The Asia-Pacific region is a home to 60% of the population in the world and to approximately one quarter of people with HIV/AIDS. Close to a million of people has been infected and a half million people died of AIDS annually in Asia,becoming the second largest epicenter of global AIDS epidemic. Molecular epidemiology has been useful tool to track a course of HIV spread. In-depth knowledge from the studies on molecular epidemiology elucidates the dynamics of HIV spread and the interrelationship of epidemics in the different regions in Asia.

A quantum cascade laser infrared spectrometer for CO_2 stable isotope analysis：Field implementation at a hydrocarbon contaminated site under bio-remediation

Real-time methods to monitor stable isotope ratios of CO_2 are needed to identify biogeochemical origins of CO_2 emissions from the soil–air interface. An isotope ratio infra-red spectrometer（IRIS） has been developed to measure CO_2 mixing ratio with δ~13C isotopic signature, in addition to mixing ratios of other greenhouse gases（CH_4, N2_O）. The original aspects of the instrument as well as its precision and accuracy for the determination of the isotopic signature δ~13C of CO_2 are discussed. A first application to biodegradation of hydrocarbons is presented, tested on a hydrocarbon contaminated site under aerobic bio-treatment. CO_2 flux measurements using closed chamber method is combined with the determination of the isotopic signature δ~13C of the CO_2 emission to propose a non-intrusive method to monitor in situ biodegradation of hydrocarbons. In the contaminated area, high CO_2 emissions have been measured with an isotopic signature δ~13C suggesting that CO_2 comes from petroleum hydrocarbon *** first field implementation shows that rapid and accurate measurement of isotopic signature of CO_2 emissions is particularly useful in assessing the contribution of contaminant degradation to the measured CO_2 efflux and is promising as a monitoring tool for aerobic bio-treatment.

Objective: The aim of this study was to characterize the polymorphisms of the DC-SIGN (-336 A/G, rs4804803) gene and their association with the immunopathogenicity of dengue fever in Burkina Faso. Methods: A total of three hundred forty-one subjects, patients of all ages have been included in the study: 208 persons presenting clinical signs of dengue fever which were confirmed by diagnostic and 133 Healthy Controls. Genotyping for the CD209 variant (-336 A/G, rs4804803) was carried out using TaqMan SNP Genotyping Assays. Haplotype frequencies were inferred and compared between the study groups. Results: The percentage of men was 61.88% (211/341) and 38.12% (130/341) for women. The highest frequency of dengue fever (77.42%) was noted in patients with age between 20 to 40 years. Around 1.52% of the study population was positive for HIV, 40.55% were carriers of HBV and 3.83% of HCV. Genotype distribution of the CD209 variant (-336 A/G, rs4804803) was in Hardy-Weinberg equilibrium in both patients and controls. The frequency of allele A was higher than allele G;however, statistical analyses showed that there is no significant difference in genotypes GG, AG and AA in patients and controls. Conclusion: This related no significant association with dengue for the variant of ?336 A/G in the DC-SIGN gene in an Ouagadougou population. However, our results offered the SNP frequencies in a West African population, which might be useful for the study of ethnic groups.

Intraocular complications of IFN-α and ribavirin therapy in patients with chronic viral hepatitis C

We report a panel of severe inflammatory and vascular intraocular disorders occurring during interferon-alpha (IFN-α) treatment in eight hepatitis C virus (HCV)-infected patients. These events include three cases of Vogt-Koyanagi-Harada like (VKH) disease (an association of panuveitis, retinal detachment, ear and meningeal detachment and skin and hair changes), two cases of central retinal vein occlusion, one case of central retinal artery occlusion, one case of severe hypertensive retinopathy and one case of bilateral ischemic optic neuropathy with severe visual impairment. Rare as they are, such severe ophthalmological complications require a close follow-up of HCV-infected patients under IFN-α treatment with ophthalmological monitoring if any ocular manifestation occurs.

