Dioxins in the Food Chain: Contamination Fingerprint Analysis in Breeding Hens, Hatching Eggs and Broilers

While routine monitoring poultry meat was obtained from breeding hens, dioxins contaminations were detected in Portugal. Levels of 430.9 pg PCDD/F-WHO-TEQ/g1 were found, which are higher than the official limits legally allowed for this matrix (1.75 pg PCDD/F-WHO-TEQ/g). To identify the magnitude of the contaminations, 60 samples were collected from poultry farms and different matrices, namely: feed, water, wood shavings from the litters, muscle of the breeding hens, hatching eggs collected in the positive farm and muscle collected from broilers farms supplied by the positive breeding farm. The comparison of the dioxins congeners profiles showed that there was a coincidence of peaks of higher relative concentrations in the wood shavings, with the peaks of the highest relative concentration in the hatching eggs, especially the relative concentrations of the congeners 1,2,3,4,6,7,8-HpCDD and OCDD, which may be explained by the role of VLDLy in the delivery of triglycerides to the oocyte, where they will be used as the energy source for the developing embryo. The comparison of the dioxins congeners profiles of the breeding hens muscle with the poultry muscle, showed a coincidence of peaks of higher relative concentrations in the congeners 1,2,3,7,8-PeCDD, 1,2,3,6,7,8-HxCDD, 1,2,3,4,6,7,8-HpCDD and OCDD which may indicate a dechlorination pathway “in vivo”. Results allowed concluding that those wood shavings, improperly used as poultry litters, were certainly the source of contamination of the food chain.

Virtual reality interface devices in the reorganization of neural networks in the brain of patients with neurological diseases

Two key characteristics of all virtual reality applications are interaction and immersion. Systemic interaction is achieved through a variety of multisensory channels （hearing, sight, touch, and smell）, permitting the user to interact with the virtual world in real time. Immersion is the degree to which a person can feel wrapped in the virtual world through a defined interface. Virtual real- ity interface devices such as the Nintendo~ Wii and its peripheral nunchuks-balance board, head mounted displays and joystick allow interaction and immersion in unreal environments created from computer software. Virtual environments are highly interactive, generating great activation of visual, vestibular and proprioceptive systems during the execution of a video game. In addi- tion, they are entertaining and safe for the user. Recently, incorporating therapeutic purposes in virtual reality interface devices has allowed them to be used for the rehabilitation of neurological patients, e.g., balance training in older adults and dynamic stability in healthy participants. The improvements observed in neurological diseases （chronic stroke and cerebral palsy） have been shown by changes in the reorganization of neural networks in patients＇ brain, along with better hand function and other skills, contributing to their quality of life. The data generated by such studies could substantially contribute to physical rehabilitation strategies.

Differences in standing balance between patients with diplegic and hemiplegic cerebral palsy

Maintaining standing postural balance is important for walking and handling abilities in patients with cerebral palsy. This study included 23 patients with cerebral palsy （seven with spastic diplegia and 16 with spastic hemiplegia）, aged from 7 to 16 years of age. Standing posture balance measurements were performed using an AMTI model OR6-7 force platform with the eyes open and closed. Patients with diplegic cerebral palsy exhibited greater center of pressure displacement areas with the eyes open and greater center of pressure sway in the medial-lateral direction with the eyes open and closed compared with hemiplegic patients, Thus, diplegic patients exhibited weaker postural balance control ability and less standing stability compared with hemiplegic cerebral palsy patients.

Tree sapling vitality and recovery following the unprecedented 2018 drought in central Europe

H pylori (CagA) and Epstein-Barr virus infection in gastric carcinomas:Correlation with p53 mutation and c-Myc,Bcl-2 and Bax expression

AIM: To investigate the interrelationship between H pylori and Epstein-Barr virus (EBV) infection in the gastric carcinogenesis having in focus the p53 mutation and the c-Myc, Bcl-2 and Bax expression. METHODS: seventy-one gastric carcinoma tissues were assessed by polymerase chain reaction (PCR) for H pylori and in situ hybridization for EBV. c-Myc, Bcl-2 and Bax expression were detected by immunohistochemistry and single-stranded conformational polymorphism (SSCP) for p53 mutation. RESULTS: The positivity rates for H pylori and EBV were 94.4% and 8.45%, respectively. The majority of the cases displayed only the H pylori presence. All EBV positive cases were also H pylori positive. None infectious agent was observed in 5.55% of the cases. The intestinal type tumor was more frequent in the co-infected and non-infected groups. The female predominated in the non-infected group showing statistical significance (70.4% vs 29.6%, P=0.039). The Bcl-2 was only detected in the group exclusively infected by H pylori. However, c-Myc and Bax were detected in the three groups but with a low frequency in the co-infected group. Mutation of p53 was present in all groups, with the highest frequencies in the H pylori positive groups. CONCLUSION: The frequency of H pylori infection in gastric carcinomas was high. The presented data indicated that gastric carcinogenesis has different pathways depending of the presence of the two investigated infectious agents, suggesting a possible involvement of H pylori with apoptotic process. The low expression of c-Myc and Bax in the EBV-positive groups suggests that EBV may inhibit the expression of these proteins. Nevertheless, p53 mutation shows to be a relevant alteration, independent of both infectious agents.

