Impact of humic acids on the colonic microbiome in healthy volunteers

AIMTo test the effects of humic acids on innate microbial communities of the *** followed the effects of oral supplementation with humic acids (Activomin®) on concentrations and composition of colonic microbiome in 14 healthy volunteers for 45 d. 3 × 800 mg Activomin® were taken orally for 10 d followed by 3 × 400 mg for 35 d. Colonic microbiota were investigated using multicolor fluorescence in situ hybridization (FISH) of Carnoy fixated and paraffin embedded stool cylinders. Two stool samples were collected a week prior to therapy and one stool sample on days 10, 31 and 45. Forty-one FISH probes representing different bacterial groups were *** sum concentration of colonic microbiota increased from 20% at day 10 to 30% by day 31 and remained stable until day 45 (32%) of humic acid supplementation (P < 0.001). The increase in the concentrations in each person was due to growth of preexisting groups. The individual microbial profile of the patients remained unchanged. Similarly, the bacterial diversity remained stable. Concentrations of 24 of the 35 substantial groups increased from 20% to 96%. Two bacterial groups detected with Bac303 (Bacteroides) and Myc657 (mycolic acid-containing Actinomycetes) FISH probes decreased (P > 0.05). The others remained unaffected. Bacterial groups with initially marginal concentrations (< 0.1 × 109/ml) demonstrated no response to humic acids. The concentrations of pioneer groups of Bifidobacteriaceae, Enterobacteriaceae and Clostridium difficile increased but the observed differences were statistically not *** acids have a profound effect on healthy colonic microbiome and may be potentially interesting substances for the development of drugs that control the innate colonic microbiome.

摘要：茄病镰刀菌（Fusarium solani ***）是一种土生细菌，通过侵染大豆根系引发猝死综合症。利用温室盆栽试验和须根培养试验研究了接种茄孢镰刀菌对13个不同基因型大豆的影响。结果表明：接种后所有盆栽大豆主根都有明显深褐色的侵染病斑；移植后21d测定了盆栽植株叶部发病程度，Peking表现最为严重，然后依次为Spencer，Ripley，P3981，Williams82，Essex，Forrest，Iroquois，PI520733，Hartwig，PI567650B，Jack，和PI567374。叶部发病程度与冠高（r=-0．422，P=0．0018）、冠重（r=-0．857，P〈0．0001）和根重（r=-0．732，P〈0．0001）呈显著负相关。主根病斑长度与叶部发病程度没有相关性，表明大豆对病原菌的抗性不能仅通过根系得到充分控制。对培养的大豆须根接种茄病镰刀菌菌丝体10d后，不同基因型大豆的菌落直径的变化范围为17—40mm，差异显著（P=0．05），其中Spencer和Peking须根上的菌落直径显著（P=0．05）大于PI567374和PI520733。对Spencer和PI567374的须根接种10灿茄病镰刀菌常量成分悬浮液，10d后菌落直径分别为50和38mm，差异显著（P=0．05）。通常，不同基因型大豆间茄病镰刀菌在培养须根上的生长与整株的症状间有一定的相关性，但不总是这样，这是因为即使根系对毒素产生抗性来减少叶部病害症状，但并不是所有的大豆都表现出明显的根系抗性。

To compare a novel, fully synthetic, polyurethane based glue (MAR-1) to fibrin sealant in a partial liver resection rat model. METHODSAfter 50% resection of the lateral left liver lobe in male Wistar rats (n = 7/group/time point), MAR-1, Fibrin or NaCl was applied. After 14, 21 and 90 postoperative days, sealant degradation, intra-abdominal adhesions were scored, and histological examination of liver tissue was performed. RESUlTS(Mean ± SEM) (MAR-1 vs Fibrin vs NaCl). Bleeding mass was significantly higher in NaCl (3.36 ± 0.51 g) compared to MAR-1 (1.44 ± 0.40 g) and Fibrin (1.16 ± 0.32 g). At 14 and 90 d, bleeding time was significantly lower in MAR-1 (6.00 ± 0.9 s; 13.57 ± 3.22 s) and Fibrin (3.00 ± 0.44 s; 22.2 ± 9.75 s) compared to NaCl (158.16 ± 11.36 s; 127.5 ± 23.3 s). AlT levels were significantly higher in MAR-1 (27.66 ± 1 U/l) compared to Fibrin (24.16 ± 0.98 U/l) and NaCl (23.85 ± 0.80 U/l). Intrabdominal adhesions were significantly lower in MAR-1 (11.22% ± 5.5%) compared to NaCl (58.57% ± 11.83%). Degradation of the glue was observed and MAR-1 showed almost no traces of glue in the abdominal cavity as compared to the Fibrin (10% ± 5% 14 d; 7% ± 3% 21 d). Survival showed no significant differences between the groups. CONClUSIONCompared to Fibrin, MAR-1 showed similar hemostatic properties, no adverse effects, and is biocompatible. Further studies on adhesion strength and biodegradability of synthetic sealants are warranted.

