NSD3S stabilizes MYC through hindering its interaction with FBXW7

The MYC transcription factor plays a key role in cell growth control. Enhanced MYC protein stability has been found to promote tumorigenesis. Thus, understanding how MYC stability is controlled may have significant implications for revealing MYC-driven growth regulatory mechanisms in physiological and pathological processes. Our previous work identified the histone lysine methyltransferase nuclear receptor binding SET domain protein 3 (NSD3) as a MYC modulator. NSD3S, a noncatalytic isoform of NSD3 with oncogenic activity, appears to bind, stabilize, and activate the transcriptional activity of MYC. However, the mechanism by which NSD3S stabilizes MYC remains to be elucidated. To uncover the nature of the interaction and the underlying mechanism of MYC regulation by NSD3S, we characterized the binding interface between both proteins by narrowing the interface to a 15-amino acid region in NSD3S that is partially required for MYC regulation. Mechanistically, NSD3S binds to MYC and reduces the association of F-box and WD repeat domain containing 7 (FBXW7) with MYC, which results in suppression of FBXW7-mediated proteasomal degradation of MYC and an increase in MYC protein half-life. These results support a critical role for NSD3S in the regulation of MYC function and provide a novel mechanism for NSD3S oncogenic function through inhibition of FBXW7-mediated degradation of MYC.

Clinical features of alcoholic hepatitis in latinos and caucasians: A single center experience

AIM To study differences of presentation, management, and prognosis of alcoholic hepatitis in Latinos compared to Caucasians. METHODS We retrospectively screened 876 charts of Caucasian and Latino patients who were evaluated at University of California Davis Medical Center between 1/1/2002-12/31/2014 with the diagnosis of alcoholic liver disease. We identified and collected data on 137 Caucasians and 64 Latinos who met criteria for alcoholic hepatitis, including chronic history of heavy alcohol use, at least one episode of jaundice with bilirubin ≥ 3.0 orcoagulopathy, new onset of liver decompensation or acute liver decompensation in known cirrhosis within 12 wk of last drink. RESULTS The mean age at presentation of alcoholic hepatitis was not significantly different between Latinos and Caucasians. There was significant lower rate of overall substance abuse in Caucasians compared to Latinos and Latinos had a higher rate of methamphetamine abuse(12.5% vs 0.7%) compared to Caucasians. Latinos had a higher mean number of hospitalizations(5.3 ± 5.6 vs 2.7 ± 2.7, P = 0.001) and mean Emergency Department visits(9.5 ± 10.8 vs 4.5 ± 4.1, P = 0.017) for alcohol related issues and complications compared to Caucasians. There was significantly higher rate of complications of portal hypertension including gastrointestinal bleeding(79.7% vs 45.3%, P < 0.001), spontaneous bacterial peritonitis(26.6% vs 9.5%, P = 0.003), and encephalopathy(81.2% vs 55.5%, P = 0.001) in Latinos compared to *** Latinos have significant higher rates of utilization of acute care services for manifestations alcoholic hepatitis and complications suggesting poor access to outpatient care.

Molecular survey of tick-borne pathogens in Ixodid ticks collected from hunted wild animals in Tuscany, Italy

Objective: To determine the prevalence of zoonotic tick-borne bacteria in feeding ticks removed from hunted wild animals. Methods: PCR was executed on DNA extracted from 77 tick pools to detect Anaplasma phagocytophilum, Bartonella spp., Borrelia burgdorferi sensu lato, Coxiella burnetii and Rickettsia spp. Results: A total of 432 ticks were collected: 30(6.94%) Haemaphysalis punctata, 72(16.7%) Dermacentor marginatus and 330(76.38%) Ixodes ricinus. For each animal one or two pools of 3 ticks of the same species was constituted. Seventy-seven tick pools were examined by PCR: 58(75.32%) resulted infected and among them 14(18.18%) showed co-infections. In particular, 29(37.66%) pools were positive for Bartonella spp., 23(29.87%) for Anaplasma phagocytophilum, 16(20.78%) for Rickettsia spp., and 5(6.49%) for Borrelia burgdorferi s.l. All samples were negative for Coxiella burnetii. Conclusions: The results demonstrate the presence of several zoonotic tick-borne pathogens in the studied area, and underline the risk of exposure to infections for hunters not only during the outdoor activity, but also when they manipulate hunted animals infested by infected ticks.

Feeding broiler chickens with arginine above recommended levels:effects on growth performance,metabolism,and intestinal microbiota

Occurrence of Escherichia coli virulence genes in feces of wild birds from Central Italy

Objective: To investigate the potential role of wild birds as fecal spreaders of enteropathogenic,enterohemorrhagic and Shiga-toxins producing Escherichia coli(E. coli),enteropathogenic E. coli,enterohemorrhagic E. coli and Shiga toxin-producing E. coli strains. Methods: Fecal samples collected from 121 wild birds of different orders and species were submitted to molecular analyses. In particular,eaeA encoding intimin,hlyA encoding for hemolysin,stx1 and stx2 genes encoding Shiga-toxins 1 and 2,respectively,were investigated. Results: Overall,21(17.35%) fecal samples resulted positive for at least one of the investigated genes. In detail,12(9.91%) samples were positive for eaeA,10(8.26%) for stx1,4(3.31%) for hylA and 1(0.83%) for stx2. An owl(Athene noctua) positive for the four investigated genes suggesting that it harbored a STEC strain. However,virulence genes characterizing EPEC,and EHEC strains were mainly found among seagulls,waterfowl and feral pigeons. Conclusions: Seagulls,waterfowl and feral pigeons,which frequently reach and contaminate rural,urban and peri-urban areas with their droppings,may be important sources of E. coli infection for other animals and humans.
