Screening of Rice Accessions for Tolerance to Drought and Salt Stress Using Morphological and Physiological Parameters

Drought and salinity are the most widespread soil problems, posing a big threat to food security in rice growing regions. The present study evaluated the performance of eleven rice genotypes using morphological and physiological parameters, under induced drought and salinity conditions. The seedlings were raised in 5 kg of homogenous soil in plastic bags in the greenhouse. For the drought experiment, each bag was watered with 200 ml of water twice daily until plants reached the five-leaf stage when watering was suspended for 2 weeks for the drought stressed plants but not suspended for the control plants. The experiment was a 2 × 11 factorial and the set up was arranged using the completely randomized design with three replications. Data were taken on Plant height, Number of tillers, leaf length, Number of green leaves, Number of dead leaves, Leaf rolling score (LRS) and Rate of water loss. The salinity experiment was set up in a similar manner except that the plants were irrigated twice a day for 2 weeks with 200 ml of treatment solution containing either 0 mM NaCl or 75 mM and data were collected on plant height, number of tillers, shoot fresh weight, shoot dry weight, Na+ and K+ concentrations, relative water content and chlorophyll content. Data from both experiments were subjected to Analysis of variance test using the GenStat software 10th edition and the means separated using least significant difference test. Cucumber mosaic virus is one of the most constraints to the production of tomato and other vegetable crops worldwide. Here, we generated an RNAi construct containing inverted repeat of 1138 bp fragment of a partial replicase gene of CMV-O and used it to produce transgenic tomato plants expressing CMV-specific dsRNA of the replicase gene. Inoculation of transgenic plants with CMV strain O discriminated three categories of plants: plants that showed complete resistance, which were free of symptoms;highly resistant plants, which had mild symptoms, but later recovered because new leaves that emerged were free of symptoms;and susceptible plants, which showed severe symptoms similar to wild-type plants. The completely resistant lines were selected and challenged with a closely related strain, CMV-Y. Interestingly, the transgenic plant lines either remained immune or showed high levels of resistance to the strain. No virus could be detected in uninoculated new leaves of the resistant lines after RT-PCR and Dot immunobinding assay (DIBA) analyses. We could show that the resistance is correlated with post-transcriptional gene silencing because of the production of transgenic specific siRNA.

Dear Editor Strigolactones （SLs） are novel phytohormones that shape plant architecture by inhibiting shoot branching and regulating root growth, besides their established functions in stimulating seed germination of root-parasitic weeds, such as Striga and Phelipanche species, and inducing hyphal branching in arbuscular mycorrhizal （AM） fungi （AI-Babili and Bouwmeester, 2015）. Canonical SLs are divided into strigoland orobanchollike subfamilies with a typical structure consisting of a tricyclic （ABC-ring） and a monocyclic lactone （D-ring）, which are connected by an enol ether bridge （Figure 1A and Supplemental Figure 1）. SLs are synthesized from carotenoids via carlactone, which lacks the B/C-ring （Alder et al., 2012）. In Arabidopsis, carlactone （Figure 1A） is converted by MAX1, a member of the CYP711 family, into carlactonoic acid, after methylation, which, can bind to SL receptor （Abe et al., 2014）.
