Decrease of serum carnitine levels in patients with or without gastrointestinal cancer cachexia

AIM： To evaluate the levels of serum carnitine in patients with cancer in digestive organs and to compare them with other cancers in order to provide new insights into the mechanisms of cachexia. METHODS： Fifthy-five cachectic patients with or without gastrointestinal cancer were enrolled in the present study. They underwent routine laboratory investigations, including examination of the levels of various forms of carnitine present in serum （i.e., long-chain acylcarnitine, short-chain acylcarnitine, free carnitine, and total carnitine）. These values were compared with those found in 60 cancer patients in good nutritional status as well as with those of 30 healthy control subjects. RESULTS： When the cachectic patients with gastrointestinal cancer were compared with the cachectic patients without gastrointestinal cancer, the difference was -6.8 μmol/L in free carnitine （P 〈 0.005）, 0.04 μmol/ L in long chain acylcarnitine （P 〈 0.05）, 8.7 μmol/L in total carnitine （P 〈 0.001）. In the cachectic patients with or without gastrointestinal cancer, the difference was 12.2 μmol/L in free carnitine （P 〈 0.001）, 4.60 μmol/L in short chain acylcarnitine （P 〈 0.001）, and 0.60 μmol/L in long-chain acylcarnitine （P 〈 0.005） and 17.4 μmol/L in total carnitine （P 〈 0.001）. In the cachectic patients with gastrointestinal cancer and the healthy control subjects, the difference was 15.5 μmol/L in free carnitine （P 〈 0.001）, 5.2 μmol /L in short-chain acylcarnitine （P 〈 0.001）, 1.0 μmol/L in long chain acylcarnitine （P 〈 0.001）, and 21.8 μmol/L in total carnitine （P 〈 0.001）. CONCLUSION： Low serum levels of carnitine in terminal neoplastic patients are decreased greatly due to the decreased dietary intake and impaired endogenous synthesis of this substance. These low serum carnitine levels also contribute to the progression of cachexia in cancer patients.

Insufficient metabolic energy,in the form of adenosine triphosphate(ATP),and bacterial infections are among the main causes for the development of chronic *** we showed that the physi-ological inorganic polymer polyphosphate(polyP)massively accelerates wound healing both in animals(diabetic mice)and,when incorporated into mats,in patients with chronic ***,we focused on a hydrogel-based gel formulation,supplemented with both soluble sodium polyP(Na-polyP)and amor-phous calcium polyP nanoparticles(Ca-polyP-NP).Exposure of human epidermal keratinocytes to the gel caused a significant increase in extracellular ATP level,an effect that was even enhanced when Na-polyP was combined with ***,it is shown that the added polyP in the gel is converted into a coacervate,leading to encapsulation and killing of *** data on human chronic wounds showed that the administration of hydrogel leads to the complete closure of these *** analysis of biopsies showed an increased granulation of the wounds and an enhanced microvessel *** results indicate that the polyP hydrogel,due to its properties to entrap bacteria and generate metabolic energy,is a very promising formulation for a new therapy for chronic wounds.

AIM：To analyze the visual outcomes and the posterior capsule opacification（PCO）with the new Incise？MJ14intraocular lens（IOL）implanted through a 1.4 mm clear corneal incision（CCI）in patients who underwent bimanual microincision cataract surgery（B-MICS）.METHODS：Eighty eyes which underwent cataract surgery using B-MICS technique performed by the same experienced surgeon were included in the study：40 eyes were implanted with an Incise？MJ14 IOL through a 1.4 mm CCI（group A）without enlargement of the main CCI,while 40 eyes were implanted with an Akreos？MI60 IOL with enlargement of the main CCI to 1.8 mm（group B）.Best corrected visual acuity（BCVA）,astigmatism and endothelial cell loss were evaluated before and after surgery at 7,30d and *** segment-optical coherence tomography（AS-OCT）of CCI was performed at 1,3,7,30d,6 and *** incidence was evaluated at 18mo using EPCO 2000 ***：Mean BCVA improvement and endothelial cell loss were statistically significant at 18mo in both groups with no difference between the two groups;no statistically significant difference in surgically induced astigmatism（SIA）was noticed in the two *** AS-OCT the only significant alterations in the CCI were endothelial gaping and local detachment of Descemet’s membrane at 1 and 7d after surgery;no statistically significant alterations were found at 1,6 and *** score at 18mo was 0.03±0.07for group A and 0.08±0.18 for group B（P=0.11）with no sign of central optic plate invasion in both ***：The implant of the new Incise？MJ14 IOL through a 1.4 mm CCI and B-MICS technique appeared to be a safe and effective procedure with rapid visual *** rate resulted very low and the CCI presented few morphological alterations which were only detectable in the first days postoperatively and achieved fast corneal healing during the long-term follow-up.