Effects on coagulation factor production following primary hepatomitogen-induced direct hyperplasia

AIM:To investigate the molecular mechanisms involved in coagulation factor expression and/or function during direct hyperplasia(DH) -mediated liver ***:Direct hyperplasia-mediated liver regeneration was induced in female C57BL/6 mice by administering 1,4-bis[2-(3,5-dichloropyridyloxy) ] benzene(TCPOBOP) ,a representative *** were weighed and sacrificed at various time points [Day 0(D0:prior to injection) ,3 h,D1,D2,D3,and D10] after TCPOBOP administration to obtain liver and blood *** the RNA samples extracted from the liver,a comprehensive analysis was performed on the hepatic gene expression profiling of coagulation-related factors by real-time RT-PCR(fibrinogen,prothrombin,factors Ⅴ,Ⅶ,Ⅷ,Ⅸ,Ⅹ,Ⅺ,Ⅻ,XⅢβ,plasminogen,antithrombin,protein C,protein S,ADAMTS13,and VWF) .The corresponding plasma levels of coagulation factors(fibrinogen,prothrombin,factors Ⅴ,Ⅶ,Ⅷ,Ⅸ,Ⅹ,Ⅺ,Ⅻ,XⅢ,and VWF) were also analyzed and compared with their mRNA ***:Gavage administration of TCPOBOP(3 mg/kg body weight) resulted in a marked and gradual increase in the weight of the mouse livers relative to the total body weight to 220% by D10 relative to the D0(control) *** the peak of liver regeneration(D1 and D2) ,the gene expression levels for most of the coagulationrelated factors(fibrinogen,prothrombin,factors Ⅴ,Ⅶ,Ⅷ,Ⅸ,Ⅺ,Ⅻ,XⅢβ,plasminogen,antithrombin,protein C,ADAMTS13,VWF) were found to be downregulated in a time-dependent manner,and gradually recovered by D10 to the basal *** mRNA levels of factor Ⅹ and protein S failed to show any decrease during the regenerative *** for the plasma levels,5 clotting factors(prothrombin,factors Ⅷ,Ⅸ,Ⅺ,and Ⅻ) demonstrated a significant decrease(P < 0.05) during the regeneration phase compared with *** these 5 factors,factor Ⅸ and factor Ⅺ showed the most dramatic decline in their activities by about 50% at D2 compared to the basal levels,and these reductions in plasma activity for both factors

Selective decrease in colonic CD56^+ T and CD161^+ T cells in the inflamed mucosa of patients with ulcerative colitis

AIM: To investigate the role of local colonic mucosal NK receptor-positive T (NKR+ T) cells in the regulation of intestinal inflammation, we analyzed the population and function of these cells in ulcerative colitis (UC). METHODS: Colonic mucosal tissues were obtained from colonoscopic biopsies of the descending colon from 96 patients with UC (51 endoscopically uninfl amed, 45 inflamed) and 18 normal controls. Endoscopic appearance and histologic score at the biopsied site were determined by Matts’ classification. A single cell suspension was prepared from each biopsy by collagenase digestion. Two NKR+ T cell subsets, CD56+ (CD56+CD3+) T cells and CD161+ (CD161+CD3+) T cells, were detected by flow cytometric analysis. Intracellular cytokine analysis for anti-in? ammatory cytokine interleukin-10 (IL-10) was performed by in vitro stimulation with phorbol-myristate- acetate (PMA) and ionomycin. RESULTS: CD56+ T cells and CD161+ T cells are present in the normal human colon and account for 6.7% and 21.3% of all mononuclear cells, respectively. The populations of both CD56+ T cells and CD161+ T cells were decreased significantly in the inflamed mucosa of UC. In contrast, the frequency of conventional T cells (CD56-CD3+ cells and CD161-CD3+ cells) was similar among the patient and control groups. The populations of NKR+ T cells were correlated inversely with the severity of infl ammation, which was classif ied according to the endoscopic and histologic Matts’ criteria. Interestingly, approximately 4% of mucosal NKR+ T cells expressing IL-10 were detected by in vitro stimulation with PMA and ***: Selective reduction in the population of colonic mucosal NKR+T cells may contribute to the development of intestinal inflammation in UC.
