free-survival,NRM,or graft-versus-host-relapse-free survival were observed for patients diagnosed locally vs in another institution,nor pat

Heating of nanoparticles and their environment by laser radiation and applications

This review considers the fundamental dynamic processes involved in the laser heating of metal nanoparticles and their subsequent cooling.Of particular interest are the absorption of laser energy by nanoparticles,the heating of a single nanoparticle or an ensemble thereof,and the dissipation of the energy of nanoparticles due to heat exchange with the environment.The goal is to consider the dependences and values of the temperatures of the nanoparticles and the environment,their time scales,and other parameters that describe these processes.Experimental results and analytical studies on the heating of single metal nanoparticles by laser pulses are discussed,including the laser thresholds for initiating subsequent photothermal processes,how temperature inﬂuences the optical properties,and the heating of gold nanoparticles by laser pulses.Experimental studies of the heating of an ensemble of nanoparticles and the results of an analytical study of the heating of an ensemble of nanoparticles and the environment by laser radiation are considered.Nanothermometry methods for nanoparticles under laser heating are considered,including changes in the refractive indices of metals and spectral thermometry of optical scattering of nanoparticles,Raman spectroscopy,the thermal distortion of the refractive index of an environment heated by a nanoparticle,and thermochemical phase transitions in lipid bilayers surrounding a heated nanoparticle.Understanding the sequence of events after radiation absorption and their time scales underlies many applications of nanoparticles.The applicationﬁelds for the laser heating of nanoparticles are reviewed,including thermochemical reactions and selective nanophotothermolysis initiated in the environment by laser-heated nanoparticles,thermal radiation emission by nanoparticles and laser-induced incandescence,electron and ion emission of heated nanoparticles,and optothermal chemical catalysis.Applications of the laser heating of nanoparticles in laser nanomedicine are of particular interest.Sign

Seamless morphing trailing edge flaps for UAS-S45 using high-fidelity aerodynamic optimization

The seamless trailing edge morphing flap is investigated using a high-fidelity steady-state aerodynamic shape optimization to determine its optimum configuration for different flight conditions,including climb,cruise,and gliding descent.A comparative study is also conducted between a wing equipped with morphing flap and a wing with conventional hinged flap.The optimization is performed by specifying a certain objective function and the flight performance goal for each flight condition.Increasing the climb rate,extending the flight range and endurance in cruise,and decreasing the descend rate,are the flight performance goals covered in this study.Various optimum configurations were found for the morphing wing by determining the optimum morphing flap deflection for each flight condition,based on its objective function,each of which performed better than that of the baseline wing.It was shown that by using optimum configuration for the morphing wing in climb condition,the required power could be reduced by up to 3.8%and climb rate increases by 6.13%.The comparative study also revealed that the morphing wing enhances aerodynamic efficiency by up to 17.8%and extends the laminar flow.Finally,the optimum configuration for the gliding descent brought about a 43%reduction in the descent rate.

Dynamics of Low-Viscosity Liquids Interface in an Unevenly Rotating Vertical Layer

The behavior of two immiscible low-viscosity liquids differing in density and viscosity in a vertical flat layer undergoing modulated rotation is experimentally studied.The layer has a circular axisymmetric boundary.In the absence of modulation of the rotation speed,the interphase boundary has the shape of a short axisymmetric cylinder.A new effect has been discovered,under the influence of rotation speed modulation,the interface takes on a new dynamic equilibrium state.A more viscous liquid covers the end boundaries of the layer in the form of thin films,which have the shape of round spots of almost constant radius;with increasing amplitude of the velocity modulation,the wetting boundary expands.It is found that upon reaching the critical amplitude of oscillations,the film of a viscous liquid loses stability,and the outer edge of the wetting spot collapses and takes on a feathery structure.It is shown that this threshold is caused by the development of the Kelvin-Helmholtz oscillatory instability of the film.The spreading radius of a spot of light viscous liquid and its stability are studied depending on the rotation rate,amplitude,and frequency of rotation speed modulation.The discovered averaged effects are determined by different oscillatory interaction of fluids with the end-walls of the cell,due to different viscosities.The effect of films forming can find application in technological processes to intensify mass transfer at interphase boundaries.

