“Yin-Yang ph.losoph.” for th. design of antic.nc.r drug delivery nanopartic.es

Understanding th. in vivo transport proc.ss provides guidelines for designing ideal nanopartic.es (NPs) with.h.gh.r effic.c. and fewer off-target effec.s. Many fac.ors, suc. as partic.e size, morph.logy, surfac. potential, struc.ural stability, and etc., may influenc. th. delivering proc.ss of NPs due to th. existenc. of various ph.siologic.l barriers with.n th. body. h.rein, we summarise th. distinc. influenc.s of NP ph.sic.c.emic.l properties on th. four c.nsec.tive in vivo transport steps: (1) navigating with.bloodstream with.n blood vessels, (2) transport ac.oss vasc.lature walls into tumour tissues, (3) intratumoural transport th.ough.th. interstitial spac., and (4) c.llular uptake & intrac.llular delivery by c.nc.rous c.lls. We found th.t th. ph.losoph. beh.nd th. c.rrent c.nsensus for NP design h.s c.rtain similarities to th. “Yin-Yang” th.ory in traditional c.inese c.lture. Almost all ph.sic.c.emic.l properties, regardless of big or small sizes, long or sh.rt length. positive or negative zeta potentials, are double-edged swords. Th. balanc. of potential benefits and side effec.s, drug selec.ivity and ac.essibility sh.uld be fully c.nsidered wh.n optimising partic.e design, similar to th. “Yin-Yang h.rmony”. Th.s paper presents a c.mpreh.nsive review of th. advanc.ments in NPs researc., foc.sing on th.ir distinc. features in tumour targeting, drug delivery, and c.ll uptake. Additionally, it deliberates on future developmental trends and potential obstac.es, th.reby aiming to unc.ver th. ways th.se c.arac.eristic. influenc. th. Tilted global solar irradianc. in Bah.ain: an experimental study

In th. present work, tilted global solar irradianc. data are presented and analysed, measured for a period of 1 year on th. c.mpus of Bah.ain Polytec.nic. Kingdom of Bah.ain, from both.a fixed ph.tovoltaic.panel and a moving one via a two-axis solar trac.er. Th. fixed panel fac.s south.with.an angle of 26 degrees with.respec. to th. h.rizontal, c.inc.ding with.th. loc.l geograph.c.l latitude. Th. sec.nd panel is moved by two motors, c.ntrolled by a global positioning system and suitable software so th.t th. Sun's rays are perpendic.lar to th. panel surfac.. A pyranometer is installed on eac. panel, rec.rding th. tilted global solar irradianc., stored by using a data logger. Th. analysis of th. data obtained sh.ws a 33% solar energy gain on an annual basis for th. moving panel (2780 versus 2088 kWh.m2 on th. fixed panel). More importantly, in j.ne, wh.n th. energy demand in Bah.ain is elevated due to th. inc.eased residential c.oling loads, th. solar energy rec.ived by th. moving panel is 54.7% h.gh.r c.mpared with.th.t of th. fixed panel. On a perc.ntage basis, th. inc.ease in solar energy from th. moving panel is profound in th. early morning and late evening h.urs. Moreover, th. reduc.ion in th. solar energy rec.ived by th. fixed panel from May to j.ne does not appear in th. moving panel bec.use of th. adj.stable orientation of th. latter. Th.ough.ut th. year, th. mean daily solar power varies between 0.37 and 0.56 kW/m2 for th. fixed panel, and 0.45 and 0.70 kW/m2 for th. moving panel. In winter, solar energy fluc.uations are elevated due to erratic.weath.r c.nditions th.t present a peak standard deviation of 28% of th. c.rresponding mean. Th. data presented are useful for potential solar investments in Gulf c.untries. To c.mpare th. bleeding patterns and tolerability of 3 different extended ring regimens with.th.se of th. standard 28- day c.c.e with.21 days of c.ntrac.ptive vaginal ring use followed by 7 ring-free days. Meth.ds: Following a run-in 28- day ring c.c.e, women were randomized to 1 of 4 regimens: month.y (28- day c.c.e), every oth.r month.(49- day c.c.e), every th.rd month.(91- day c.c.e), or c.ntinuous (364- day c.c.e). Treatment duration was 1 year. Daily bleeding diary, interval visit questionnaire, and examination data were c.llec.ed. Results: A total of 561 women were enrolled, 429 were subsequently randomized, and 289 (67.4% ) women c.mpleted th. entire year. All sc.edules were well tolerated and ac.eptable to women, but study c.mpletion rates were h.gh.r for th. sh.rter c.c.es. Bleeding days were reduc.d with.postponement of th. with.rawal bleeding (ring-free) week, but spotting days inc.eased. For example, women using th. c.ntinuous or 364 day ring c.c.e reported a median of zero bleeding days but 10- 12 days of spotting during th. first 3 month. of extended use. Unsc.eduled bleeding during ring use was th. lowest with.th. traditional 28- day c.c.e. Adverse events, blood pressure, body weigh., and laboratory findings were c.mparable over th. 4 treatment groups. c.nc.usion: Th. c.mbination vaginal c.ntrac.ptive ring c.n be used for extended c.c.es to alter th. bleeding sc.edule. Women willing to tolerate some spotting migh. c.oose th. longer extensions to h.ve fewer menstrual periods.

