In a recent study published in Nature,Garcia et *** a sophisticated approach to identify fitness-enhancing mutations for T cells that was inspired by cancer evolution.1 The identified CARD11-PIK3R3 gene fusion enhanced tumor rejection and persistence of engineered T cells in multiple tumor models and might have the potential to improve efficacy of adoptive T-cell therapies in cancer patients.

Novel analytical methods were developed for the determination of nitrapyrin and 6-chloropicolinic acid (6-CPA) residues in agricultural products. Nitrapyrin was extracted with hexane: toluene (1:1, v/v) and 6-CPA with 0.1 N sodium hydroxide (NaOH). For nitrapyrin, samples were cleaned with silica solid phase extraction (SPE) and for 6-CPA with C18 or carbon SPE. Analysis was performed by liquid chromatography-tandem mass spectroscopy (lC-MS/MS) with external calibration (0.010 to 0.0010 µg∙ml−1 for nitrapyrin and 0.0050 to 0.00025 µg∙ml−1 for 6-CPA). Recoveries for nitrapyrin and 6-CPA were 68% - 102% and 66% - 90%, respectively. Nitrapyrin was detected in celery and onion (−1) and 6-CPA in onion, mustard greens and lettuce (−1). lower level method validation (llMV) for nitrapyrin and 6-CPA was 0.020 µg∙g−1 and 0.050 µg∙g−1, respectively. The methods presented in this study are robust and were utilized for the registration of nitrapyrin on agricultural commodities in the United States.

Monitoring and predicting highly localized weather events over a very short-term period,typically ranging from minutes to a few hours,are very important for decision makers and public *** these events usually relies on radar observations through monitoring and *** advanced high-resolution imaging and sounding observations from weather satellites,nowcasting can be enhanced by combining radar,satellite,and other data,while quantitative applications of those data for nowcasting are advanced through using machine learning *** applications include monitoring the location,impact area,intensity,water vapor,atmospheric instability,precipitation,physical properties,and optical properties of the severe storm at different stages(pre-convection,initiation,development,and decaying),identification of storm types(wind,snow,hail,etc.),and predicting the occurrence and evolution of the *** observations can provide information on the environmental characteristics in the preconvection stage and are very useful for situational awareness and storm *** paper provides an overview of recent progress on quantitative applications of satellite data in nowcasting and its challenges,and future perspectives are also addressed and discussed.

The South China,Indochina,and Borneo margins surrounding the South China Sea contain long-lived arcs that became inactive at approximately 85 Ma,even though an embayment of oceanic crust(the‘Proto-South China Sea’)remained in the intervening *** oceanic crust eventually subducted in the Cenozoic below Borneo and the Cagayan arc,while the modern South China Sea opened in its *** investigate the enigmatic cessation of Mesozoic subduction below South China and Borneo,we studied a fragment of oceanic crust and overlying trench-fill sediments that accreted to NW Borneo during the final stages of Paleo-Pacific *** on radiolarian biostratigraphy of cherts overlying the pillow basalts and detrital zircon geochronology of the trench-fill,we constrained the minimum age of the oceanic crust during accretion to 40 *** shows that subduction cessation was not related to ridge *** analysis of pillow basalts revealed an enriched mid-ocean ridge basalt signature comparable to oceanic *** paleomagnetism,we show that this fragment of oceanic crust was not part of the Izanagi Plate but was part of a plate(the‘Pontus’Plate)separated from the Izanagi Plate by a subduction *** on the minimum 40 Ma age of the oceanic crust and its geochemistry,we suggest that Mesozoic subduction below South China and Borneo stopped when an oceanic plateau entered the trench,while the eastern plate margin with the Izanagi Plate remained *** show how our findings offer opportunities to restore plate configurations of the Panthalassa-Tethys junction region.

