Targeting endothelial senescence by natural products to improve vascular health

It is well established in experimental animals and Humans that endothelial cells,which cover the luminal surface of all blood vessels,have a pivotal role in the control of vascular *** protective effect of endothelial cells is mostly due to their ability to respond to circulating hormones,autacoids,blood-and platelet-derived factors,and also to blood flow by inducing potent vasoprotectivemechanismsincluding predominantlythe formation of nitric oxide(NO),and,often also to some extent,endothelium-derived hyperpolarization,and prostacyclin(PGI2).Besides being a potent vasodilator,NO also effectively prevents platelet activation and has anti-thrombotic and anti-atherosclerotic *** endothelial dysfunction characterized by a reduced generation of these endothelium-dependent vasodilator mechanisms associated with vascular oxidative stress and the formation of endothelium-derived contracting factors such as contractile prostanoids is often observed in most types of cardiovascular diseases including hypertension,hypercho⁃lesterolemia,diabetes,and also during physiological ageing in both experimental animals and ***-derived polyphenols such as those from grapes,tea,cocoa,and berries have been shown to stimulate the endothelial formation of NO by activating the Src/PI3-kinase/Akt-dependent phosphorylation of endothelial NO synthase(eNOS)and to enhance eNOS *** a consequence,the active polyphenols will promote a sustained forma⁃tion of NO that contributes to protect the vascular ***-rich products have also been shown to improve an established endothelial dysfunction and to delay the onset of the induction of an endothelial dysfunction in several experimental models of cardiovascular diseases including hypertension and in ***,endothelial senescence characterized by cell cycle arrest and the acquisition of a pro-inflammatory and pro-atherothrombotic phenotype has been identified as an early event promotingthe

Evolutionary history and phylogeography of Scots pine (Pinus sylvestris L.) in Europe based on molecular markers

In this review we summarized recent historical records and molecular studies on evolutionary history and phylogeography of Scots pine with focus on the European highly fragmented distribution area of the species. Fossilized pollen, plant micro- and macrofossil records provided evidences on the large-scale species’ range shifts and demographic changes during the Quaternary. Populations of Scots pine were documented both in the glacial (incl. full glaciation) and interglacial periods. Recolonization of Europe after the glaciation originated from the (Sub) Mediterranean areas like the Balkan Peninsula but also from around the Eastern Alps and the surroundings of the Danube plain. Fennoscandia and northern European Baltic regions were most probably colonized from two main directions, from Western Europe and from the Russian Plain. Modern history of Scots pine was hardly affected by anthropogenic activities that started to strengthen in the Bronze and Iron Age. Along with the fossil records, molecular genetic tools were used to infer the origin and putative history including migration, differentiation and demography of the species. In this paper we compiled the major publications (30) of molecular genetic studies of the past 20 years derived from distinctly inherited organelle genomes (mitochondrial, chloroplast, nuclear) revealed by different marker systems (mtDNA-cox1, -nad1, -nad3, -nad7, ISSR, cpSSR, nSSR, B-SAP, SNP). It is important to consider that different phylogeographic patterns can be drawn by the analysis of different DNA marker types. Accordingly the use of more than one marker simultaneously outlines the most sophisticated phylogeographical pattern on the genetic lineages and can reveal high differentiation of the European distribution. Combined marker systems and markers derived from coding sequences have also been used to detect species’ phylogeographic patterns, but these were rarely applied to Scots pine. Chalcone analogue as potent anti-malarial compounds against Plasmodium falciparum:Synthesis,biological evaluation,and docking simulation study

To investigate in vitro antimalarial activity of chalcone derivative compounds against Plasmodium falciparum 3D7 (Pf3D7) strain and in silico antimalarial *** of the chalcone derivatives was conducted via Claisen-Schmidt method using NaOH 60% base as catalyst. An in vitro antimalarial activity assay was carried out according to the Rieckmann method against the chloroquine-sensitive Pf3D7 strain. Molecular docking studies of the prepared compounds were performed using Discovery Studio 3.1 (Accelrys, Inc., San Diego, USA) software to dihydrofolate reductases-thymidylate synthase (PfDHFR-TS) protein with Protein Data Bank ID of *** (sensitive-protein) and ID: *** (resistance-protein).ResultsThis work has successfully synthesized seven chalcone derivatives with a great antimalarial activity. It has been revealed that allyloxy, hydroxy and alkoxy functional groups could increase the antimalarial activity of the chalcone derivatives. The best antimalarial activity of the prepared compounds was possessed by 3b with an IC50 value of 0.59 μM and categorized as an excellent antiplasmodial. Molecular docking studies of 3b showed binding interaction with the amino acid residues such as Ala16, Ile164, Phe58, Tyr170 of the *** protein and also Ala16, Phe58, Ile112, Met55 of the *** *** in vitro antimalarial assay of the prepared chalcone derivative (3a-g) showed an excellent and good antiplasmodial activity against the chloroquine-sensitive Pf3D7 strain. In silico antimalarial studies revealed that 3a-g made binding interaction with both sensitive-protein (***) and resistance-protein (***), which means that they were both active against chloroquine-sensitive and resistant plasmodium strain.

Is autoimmune hepatitis a frequent finding among HCV patients with intense interface hepatitis?

