Overexpression of the cholesterol-binding protein MLN64 induces liver damage in the mouse

AIM： To examine the in vivo phenotype associated with hepatic metastatic lymph node 64 （MLN64） overexpression.METHODS： Recombinant-adenovirus-mediated MLN64 gene transfer was used to overexpress MLN64 in the livers of C57BL/6 mice. We measured the effects of MLN64 overexpression on hepatic cholesterol content, bile flow, biliary lipid secretion and apoptosis markers. For in vitro studies cultured CHO cells with transient MLN64 overexpression were utilized and apoptosis by TUNEL assay was measured.RESULTS： Livers from ***64-infected mice exhibited early onset of liver damage and apoptosis. This response correlated with increases in liver cholesterol content and bilian/bile acid concentration, and impaired bile flow. We investigated whether liver MLN64 expression could be modulated in a murine model of hepatic injury. We found increased hepatic MLN64 mRNA and protein levels in mice with chenodeoxycholic acid-induced liver damage. In addition, cultured CliO cells with transient MLN64 overexpression showed increased apoptosis.CONCLUSION： In summary, hepatic MLN64 overexpression induced damage and apoptosis in murine livers and altered cholesterol metabolism. Further studies are required to elucidate the relevance of these findings under physiologic and disease conditions.

Effects of fertilization on radial growth of Pinus pinea explored hourly using dendrometers

Background Stone pine(Pinus pinea), a drought-resistant species, has significant socio-economic benefits and increasing interest for the establishment of productive plantations in several countries, especially in a climate change context. Monitoring hourly stem diameter variations contributes to the understanding of the tree-growth response to changes in environmental conditions and management. By monitoring the diameter expansion of tree stems, high-resolution band dendrometers were used to study the development of adult trees growing in a semi-arid coastal environment of central Chile under fertilized and non-fertilized soil conditions through the span of a *** Short cycles( 28 h) occurred at a higher frequency in fertilized trees(16 vs 6). Most of the circadian cycles were regular(24 ± 3 h). The longest cycle duration(59 h) was observed in fertilized trees during spring. In all seasons, each phase of the circadian cycle, especially during the stem diameter increment phase(P3, irreversible growth), started earlier in fertilized than in control trees. P3 duration was significantly longer in fertilized than in control trees in springtime. The maximum shrinkage(P1) was observed in summer for both treatments. Stem diameter increased faster in fer-tilized than in control trees throughout the year, with the highest accumulation occurring in spring and the lowest in autumn. The daily variability pattern showed lower growth under high temperature across *** This study highlights the importance of fertilization in enhancing stone pine diameter growth. This cultural practice should be further explored to contribute to the mitigation of climate change effects in semi-arid environments.

Background Tree regeneration is a key component of resilience because it promotes post-disturbance recovery of forests. Northwestern Patagonia from Argentina is occupied by Nothofagus alpina（Na）, N. obliqua（No）, and N. dombeyi（Nd） forests that grow along an intense precipitation gradient, managed throughout shelterwood silvicultural system by technicians of the Lanin Natural Reserve. The objective of this study was to evaluate the influence of seeding cuttings over the dynamics of Nothofagus mixed forests across landscape（precipitation gradient） based mainly on the composition and abundance of tree regeneration, permanent sampling plots and generalized linear mixed models. In particular, we analysed:（i） the structure of sexual and asexual regeneration during 20 years after harvest（the dynamics of managed forests）, and（ii） the structure of sexual regeneration in primary and managed forests after > 20 years of harvest（the effect of silviculture）.Results Nd was the most abundant species in the regeneration of managed forests during all periods in both sites despite its lower contribution to the adult cohort. During the 10–20 years period after harvest, the humid site exhibited higher regeneration density than the mesic site(120,000 and 6000 ind ha-1, respectively), and it decreased afterwards. The number of established regeneration（> 2 m height） was lower for Na in the mesic site and for No in the humid site(0 and 57 ind ha-1, respectively). However, in comparison to No, Na showed a higher number of sprouted stumps and sprouts per stump, and a higher sprout height in the mesic site. No exhibited higher sprout mortality in the humid site. Transcriptomic responses to drought stress in the Patagonian southern beech Nothofagus alpina

Background Deciphering the genetic architecture of drought tolerance could allow the candidate genes identification responding to water stress. In the Andean Patagonian forest, the genus Nothofagus represents an ecologically relevant species to be included in different genomic studies. These studies are scarce in South American ecosystems however represent an important source of genomic data in order to interpret future climate-change environment scenarios of these emblematic forests. Here, we achieved the assemblage of the transcriptome of N. alpina while searching for key genes of activated or suppressed metabolic pathways in response to drought *** De novo transcriptome assembly resulted in 104,030 transcripts. Following confirmation of drought conditions, based on reduction of leaf water potential and stomatal conductance, a differential gene expression analysis resulted in 2720 significantly expressed genes(1601 up-regulated and 1119 down-regulated). Enrichment analysis(over-representation analysis and gene set enrichment analysis) resulted in more than one hundred stress-responsive term ontologies(i.e. biological processes) and pathways. Terms such as response to abscisic acid and pathways such as plant hormone signal transduction or starch and sucrose metabolism were over-represented. Protein–protein interaction assessment resulted in networks with significantly expressed top common hub gene clusters(e.g. planttype cell wall biogenesis among down-regulated or ABA-signalling among up-regulated). Geochemical Characterization of Soils in the Playa Lake Environment of Laguna de Encinillas, Chihuahua, Mexico

Laguna de Encinillas, located in Chihuahua, Mexico, is an ephemeral lake situated within a playa lake sedimentary depositional environment. This region plays a significant role in the aquifer supplying water to Chihuahua City. A surficial soil sampling campaign was conducted in 2017, 2018, and 2021 to assess the potential impact of surface soil composition on groundwater quality. The collected soil samples were analyzed using inductively coupled plasma mass spectrometry (ICP-MS) following microwave-assisted digestion. The analytical results were employed to generate spatial distribution maps of elemental concentrations using QGIS, applying interpolation methods such as inverse distance weighting (IDW) and Kriging. The analysis revealed elevated concentrations of beryllium (Be), calcium (Ca), cadmium (Cd), chromium (Cr), magnesium (Mg), and strontium (Sr) in the eastern part of the study area. In contrast, arsenic (As) and iron (Fe) were more prevalent in the western sector. Notably, high levels of barium (Ba), silver (Ag), cobalt (Co), potassium (K), and scandium (Sc) were identified in the northeastern region. At the same time, nickel (Ni), titanium (Ti), and vanadium (V) were concentrated in the northern portion. Manganese (Mn) was particularly prominent in the southern area. The geogenic source of these elements is likely linked to the volcanic rocks from the surrounding mountain ranges.