Bactrocera tryoni and Bactrocera neohumeralis are morphologically similar sibling pest fruit fly species that possess different biological attributes,geographic distributions,and host *** need to differentiate between the two species is critical for accurate pest status assessment,management,biosecurity,and maintenance of reference *** morphologically similar,adults may be separated based on subtle characters;however,some characters exhibit intraspecific variability,creating overlap between the two ***,there is currently no single molecular marker or rapid diagnostic assay that can reliably distinguish between *** and ***;therefore,ambiguous samples remain *** we report the first molecular marker that can consistently distinguish between *** and *** diagnostic region consists of two adjacent single nucleotide polymorphisms(SNPs)within the pangolin(pan)gene *** confirmed the genotypes of each species are consistent across their distributional range,then developed a tetra-primer amplification refractory mutation system(ARMS)PCR assay for rapid diagnosis of the *** assay utilizes four primers in multiplex,with two outer universal primers,and two internal primers:one designed to target two adjacent SNPs(AA)present in *** and the other targeting adjacent SNPs present in ***(GG).The assay accurately discriminates between the two species,but their SNP genotypes are shared with other nontarget tephritid fruit fly ***,this assay is most suited to adult diagnostics where species confirmation is necessary in determining ambiguous surveillance trap catches;maintaining pure colony lines;and in Sterile Insect Technique management responses.

The electrochemical CO_(2) reduction reaction(CO_(2)RR) to controllable chemicals is considered as a promising pathway to store intermittent renewable energy. Herein, a set of catalysts based on copper-nitrogendoped carbon xerogel(Cu-N-C) are successfully developed varying the copper amount and the nature of the copper precursor, for the efficient CO_(2)RR. The electrocatalytic performance of Cu-N-C materials is assessed by a rotating ring-disc electrode(RRDE), technique still rarely explored for CO_(2)RR. For comparison, products are also characterized by online gas chromatography in a H-cell. The as-synthesized Cu-NC catalysts are found to be active and highly CO selective at low overpotentials(from -0.6 to -0.8 V ***) in 0.1 M KHCO_(3), while H_(2) from the competitive water reduction appears at larger overpotentials(-0.9 V vs. RHE). The optimum copper acetate-derived catalyst containing Cu-N_(4) moieties exhibits a CO_(2)-to-CO turnover frequency of 997 h^(-1) at -0.9 V vs. RHE with a H_(2)/CO ratio of 1.8. These results demonstrate that RRDE configuration can be used as a feasible approach for identifying electrolysis products from CO_(2)RR.

An in-process technology approach is proposed to identify the source of acid mine drainage(AMD)generation and prevent its formation in a porphyry copper waste rock(WR).Adopting actions before stockpiling the WR enables the establishment of potential contaminants and predicts the more convenient method for AMD prevention.A WR sample was separated into size fractions,and the WR’s net acidgenerating potential was quantified using chemical and mineralogical *** diameter of physical locking of sulfides(DPlS)was determined,and the fractions below the DPlS were desulfurized using ***,the WR fractions and tailing from the flotation test were submitted to acid-base accounting and weathering tests to evaluate their acid-generating *** show that the WR’s main sulfide mineral is pyrite,and the DPlS was defined as 850μm.A sulfide recovery of 91%was achieved using a combination of HydroFloat^(®)and Denver cells for a size fraction lower than *** grinding was *** results show that size fractions greater than DPlS and the desulfurized WR are unlikely to produce *** outcomes show that in-processing technology can be a more proactive approach and an effective tool for avoiding AMD in a porphyry copper WR.

The gray mold fungus Botrytis cinerea is a necrotrophic pathogen that causes diseases in hundreds of plant species,including high-value *** polyxenous nature and pathogenic success are due to its ability to perceive host signals in its *** this study,we found that laticifer cells of Euphorbia lathyris are a source of susceptibility factors required by *** to cause ***,poor-in-latex(pil)mutants,which lack laticifer cells,show full resistance to this pathogen,whereas lot-of-latex mutants,which produce more laticifer cells,are *** S factors are triterpenoid saponins,which are widely distributed natural products of vast structural *** downregulation of laticifer-specific oxydosqualene cyclase genes,which encode the first committed step enzymes for triterpene and,therefore,saponin biosynthesis,conferred disease resistance to ***,the Medicago truncatula Iha-1 mutant,compromised in triterpenoid saponin biosynthesis,showed enhanced ***,the application of different purified triterpenoid saponins pharmacologically complemented the diseaseresistant phenotype ofpil and hla-1 mutants and enhanced disease susceptibility in different plant *** found that triterpenoid saponins function as plant cues that signal transcriptional reprogramming in ***,leading to a change in its growth habit and infection strategy,culminating in the abundant formation of infection cushions,the multicellular appressoria apparatus dedicated to plant penetration and biomass destruction in *** together,these results provide an explanation for how plant triterpenoid saponins function as disease susceptibility factors to promote *** pathogenicity.