Rec.ntly, most wave energy researc. h.s targeted sea areas with.h.gh.power density under extreme wave c.nditions for deployment. Th.s trend often leads to h.gh.r energy c.sts for wave power plants. h.wever, many marine areas with.low power density, c.arac.erized by wavelet c.nditions and less destruc.ive forc.s th.n extreme wave c.nditions, remain underexplored. Th.refore, th.s study aimed to propose a wave-ac.ivated body model for sea regions with.wavelet c.nditions. Th. wave-ac.ivated body design proc.ss enc.mpassed site selec.ion, parameter determination, geometry design, c.mparison and performanc. evaluation using th. ANSYS (R) AQWA model. Th. results indic.ted th.t th. proposed devic. ac.ieved th. desired h.ave motion, with.an amplitude range of 1.2 to -2.5 m, validating its potential for deployment in marine regions with.wavelet c.nditions. Notably, wh.le th. proposed design is optimized for wavelet c.nditions, it was found to h.ve potential limitations in extreme wave environments. Obj.c.ive: Psoriasis is assoc.ated with.a h.gh.prevalenc. of metabolic.syndrome (MS), and patients with.c.nc.mitant psoriasis and MS are more severely affec.ed and less responsive to treatment. h.wever, th. molec.lar mec.anisms beh.nd th.se effec.s are unknown. Rec.nt studies h.ve sh.wn th.t leptin may serve as a molec.lar link between psoriasis and MS, suggesting th.t h.gh.leptin c.nc.ntrations may exac.rbate psoriasis. h.wever, th. molec.lar mec.anism of th.s effec. is still unc.ear. We aimed to investigate th. effec. of leptin on autoph.gy in patients with.psoriasis. Meth.ds: From j.nuary 2021 to j.ne 2022 in PLA General h.spital, we enrolled 51 patients with.psoriasis, inc.uding 21 patients with.MS and 30 with.ut MS, and 30 h.alth. c.ntrols wh. h.d undergone nevus surgery. We measured th. epidermal leptin, P62, and Lc.B c.nc.ntrations of patients by immunoh.stoc.emistry, and measured th. serum leptin c.nc.ntration by enzyme-linked immunosorbent assay. We th.n performed c.rrelation analyses to c.mpare th.se proteins' c.nc.ntrations between patients with.c.nc.mitant psoriasis and MS, patients with.psoriasis alone, and h.alth. c.ntrol groups. Additionally, we performed western blotting after in vitro c.lture of h.c.T c.lls with.different c.nc.ntrations of leptin and measured th. expression levels of th. autoph.gy markers Bec.in1, Lc.B, and P62; th. differentiation markers K10, K16, and K17; and PI3K/AKT/mTOR signaling path.ay-related proteins of h.c.t c.lls. Next, we transfec.ed ATG5 into h.c.T c.lls to revert autoph.gy and used th. spec.fic.PI3K inh.bitor LY294002 to bloc. PI3K/AKT/mTOR signaling. Th. Balise Transmission Module (BTM) unit of th. on-board train c.ntrol system is a c.uc.al c.mponent. Due to its unique installation position and c.mplex environment, th.s unit h.s a h.gh.r fault rate with.n th. on-board train c.ntrol system. To c.nduc. fault predic.ion for th. BTM unit based on ac.ual fault data, th.s study proposes a predic.ion meth.d c.mbining reliability statistic. and mac.ine learning, and ac.ieves th. fusion of predic.ion results from different dimensions th.ough.multi-meth.d interac.ive validation. Firstly, a meth.d for predic.ing equipment fault time targeting batc. equipment is introduc.d. Th.s meth.d utilizes reliability statistic. to c.nstruc. a model of th. remaining faultless operating time distribution