Secretoneurin （SN） is a neuropeptide derived from specific proteol.tic processing of the precursor secretogranin II （Sgl.）. In zebrafish and other tel.osts, there are two paral.gs named sgl.a and sgl.b. Our resul.s showed that neurons expressing sgl.b were al.gned with central.arteries in the hindbrain, demonstrating a cl.se neurovascul.r association. Both sgl.b-/- and sgl.o-/-/sgl.b-/- mutant embryos were defective in hindbrain central.artery devel.pment due to impairment of migration and prol.feration of central.artery cel.s. Further study reveal.d that sgl.b is non-ceU autonomous and required for central.artery devel.pment. Hindbrain arterial.and venous network identities were not affected in sgl.b-/- mutant embryos, and the mRNA l.vel. of Notch and VEGF pathway-rel.ted genes were not al.ered. However, the activation of MAPK and PI3K/AKT pathways was inhibited in sgl.b-/- mutant embryos. Reactivation of MAPK or PI3K/AKT in endothel.al.cel.s coul. partial.y rescue the central.artery devel.pmental.defects in the sgl.b mutants. This studV provides the first in vivo evidence that sgl.b ptavs a critical.rote in neurovascutar model.ng of the hindbrain. Targeting the Sgl. system may, therefore, represent a new avenue for the treatment of vascul.r defects in the central.nervous system.
