Skeletal stem and progenitor cells(SSPCs) perform bone maintenance and repair. With age, they produce fewer osteoblasts and more adipocytes leading to a loss of skeletal integrity. The molecular mechanisms that underlie this detrimental transformation are largely unknown. Single-cell RNA sequencing revealed that Notch signaling becomes elevated in SSPCs during aging. To examine the role of increased Notch activity, we deleted Nicastrin, an essential Notch pathway component, in SSPCs in vivo. Middle-aged conditional knockout mice displayed elevated SSPC osteo-lineage gene expression, increased trabecular bone mass, reduced bone marrow adiposity, and enhanced bone repair. Thus, Notch regulates SSPC cell fate decisions, and moderating Notch signaling ameliorates the skeletal aging phenotype, increasing bone mass even beyond that of young mice. Finally, we identified the transcription factor Ebf3 as a downstream mediator of Notch signaling in SSPCs that is dysregulated with aging, highlighting it as a promising therapeutic target to rejuvenate the aged skeleton.

Optimization of Wire-EDM Parameters for Ti-6Al-4v Using Taguchi-MRSN Optimization Approach

Ti-6Al-4v is a lightweight， extremely strong material with high resistance to corrosion and is widely used in aerospace industries， medical implants and many more. Contour cutting in such material with sharp corner radius， narrow kerf width， and maintaining high dimensional accuracy and surface integrity is a challenge for the modern manufacturing industry. Wire electrical discharge machining （WEDM） is a feasible process to meet the above challenges. However， proper control of process parameters is highly necessary for quality cutting of such material. In order to meet the above challenges， the present work focuses on the optimum selection of different process parameters such as pulse-on time (TON)， pulse-off time (TOFF)， spark gap voltage （Sv） and wire feed rate （WF） using Taguchi-based multi response signal to noise ratio （MRSN） technique. This is a very simple multi response optimization technique to simultaneously optimize vital responses like thickness of white layer formed on the machined surfaces， average crack density， surface roughness and dimensional accuracy， without the assistance of any software. Moreover， simple regression models are developed to determine the above responses for any combination of process parameters. Industry personnel will find this study useful in utilizing this expensive non-conventional machining method in a techno-economic manner.

B-L model with D_(4)×Z_(4)×Z_(2)symmetry for fermion mass hierarchies and mixings

We constructed a gauge B-L model with D_(4)×Z_(4)×Z_(2)symmetry to explain the quark and lepton mass hierarchies and their mixings with realistic CP phases via the type-I seesaw *** quark mases,three quark mixing angles,and the CP phase in the quark sector take the central values whereas Yukawa couplings in the quark sector are diluted in a range of difference of three orders of magnitude by the perturbation theory at the first *** the neutrino sector,a small neutrino mass is achieved by the type-I seesaw *** inverted and normal neutrino mass hierarchies are consistent with the experimental *** predicted sum of neutrino masses for normal and inverted hierarchies,the effective neutrino masses,and the Dirac CP phase are also consistent with recently reported limits.

A generalized kinetic model of atomic level populations in an optically dense plasma excited by laser pulses of arbitrary duration is formulated and *** model is based on a nonstationary expression for the probability of excitation of an atomic transition and takes into account the effects of laser pulse penetration into an optically dense medium.A universal formula for the excitation probability as a function of time and propagation length is derived and applied to the case of a Lorentzian spectral profile of an atomic transition excited by a laser pulse with a Gaussian *** features of nonstationary excitation probabilities are presented for different optical depths of the plasma,laser pulse durations,and carrier *** formulas derived here will be useful for the description of atomic populations excited by laser pulses under realistic conditions of dense plasmas.

Characterization of early maturing elite genotypes based on MTSI and MGIDI indexes:an illustration in upland cotton(Gossypium hirsutum L.)

Background Globally,the cultivation of cotton is constrained by its tendency for extended periods of *** maturity plays a potential role in rainfed-based multiple cropping system especially in the current era of climate *** the current study,a set of 20 diverse Gossypium hirsutum genotypes were evaluated in two crop seasons with three planting densities and assessed for 11 morphological traits related to early *** study aimed to identify genotype(s)that mature rapidly and accomplish well under diverse environmental conditions based on the two robust multivariate techniques called multi-trait stability index(MTSI)and multi-trait genotype-ideotype distance index(MGIDI).Results MTSI analysis revealed that out of the 20 genotypes,three genotypes,viz.,NNDC-30,A-2,and S-32 accomplished well in terms of early maturity traits in two ***,three genotypes were selected using MGIDI method for each planting densities with a selection intensity of 15%.The strengths and weaknesses of the genotypes selected based on MGIDI method highlighted that the breeders could focus on developing early-maturing genotypes with specific traits such as days to first flower and boll *** selected genotypes exhibited positive genetic gains for traits related to earliness and a successful harvest during the first and second ***,there were negative gains for traits related to flowering and boll *** The study identified three genotypes exhibiting early maturity and accomplished well under different planting *** multivariate methods(MTSI and MGIDI)serve as novel approaches for selecting desired genotypes in plant breeding programs,especially across various growing *** methods offer exclusive benefits and can easily construe and minimize multicollinearity issues.