Digital maps of soil properties are now widely ***-users now can access several digital soil mapping(DSM)products of soil properties,produced using different models,calibration/training data,and covariates at various spatial scales from global to ***,there is an urgent need to provide easy-to-understand tools to communicate map uncertainty and help end-users assess the reliability of DSM products for use at local *** this study,we used a large amount of hand-feel soil texture(HFST)data to assess the performance of various published DSM products on the prediction of soil particle size distribution in Central *** tested four DSM products for soil texture prediction developed at various scales(global,continental,national,and regional)by comparing their predictions with approximately 3200 HFST observations realized on a 1:50000 soil survey conducted after release of these DSM *** used both visual comparisons and quantitative indicators to match the DSM predictions and HFST *** comparison between the low-cost HFST observations and DSM predictions clearly showed the applicability of various DSM products,with the prediction accuracy increasing from global to regional *** simple evaluation can determine which products can be used at the local scale and if more accurate DSM products are required.

A Rare Case of Diaphragmatic Mass: The Key to Diagnosis of a lung Adenocarcinoma

lung cancer is a leading cause of death due to malignancy worldwide. Diaphragmatic metastasis secondary to primary lung cancer is rare. In the literature, only a few cases have been described as the majority of cases of diaphragmatic metastasis which are secondary to gastrointestinal neoplasia. The authors report a rare case of diaphragmmatic metastasis from a primary adenocarcinoma of the lung in a 76-year-old female patient with no history of active or passive smoking with progressive worsening fatigue, asthenia, malaise, and unquantified weight loss with three months of evolution, associated with pleuritic chest pain and edema of the lower limbs for fifteen days previous to hospital admission. Chest x-ray showed elevation of the right hemi-diaphragm and thoracic-abdominal-pelvic computed tomography revealed a compl.x multilobuled mass with neoformative characteristics intersecting the diaphragm and invading the dome liver and the right lung region. Biopsy of the mass was performed whose histological examination was consistent with moderately differentiated adenocarcinoma. Immunohistochemical examination was positive for cytokeratin 7 and negative forcytokeratin 20, suggesting primary lung tumor. A positron emission tomography was performed without uptake in other locations. Diagnosis of adenocarcinoma with diaphragmatic metastasis was then admitted and the patient started chemotherapy with carboplatin and gemcitabine. The patient died six months after the diagnosis. The authors also present a brief theoretical discussion based on the current and scarce literature on this rare entity.

Thymoma is the most common neoplasm of the anterior mediastinum and accounts for one quarter of all mediastinal tumours and half of all anterior mediastinal masses. Pericardial effusion may be present in approximately 20% of cases. This report presents a patient with a previously diagnosed thymoma without surgical indication and with poor response to chemotherapy and radiotherapy who had a pericardial effusion that likely resulted from the malignancy dissemination to pericardium.

The diagnosis and management of blood culture-negative endocarditis constitute a real clinical challenge and a systemic approach is necessary for a successful outcome. The authors report a case of a female patient aged 26, with previous clinical history of valve disease and heart failure NYHA class II, with decompensation of co-morbidities associated with fever, productive cough, nonselective anorexia and unquantified weight loss with one month of evolution with negative blood cultures. Transesophageal echocardiography revealed the presence of severe pulmonary hypertension and mitral valve vegetations in the context of positive serology for Q fever. Herein, the authors report a case of blood culture-negative endocarditis and present a brief review on the management of this medical condition. We highlighted the diagnostic difficulties of blood culture-negative endocarditis and subacute clinical presentation, which sometimes present with fever of unknown origin and complaints of deterioration of cardiac function, thus creating a challenging differential diagnosis.

目的:探索体外诱导人类B细胞产生特异性HlA抗体的理想培养条件。方法:以EBV转化致敏肾移植患者B淋巴细胞系(BlCl)为模型,采用ElISA和ElISPOT方法,比较3种培养体系:(1)RPMI1640+抗OPTI单克隆抗体(mAb);(2)RPMI1640+0·1mmol/l非必需氨基酸液+1mmol/l丙酮酸钠+40mg/l转铁蛋白;(3)Hybridoma无血清培养液,以及3种培养条件:(1)对照组(仅培养液),(2)培养液+rIl-4+rIl-10+鼠抗人CD40(α-CD40),(3)培养液+rIl-4+rIl-10+重组人CD40配体(CD40l),人类B细胞产生免疫球蛋白(Ig)和特异性抗HlA抗体的能力。结果:BlCl细胞在Hybridoma SFM培养体系所产生的IgG和IgM浓度分别为25·2～28·1mg/l和7·14～7·95mg/l,高于其他两种培养条件(PlCl细胞在添加了Il-4、Il-10和α-CD40或CD40l的杂交瘤无血清培养液中能产生特异性抗HlA-I抗体,所获得的斑点频数分别为3·06/每104个BlCl细胞和3·55/每104个BlCl细胞,高于对照组(1·77/每104个BlCl细胞)。结论:BlCl细胞在杂交瘤无血清培养液中和添加了Il-4、Il-10和α-CD40或CD40l的培养条件下,提高分泌IgG和IgM的能力和特异性抗HlA-I类分子的抗体。
