Genetic differentiation in functional traits among wild cherry (Prunus avium L.) half-sib lines

Understanding intra-specific variation in leaf functional traits is one of the key requirements for the evaluation of species adaptive capacity to ongoing climate change, as well as for designing long-term breeding and conservation strategies. Hence, data of 19 functional traits describing plant physiology, antioxidant properties, anatomy and morphology were determined on 1-year-old seedlings of wild cherry (Prunus avium L.) half-sib lines. The variability within and among half-sib lines, as well as the estimation of multi-trait association, were examined using analysis of variance (ANOVA) followed by Tukey's honestly significant difference test and multivariate analyses: principal component analysis (PCA), canonical discriminant analysis (CDA) and stepwise discriminant analysis (SDA). Pearson’s correlation coefficient was used to evaluate linear correlation between the study parameters. The results of the ANOVA showed the presence of statistically significant differences (P < 0.01) among half-sib lines for all study traits. The differences within half-sib lines, observed through the contribution of the examined sources of variation to the total variance (%), had higher impact on total variation in the majority of the examined traits. Pearson’s correlation analysis and PCA showed strong relationships between gas exchange in plants and leaf size and stomatal density, as well as between leaf biomass accumulation, intercellular CO_(2) concentration and parameters related to antioxidant capacity of plants. Likewise, the results of SDA indicate that transpiration and stomatal conductance contributed to the largest extent, to the discrimination of the wild cherry half-sib lines. In addition, PCA and CDA showed separation of the wild cherry half-sib lines along the first principal component and first canonical variable with regards to humidity of their original sites. The European glass lizard,Pseudopus apodus(Pallas,1775),is a large,legless lizard with wide distribution across south-eastern Europe and eastern and central *** date,morphological diversification among populations on a geographically small scale has not yet been reported in this ***,we investigated the morphological variations and corresponding differences in habitat utilization between two populations of *** inhabiting the same biogeographical zone within a relatively close geographic *** hypothesized that minor differences in habitat could induce a significant level of morphological differentiation,thus indicating morphological plasticity in this species on a small geographical *** sampled 164 individuals(92 from the Croatian mainland and 72 from the island of Cres).Results showed that *** indeed exhibited morphological differences between populations in the same biogeographical zone within a relatively close geographic area,with the Cres Island individuals being generally larger than the individuals from the *** ecological characteristics were similar in both populations(e.g.,soil temperature,distance to hiding place),whereas others were distinct(e.g.,air temperature and humidity).In addition,vegetation cover differed between the two sites,with more vegetation present on the mainland than on the ***,the Cres Island population showed clear sexual dimorphism,which was absent in the mainland population.

Impact of creatinine methodology on glomerular filtration rate estimation in diabetes

AIM To evaluate the influence of creatinine methodology on the performance of chronic kidney disease(CKD)-Epidemiology Collaboration Group-calculated estimated glomerular filtration rate(CKD-EPI-eGFR) for CKD diagnosis/staging in a large cohort of diabetic patients. METHODS Fasting blood samples were taken from diabetic patients attending our clinic for their regular annual examination, including laboratory measurement of serum creatinine and *** Our results indicated an overall excellent agreement in CKD staging(kappa = 0.918) between the Jaffé serum creatinine-and enzymatic serum creatinine-based CKDEPI-eGFR, with 9% of discordant cases. As compared to the enzymatic creatinine, the majority of discordances(8%) were positive, i.e., associated with the more advanced CKD stage re-classification, whereas only 1% of cases were negatively discordant if Jaffé creatinine was used for eGFR calculation. A minor proportion of the discordant cases(3.5%) were re-classified into clinically relevant CKD stage indicating mildly to moderately decreased kidney function(< 60 m L/min per 1.73 m^2). Significant acute and chronic hyperglycaemia, assessedas plasma glucose and Hb A1 c levels far above the recommended glycaemic goals, was associated with positively discordant cases. Due to a very low frequency, positive discordance is not likely to present a great burden for the health-care providers, while intensified medical care may actually be beneficial for the small number of discordant patients. On the other hand, a very low proportion of negatively discordant cases(1%) at the 60 m L/min per 1.73 m^2 eGFR level indicate a negligible possibility to miss the CKD diagnosis, which could be the most prominent clinical problem affecting patient care, considering high risk of CKD for adverse patient outcomes. CONCLUSION This study indicate that compensated Jaffé creatinine procedure, in spite of the glucose-dependent bias, is not inferior to enzymatic creatinine in CKD diagnosis/

