Overexpression of the cholesterol-binding protein MLN64 induces liver damage in the mouse

AIM： To examine the in vivo phenotype associated with hepatic metastatic lymph node 64 （MLN64） overexpression.MetHODS： Recombinant-adenovirus-mediated MLN64 gene transfer was used to overexpress MLN64 in the livers of C57BL/6 mice. We measured the effects of MLN64 overexpression on hepatic cholesterol content, bile flow, biliary lipid secretion and apoptosis markers. For in vitro studies cultured CHO cells with transient MLN64 overexpression were utilized and apoptosis by TUNEL assay was measured.RESULTS： Livers from ***64-infected mice exhibited early onset of liver damage and apoptosis. This response correlated with increases in liver cholesterol content and bilian/bile acid concentration, and impaired bile flow. We investigated whether liver MLN64 expression could be modulated in a murine model of hepatic injury. We found increased hepatic MLN64 mRNA and protein levels in mice with chenodeoxycholic acid-induced liver damage. In addition, cultured CliO cells with transient MLN64 overexpression showed increased apoptosis.CONCLUSION： In summary, hepatic MLN64 overexpression induced damage and apoptosis in murine livers and altered cholesterol metabolism. Further studies are required to elucidate the relevance of these findings under physiologic and disease conditions.

We read with great interest the study by Way et al.1 published in this journal,which described and compared the acute changes in central arterial stiffness(AS),wave reflections,and central hemodynamics(CH)in response to a bout of high-intensity interval exercise(HIIE)and moderate-intensity continuous exercise(MICE)in adults with *** authors reported a significant reduction in augmentation index(AIx)corrected by a heart rate(HR)of 75 beats/min(AIx@75)and central systolic blood pressure after HIIE,without significant changes in the pulse wave velocity(PWV).In view of this important research topic and our recent contributions to it,we would like to raise some points of discussion that might help to better elucidate the findings observed in the current study,and also suggest pertinent directions to further studies.

To the Editor： Xie et al, analyzed warfarin doses in pulmonary embolism （PE） and its adverse effects in patients with obstructive sleep apnea hypopnea syndrome （OSAHS）. The key findings were that OSAHS patients required a significantly higher dose of warfarin than their non-OSAHS counterparts （4.73 mg vs. 3.61 mg, P 〈 0.001） and that PE recurrence was higher in OSAHS than in non-OSAHS groups after withdrawal ofwarfarin （21.43% vs. 6.78%, P = 0.047）.

Comprehensive data from in situ and laboratory tests in residual soil have been presented by Zhang et al.(2023).A number of issues addressed in the paper have been the interest of the discussers,namely the characterisation and behaviour of residual soils,limitation of piezocone testing due to the capacity of the entire system,measurement of shear wave velocity,rate effect of piezocone(CPTU)testing and piezocone testing with dual pore pressure *** and complementation of these issues are required with regard to both the execution and interpretation of the tests.

Members of the ERF Family of Transcription Factors play an important role in plant development and gene expression that regulates responses to biotic and abiotic *** work identified 36 ERF family genes in Coffea arabica within the AP2/ERF full domain,using the EST-based genomic resource of the Brazilian Coffee Genome *** ERF family genes were classified into nine of the ten existing groups through phylogenetic analysis of the deduced amino acid sequences and comparison with the sequences of the ERF family genes in *** addition to the AP2 domain,other conserved domains were identified,typical of members of each *** in silico analysis and expression profiling showed high levels of expression for libraries derived from tissues of fruits,leaves and flowers as well as for libraries subjected to water *** results suggest the participation of the ERF family genes of *** in distinct biological functions,such as control of development,maturation,and responses to water *** results of this work imply in the selection of promising genes for further functional characterizations that will provide a better understanding of the complex regulatory networks related to plant development and responses to stress,opening up opportunities for coffee breeding programs.

Applying Rhizobacteria on Maize Cultivation in Northern Benin:Effect on Growth and Yield

The aim of the study was to investigate the effects of five plant growth-promoting rhizobacteria (PGPR) (Bacillus panthothenicus;Pseudomonas Cichorii;Pseudomonas Putida;Pseudomonas syringae and Serratia marcescens) on the growth and yield of maize on a ferruginous soil under field condition. Maize seeds were inoculated with 10 ml of bacterial suspension. Study was conducted in a completely randomized design with fifteen treatments and three replicates. A half-dose of recommended (13, 17, 17 kg?ha?1) NPK was applied 15 days after emergence. The results show that the Serratia marcescens + 50% NPK treatment yielded the best results for height, fresh underground biomass, dry aboveground biomass, dry underground biomass, and grain yield with respective increases of 41.09%, 217.5%, 213.34%, 93.82%, and 39.05% compared to the control. Maximum stem diameter (increases of 49.65%) was recorded in the plants treated with 100% NPK (full dose NPK) while the highest leaf area (466.36 ± 9.57 cm2), obtained on plant treated with Pseudomonas putida + 50% NPK was 32.08% greater than in the non-inoculated control. Our results suggest the use of these rhizobacteria as biological fertilizers for enhancing the growth and maize seed yield in ferruginous soil in the North of Benin.

AIM：To investigate the cytotoxic effects of spray-dried extracts of Phyllanthus niruri in combination with cis- platin on two cancer cell lines. MetHODS： Colorectal carcinoma （HT29） and human hepatocellular carcinoma （HepG2） cells were treated with spray-dried extracts of Phyllanthus niruri （SDEPN） either alone or in combination with cisplatin at differ- ent concentrations （0.5 mg/mL and 1 mg/mL） for 4 h and 24 h. To verify and quantify cancer cells treated with these products as well as identify the cell cycle stage and cell viability, we stained the cells with prop- idium iodide and assessed them by flow cytometry. The percentage of cells in different cell cycle phases was quantified and data were expressed as histo- grams. Significant differences between groups were determined using analysis of variance and Bonferroni＇s test, as indicated. A value of P 〈 0.05 was considered to be statistically significant. RESULTS： SDEPN had significantly different cyto- toxic effects on HT29 （2.81 4- 0.11 vs 3.51 4- 1.13, P 〉 0.05） and HepG2 （5.07± 0.3 vs 15.9 ± 1.04, P 〈 0.001） cells when compared to control cells for 4 h. SDEPN also had significantly different cytotoxic effects on HT29 （1.91 ± 0.57 vs 4.53± 1.22, P 〉 0.05） and HepG2 （14.56 ± 1.6 vs 35.67 ± 3.94, P 〈 0.001） cells when compared to control cells for 24 h. Both cell lines were killed by cisplatin in a dose-dependent manner compared to control cells （HepG2 cells for 4 h： 10.78 ± 1.58 vs 53.89 ± 1.53, P 〈 0.001; 24 h： 8.9 ± 1.43 vs 62.78 ± 1.87, P 〈 0.001 and HT29 cells for 4 h： 9.52 ±0.913 vs 49.86 ± 2.89, P 〈 0.001; 24 h： 11.78 ± 1.05 vs 53.34 ± 2.65, P 〈 0.001）. In HT29 cells, pretreat- ment with SDEPN and subsequent treatment with cis-platin resulted in a greater number of cells being killed （12.78 ± 1.01 vs 93.76 ± 1.6, P 〈 0.001）.