Green Liver Systems for Water Purification:Using the Phytoremediation Potential of Aquatic Macrophytes for the Removal of Different Cyanobacterial Toxins from Water

The protection and reasonable use of freshwater is one of the main goals for our future, as water is most important for all organisms on earth including humans. Due to pollution, not only with xenobiotics, but also with nutrients, the status of our water bodies has changed drastically. Excess nutrient load induces eutrophication processes and, as a result, massive cyanobacterial blooms during the summer times. As cyanobacteria are known to produce several toxic secondary metabolites, the so-called cyanotoxins, exhibiting hepato-, neuro- and cell-toxicity, a potential risk is given, when using this water. There is an urgent need to have a water purification system, which is able to cope with these natural toxins. Using aquatic plants as a Green Liver, the Green Liver System?, was developed, able to remove these natural pollutants. To test the ability of the Green Liver System?, several cyanobacterial toxins including artificial and natural mixtures were tested in a small-scale laboratory system. The results showed that within 7 - 14 days a combination of different aquatic macrophytes was able to remove a given toxin amount (10 μg·L-1) by 100%. The phytoremediation technology behind the Green Liver Systems? uses the simple ability of submerged aquatic plants to uptake, detoxify and store the toxins, without formation and release of further metabolites to the surrounding water.

目的探讨氨基酮戊酸光动力疗法（ALA-PDT）对成纤维细胞生长因子10（FGF-10）诱导的HaCaT细胞过度角化及增殖的作用及其机制。方法将培养的HaCaT细胞分为4组：空白对照组（无任何处理），FGF-10组（加入FGF-10孵育24 h），ALA-PDT组（ALA孵育24 h后红光照射），FGF-10 ＋ ALA-PDT组（FGF-10孵育24 h后，再加ALA孵育24 h，红光照射）。用CCK-8法检测各组细胞增殖活性，Western印迹法检测K1、K6、K16及角质形成细胞生长因子受体（KGFR）的含量，ELISA法检测各组细胞上清液中白细胞介素1α（IL-1α）蛋白表达，免疫荧光检测各组细胞KGFR和K6蛋白表达水平。统计学分析采用析因设计的方差分析和单因素方差分析。结果析因分析显示，ALA-PDT和FGF-10对HaCaT细胞增殖无交互作用（F交互 = 1.369，P = 0.276），FGF-10对HaCaT细胞增殖有促进作用（FFGF-10 = 20.853，P 〈 0.05），而ALA-PDT有抑制作用（FALA-PDT = 24.822，P 〈 0.05）。与空白对照组细胞增殖活性（A值为0.924 ± 0.024）比较，FGF-10组（1.233 ± 0.099）显著升高（P 〈 0.05），而ALA-PDT组（0.718 ± 0.107）显著降低（P 〈 0.05），FGF-10 ＋ ALA-PDT组（0.901 ± 0.014）较FGF-10组显著降低（P 〈 0.05）。Western印迹法显示，FGF-10可促进K1、K6、K16蛋白和KGFR的表达（均P 〈 0.05），而ALA-PDT抑制这些蛋白的表达（均P 〈 0.05），FGF-10 ＋ ALA-PDT组K1、K6、K16和KGFR的相对表达量与FGF-10组比较均显著降低（均P 〈 0.05）。ELISA法显示，FGF-10会增加HaCaT细胞IL-1α蛋白分泌（P 〈 0.05），但ALA-PDT对其没有影响（P = 0.467）。此外，免疫荧光检测显示，FGF-10增加HaCaT细胞K6和KGFR免疫荧光强度（P 〈 0.05），而ALA-PDT降低K6和KGFR免疫荧光强度（P 〈 0.05），FGF-10 ＋ ALA-PDT组K6和KGFR免疫荧光强度显著低于FGF-10组（P 〈 0.05）。结论 ALA-PDT能抑制FGF-10诱导的HaCaT细胞过度角化及增殖，其机制与下调K1、K6、K16、KGFR及IL-1α有关。

目的研究氨基酮戊酸光动力疗法（ALA-PDT）对中波紫外线（UVB）应激性衰老成纤维细胞（UVB-SIPS-FB）的氧化损伤和凋亡作用.方法正常成纤维细胞及UVB-SIPS-FB均分为2h和6h孵育组,每组内再分为7组：对照组、ALA组、100 J/cm2照光组、3组不同红光照射剂量ALA-PDT处理组（25 J/cm2组、50 J/cm2组、100 J/cm2组）及抗氧化剂NAC＋ 50 J/cm2红光照射ALA-PDT处理组.采用635 nm红光（能量密度50 mW/cm2）照射处理后,使用荧光显微镜、流式细胞仪检测各组细胞中活性氧及线粒体膜电位水平;Hoechst染色、流式细胞仪检测细胞凋亡率.数据用SPSS 13.0软件分析,多组间均数行单因素方差分析,结合q检验.结果孵育2h,25、50、100 J/cm2 ALA-PDT组UVB-SIPS-FB凋亡率（分别为7.34％±0.87％、8.39％±1.16％、17.03％±1.29％）较对照组（3.81％±0.59％）上升（F=102.70,均P＜0.05）,NAC＋ 50 J/cm2 ALA-PDT组凋亡率（5.35％±0.58％）下降（P＜0.05）;孵育6h,25、50、100 J/cm2 ALA-PDT组UVB-SIPS-FB凋亡率（分别为13.85％±1.71％、23.40％±2.14％、41.02％±2.73％）较对照组（5.09％±1.64％）显著上升（F=106.00,均P＜0.05）,NAC＋50 J/cm2 ALA-PDT组凋亡率（9.97％±3.23％）显著下降（P＜0.05）.单纯ALA组或照光组UVB-SIPS-FB凋亡率与对照组差异均无统计学意义.ALA-PDT处理组细胞活性氧荧光强度显著上升,线粒体去极化增强,这些现象与ALA孵育时间及红光照射剂量均呈量效依赖性关系.使用抗氧化剂NAC预处理可以减少ALA-PDT诱导的活性氧增多及线粒体去极化增强,与凋亡率变化一致.结论 ALA-PDT可诱导UVB-SIPS-FB出现明显的细胞凋亡,其作用机制可能与ALA-PDT对细胞的氧化损伤有关.