AIM: To investigate activity, toxicity, and prognostic factors for survival of erlotinib and fixed dose-rate gemcitabine (FDR-Gem) in advanced pancreatic cancer.METHODS: We designed a single-arm prospective, multicentre, open-label phase II study to evaluate the combination of erlotinib (100 mg/d, orally) and weekly FDR-Gem (1000 mg/m2, infused at 10 mg/m2 per minute) in a population of previously untreated patients with locally advanced, inoperable, or metastatic pancreatic cancer. Primary endpoint was the rate of progression-free survival at 6 mo (PFS-6); secondary endpoints were overall response rate (ORR), response duration, tolerability, overall survival (OS), and clinical benefit. Treatment was not considered to be of further interest if the PFS-6 was 40% would be of considerable interest (p1 = 40%); with a 5% rejection error (α = 5%) and a power of 80%, 35 fully evaluable patients with metastatic disease were required to be enrolled in order to complete the study. Analysis of prognostic factors for survival was also carried out.RESULTS: From May 2007 to September 2009, 46 patients were enrolled (male/female: 25/21; median age: 64 years; median baseline carbohydrate antigen 19-9 (CA 19-9): 897 U/mL; locally advanced/metastatic disease: 5/41). PFS-6 and median PFS were 30.4% and 14 wk (95%CI: 10-19), respectively; 1-year and median OS were 20.2% and 26 wk (95%CI: 8-43). Five patients achieved an objective response (ORR: 10.9%, 95%CI: 1.9-19.9); disease control rate was 56.5% (95%CI: 42.2-70.8); clinical benefit rate was 43.5% (95%CI: 29.1-57.8). CA 19-9 serum levels were decreased by > 25% as compared to baseline in 14/23 evaluable patients (63.6%). Treatment was well-tolerated, with skin rash being the most powerful predictor of both longer PFS (P < 0.0001) and OS (P = 0.01) at multivariate analysis (median OS for patients with or without rash: 42 wk vs 15 wk, respectively, Log-rank P = 0.03). 肠道病毒属(Enterovirus)是一种小型无包膜的正义单链RNA病毒属,包括脊髓灰质炎病毒、柯萨奇病毒、新型肠道病毒、埃可病毒和人鼻病毒等临床上重要的人类病原体。肠道病毒所引起的感染在临床表现中的程度各不相同,从无明显症状到轻微的疾病如手足口病和普通感冒,再到严重甚至致命的神经、心血管和肺部疾病。目前对能广谱抗肠道病毒的药物和疫苗等治疗和预防方法都存在迫切需求。最近发现,肠道病毒2A蛋白水解酶(2A)和宿主肌动蛋白组氨酸甲基转移酶SETD3之间的相互作用对多种肠道病毒的复制至关重要,于是此相互作用便成为抗病毒药物和治疗的一个潜在新靶点。虽然之前已经有突变试验尝试研究对结合SETD3所需的2A氨基酸残基,但SETD3-2A相互作用的结构基础尚未清楚,因此限制了我们对肠道病毒2A如何利用宿主蛋白SETD3达到病毒自身繁衍目的的理解。本研究解析了SETD3与柯萨奇病毒B3 2A(CV-B3 2A)和CV-A10 2A所形成的两种复合物的晶体结构,并开展了系统的结构和生物信息学分析,以及突变和生化实验。研究发现,这两个复合物中2A结合在SETD3两叶结构之间的V字形凹陷内,由于2A和SETD3都没有经历可观察到的构象变化,两蛋白之间的相互作用基于结构互补的多种弱相互作用。通过对相互作用界面的详细分析,发现了三个主要的分子间相互作用群组,其中2A锌结合位点周围的结构元件与SETD3的SET结构域之间所形成的两个相互作用群组在两个复合物中都是保守的。经相关CV-B3 2A和SETD3突变体的生化实验表明,CV-B3 2A的V62、K68-N69-K70(特别是K70)和Y72,以及SETD3的V266、Y288和E296是维持SETD3-CV-B32A之间相互作用的关键氨基酸残基。序列比对表示大多数肠道病毒的2A共享参与SETD3结合的关键氨基酸残基,表明它们与SETD3的结合方式具有很强的相似性。最后,本研究在结构数据和相关文献基础上探讨了SETD3-2A相互作用在病毒复制中的作用。本研究阐明了肠病毒2A蛋白酶与SETD3相互作用的分子基础,为研发靶向此相互作用的广谱抗病毒药物和治疗奠定了基础。