The relationship between morphology and flight in Drosophila:a study of two pairs of sibling species from a natural population

Insect flight is a complex trait involved in different behaviors,from the search for sexual partners,food,or breeding sites.Many studies have postulated the adaptive advantages of certain morphological traits in relation to increased flight capacity,such as low values of wing loading or high values of wing:thorax ratio and wing-aspect ratio.However,few studies have evaluated the relationship between variables related to flight and morphological traits in Drosophila.This work aimed to study morphological traits in males and females of two pairs of sibling species:Drosophila buzzati Patterson and Wheeler-Drosophila koeferae Fontdevila and Wasserman,and Drosophila melanogaster Meigen-Drosophila simulans Sturtevant,and to analyze its relationship with flight.We detected the highest proportion of flight time in D.koepferae and D.simulans compared to D.buzzati and D.melanogaster,respectively.Our results also revealed sexual dimorphism,with males exhibiting a higher proportion of flight time than females.Surprisingly,we did not find a general pattern to explain the relationship between morphology and the proportion of flight time because associations varied depending upon the analyses(considering all groups together or each sex-species combination separately).Moreover,these associations explained a low percentage of variation,suggesting that other nonmorphological components related to flight,such as physiological variables,should be taken into account.This work allowed us to show the variability and complexity of an aspect of flight,suggesting that the adaptive role of the morphological traits studied might have been overestimated.

Using deep neural networks coupled with principal component analysis for ore production forecasting at open-pit mines

Ore production is usually affected by multiple influencing inputs at open-pit mines.Nevertheless,the complex nonlinear relationships between these inputs and ore production remain unclear.This becomes even more challenging when training data(e.***.truck haulage information and weather conditions)are massive.In machine learning(ML)algorithms,deep neural network(DNN)is a superior method for processing nonlinear and massive data by adjusting the amount of neurons and hidden layers.This study adopted DNN to forecast ore production using truck haulage information and weather conditions at open-pit mines as training data.Before the prediction models were built,principal component analysis(PCA)was employed to reduce the data dimensionality and eliminate the multicollinearity among highly correlated input variables.To verify the superiority of DNN,three ANNs containing only one hidden layer and six traditional ML models were established as benchmark models.The DNN model with multiple hidden layers performed better than the ANN models with a single hidden layer.The DNN model outperformed the extensively applied benchmark models in predicting ore production.This can provide engineers and researchers with an accurate method to forecast ore production,which helps make sound budgetary decisions and mine planning at open-pit mines.

New approach for particle size and shape analysis of iron-based oxygen carriers at multiple oxidation states

One of the crucial issues in the chemical looping technology lies in its bed material:the oxygen carrier.Particle size analysis of an oxygen carrier is important since in a fluidized bed the material can only work well within a specific size range.While the favorable size ranges for oxygen carrier materials have already been reported,none of the published studies has analyzed the particle size and shape of oxygen carriers in detail.Furthermore,the effect of oxygen carriers'oxidation degree on such properties has not been considered either.This study aimed to report the particle size and shape analysis of five iron-based oxygen carriers,one natural ore,one synthetic material,and three residue products,at different oxidation degrees using dynamic image analysis(DIA).The oxygen carriers were prepared at different mass conversion degrees in a fluidized bed batch reactor.The size distribution,sphericity,and aspect ratio of the oxygen carrier particles were examined experimentally using a Camsizer instrument.Our results show that the DIA method was successfully able to analyze the particle size and shape of our oxygen carriers with satisfying accuracy for comparison.The oxidation state of the investigated materials seems to only affect the particle size and shape of oxygen carriers to a minor extent.However,exposures to redox cycles in a fluidized bed reactor may alter the particle size and shape of most oxygen carriers.

Hydrogen sulfide(H_(2)S) not only presents significant environmental concerns but also induces severe corrosion in industrial equipment,even at low concentrations.Among various technologies,the selective oxidation of hydrogen sulfide(SOH_(2)S) to elemental sulfur(S) has emerged as a sustainable and environmentally friendly solution.Due to its unique properties,iron oxide has been extensively investigated as a catalyst for SOH_(2)S;however,rapid deactivation has remained a significant drawback.The causes of iron oxide-based catalysts deactivation mechanisms in SOH_(2)S,including sulfur or sulfate deposition,the transformation of iron species,sintering and excessive oxygen vacancy formation,and active site loss,are thoroughly examined in this review.By focusing on the deactivation mechanisms,this review aims to provide valuable insights into enhancing the stability and efficiency of iron-based catalysts for SOH_(2)S.