Extremely Early Diagnostic Test for Prostate Cancer

Purpose: This article reports the results of a blinded fibre diffraction study of skin samples taken from TRAMP mice and age-matched controls to determine whether changes noted in fibre diffraction studies of human skin were present in these TRAMP mice studies. These mice are bred to progress to Gleeson Type 3 to Type 5 prostate cancer. Methods: Small strips, 1 mm × 5 mm, cut from the mouse skin samples were loaded into cells in the same way as human samples and slightly stretched to remove the crimp. They remained fully hydrated throughout exposure to the synchrotron beam. Results: The added change that was reported for prostate cancer in 2009 was obtained for all TRAMP mice samples, indicating that this change can be read as High Grade Cancer in human diagnostic tests. Discussion: These changes were evident for all 3 and 7 week old TRAMP mice samples but not for any of the control samples. This indicates that the changes in the fibre diffraction patterns appear much earlier than in any other available prostate cancer diagnostic test, as none of these can verify the presence of prostate cancer in the TRAMP mice before 10 weeks of age. The fibre diffraction test is therefore the most accurate and earliest test for high grade prostate cancer.

Hydrogen is considered an attractive alternative to fossil fuels,but only a small amount of it is produced from renewable energy,making it not such a clean energy carrier after *** hydrogen through water electrolysis is promising,but using a cost-effective and high-performing catalyst that has longterm stability is still a *** study exploits,for the first time,the potential of zinc oxide nanoparticles with diverse morphologies as catalysts for the electrocatalytic production of hydrogen from *** morphology of the nanoparticles(wires,cuboids,spheres)was easily regulated by changing the concentration of sodium hydroxide,used as the shape controlling agent,during the *** spherical morphology exhibited the highest electrocatalytic activity at the lowest potential *** spherical nanoparticles had the highest number of oxygen vacancies and lowest particle size compared to the other two morphologies,features directly linked to high catalytic ***,the nanowires were much more stable with repeated ***-functional theory showed that the presence of oxygen vacancies in all three morphologies led to diminished band gaps,which is of catalytic interest.

The antibacterial potential of silver nanoparticles（AgNPs） resulted in their increasing incorporation into consumer,industrial and biomedical ***,human and environmental exposure to AgNPs（either as an engineered product or a contaminant）supports the emergent research on the features conferring them different toxicity *** this study,30 ran AgNPs coated with citrate or poly（ethylene glycol）（PEG） were used to assess the influence of coating on the effects produced on a human hepatoma cell line（HepG2）,namely in terms of viability,apoptosis,apoptotic related genes,cell cycle and cyclins gene *** types of coated AgNPs decreased cell proliferation and viability with a similar toxicity *** the concentrations used（11 and 5 μg/mL corresponding to IC50 and-IC10 levels,respectively） the amount of cells undergoing apoptosis was not significant and the apoptotic related genes BCL2（anti-apoptotic gene）and BAX（pro-apoptotic gene） were both ***,both AgNPs affected HepG2 cell cycle progression at the higher concentration（11 μg/mL） by increasing the percentage of cells in S（synthesis phase） and G2（Gap 2 phase） *** the cell-cycle related genes,the expression of cyclin B1 and cyclin E1 genes were ***,this work has shown that citrate- and PEG-coated AgNPs impact on HepG2 apoptotic gene expression,cell cycle dynamics and cyclin regulation in a similar *** research is needed to determine the properties that confer AgNPs at lower toxicity,since their use has proved helpful in several industrial and biomedical contexts.