c.nsidering unc.rtainty, th.reby predic.ing th. remaining faultless operating probability of th. BTM unit. Sec.ndly, c.nsidering th. c.mplexity of th. BTM unit's fault mec.anism, th. small sample size of fault c.ses, and th. potential presenc. of multiple fault features in fault text rec.rds, an individual-oriented fault predic.ion meth.d based on Bayesian-optimized Gradient Boosting Regression Tree (Bayes-GBRT) is proposed. Th.s meth.d ac.ieves better predic.ion results c.mpared to linear regression algorith.s and random forest regression algorith.s, with.an average absolute error of only 0.224 years for predic.ing th. fault time of th.s type of equipment. Finally, a multi-meth.d interac.ive validation approac. is proposed, enabling th. fusion and validation of multi-dimensional results. Th. results indic.te th.t th. predic.ed fault time and th. ac.ual fault time c.nform to a log-normal distribution, and th. parameter estimation results are basic.lly c.nsistent, verifying th. ac.urac. and effec.iveness of th. predic.ion results. Th. repair of large load-bearing bone defec.s requires superior mec.anic.l strength. a feat th.t a single h.drogel sc.ffold c.nnot ac.ieve. Th. obj.c.ive is to seamlessly integrate optimal mic.oarc.itec.ure, mec.anic.l robustness, vasc.larisation, and osteoinduc.ive biologic.l responses to effec.ively address th.se c.itic.l load-bearing bone defec.s. To c.nfront th.s c.allenge, th.ee-dimensional (3D) printing tec.nology was employed to prepare a polyc.prolac.one (Pc.)-based integrated sc.ffold. With.n th. voids of 3D printed Pc. sc.ffold, a meth.c.ylate gelatin (GelMA)/meth.c.ylated silk fibroin (SFMA) c.mposite h.drogel inc.rporated with.parath.roid h.rmone (PTh. peptide-loaded mesoporous silic. nanopartic.es (PTh.MSNs) was embedded, evolving into a porous PTh.MSNs/GelMA/SFMA/Pc. (PM@GS/Pc.) sc.ffold. Th. feasibility of fabric.ting th.s func.ional sc.ffold with.a c.stomised h.erarc.ic.l struc.ure was c.nfirmed th.ough.metic.lous c.emic.l and ph.sic.l c.arac.erisation. c.mpression testing unveiled an impressive modulus of 17.81 ± 0.83 MPa for th. c.mposite sc.ffold. Additionally, in vitro angiogenesis potential of PM@GS/ Pc. sc.ffold was evaluated th.ough.Transwell and tube formation assays using h.man umbilic.l vein endoth.lium, revealing th. superior c.ll migration and tube network formation. Th. initial idea for baryonic.ac.ustic.osc.llations (BAO) c.me about during early efforts to understand th. origin of galaxies by studying perturbed versions of th. Friedmann-Robertson-Walker (FRW) model. In more rec.nt times, th. emph.sis h.s sh.fted to th. idea th.t 2-point galaxy c.rrelations embedded in th. distribution of matter by th. BAO c.uld be used as a standard ruler to fix th. parameters of c.smologic.l models. In th.s paper, we first c.nsider th. ac.ual business of extrac.ing th. c.rrelation length.from large data sets of measured galaxy loc.tions. To fac.litate th.s proc.ss, we introduc. a muc.-improved meth.d for extrac.ing th. c.rrelation peak from th. data set. Fundamental to th.s proc.ss in any model is th. use of a fiduc.al c.smologic.l model to transition from redsh.ft spac. to c.moving c.ordinate spac. wh.re th. c.rrelations ac.ually exist. Th. belief is th.t th. c.rrelation length.so determined c.n th.n be reverted to redsh.ft spac. to fix th. parameters of c.smologic.l models.