AIM:To evaluate the overlap of autoimmune hepatitis in hepatitis C virus(HCV)-infected patients with intense interface ***:Among 1759 patients with hepatitis C submitted to liver biopsy,92(5.2%) presented intense interface *** patients were evaluated regarding the presence of antinuclear antibody(ANA),anti-smooth muscle antibody(SMA) and anti-liver/kidney microsomal antibody(LKM-1),levels of γ-globulin and histological findings related to autoimmune hepatitis(plasma cell infiltrate and presence of rosettes).RESULTS:Among patients with hepatitis C and intense interface hepatitis there was a low prevalence of autoantibodies(ANA=12%,SMA=5%,LKM-1=0%) and the median γ-globulin level was within the normal *** histological findings of autoimmune disease were observed in only two cases(2%).After applying the score for diagnosis of autoimmune hepatitis,only one patient was classified with a definitive diagnosis of autoimmune *** overlap with autoimmune hepatitis was not the explanation for the intense necroinflammatory activity in patients with chronic hepatitis C we sought to identify the variables associated with this *** presence of intense interface hepatitis was associated with more advanced age,both at the time of infection and at the time of the biopsy,and higher prevalence of blood transfusion and alcohol ***:Although possible,overlap with autoimmune hepatitis is a very rare association in HCV-infected patients with intense interface hepatitis,an unusual presentation which seems to be related to other host variables.

The continuous search for an antimicrobial agent led to the identification of potential antimicrobial biomaterials based on polymers naturals, such as chitosan (CS). However, the mechanism of action of antibacterial activity of CS for gram-positive and gram-negative bacteria was not completely elucidated. The aim of this work is to report the antibacterial activity of CS through ultrastructural analyses of the clinical isolates Staphylococcus aureus and Escherichia coli by Transmission Electron Microscopy. The CS has a bactericidal action against S. aureus and E. coli which alters its cellular ultrastructure, such as with collapsed cell walls, condensed chromatin and the increase of intracellulares structures like vacuoles and cell debris. In this way, the CS represents a potential model for the future design of antibacterial in order to control bacterial resistance of patients in hospital settings.

Assessment of liver ablation using cone beam computed tomography

AIM: To investigate the feasibility and accuracy of cone beam computed tomography(CBCT) in assessing the ablation zone after liver tumor ***: Twenty-three patients(17 men and 6 women, range: 45-85 years old, mean age 65 years) with malignant liver tumors underwent ultrasoundguided percutaneous tumor ablation [radiofrequency(n = 14), microwave(n = 9)] followed by intravenous contrast-enhanced CBCT. Baseline multidetector computed tomography(MDCT) and peri-procedural CBCT images were compared. CBCT image quality was assessed as poor, good, or excellent. Image fusion was performed to assess tumor coverage, and quality of fusion was rated as bad, good, or excellent. Ablation zone volumes on peri-procedural CBCT and post-procedural MDCT were compared using the nonparametric paired Wilcoxon ***: Rate of primary ablation effectiveness was 100%. There were no complications related to ablation. Local tumor recurrence and new liver tumors were found 3 mo after initial treatment in one patient(4%). The ablation zone was identified in 21/23(91.3%) patients on CBCT. The fusion of baseline MDCT and peri-procedural CBCT images was feasible in all patients and showed satisfactory tumor coverage(at least 5-mm margin). CBCT image quality was poor, good, and excellent in 2(9%), 8(35%), and 13(56%), patients respectively. Registration quality between periprocedural CBCT and post-procedural MDCT images was good to excellent in 17/23(74%) patients. The median ablation volume on peri-procedural CBCT and post-procedural MDCT was 30 cm3(range: 4-95 cm3) and 30 cm3(range: 4-124 cm3), respectively(P-value > 0.2). There was a good correlation(r = 0.79) between the volumes of the two techniques. CONCLUSION: Contrast-enhanced CBCT after tumor ablation of the liver allows early assessment of the ablation zone.

Detection of functional diversity gradients and their geoclimatic filters is sensitive to data types(occurrence ***)and spatial scales(sites ***)

Functional diversity(FD)reflects within-and between-site variation of species traits(α-and β-FD,respectively).Understanding how much data types(occurrence-based ***-weighted)and spatial scales(sites ***)change FD and ultimately interfere with the detection of underlying geoclimatic filters is still *** contribute to this debate,we explored the occurrence of 1690 species in 690 sites,abundances of 1198 species in 343 sites,and seven functional traits of the Atlantic Forest woody flora in South *** FD indices were sensitive and dependent on the data type at both scales,with occurrence particularly increasing a richness and dispersion(occurrence>abundance in 80%of the sites)while abundance increased β total,β replacement,and α evenness(abundance>occurrence in 60%of the sites).Furthermore,detecting the effect of geoclimatic filters depended on the data type and was *** the site scale,precipitation seasonality and soil depth had weak effects on α-and β-FD(max.R^(2)=0.11).However,regional-scale patterns of a richness,dispersion,and evenness strongly mirrored the variation in precipitation seasonality,soil depth,forest stability over the last 120 kyr,and cation exchange capacity(correlations>0.80),suggesting that geoclimatic filters manifest stronger effects at the regional ***,the role of edaphic gradients expands the idea of biogeographical filters beyond *** findings caution functional biogeographic studies to consider the effect of data type and spatial scale before designing and reaching ecological conclusions about the complex nature of FD.

The bone marrow niche landscape:a journey through aging,extrinsic and intrinsic stressors in the haemopoietic milieu

Inflammation and its effects in the bone marrow microenvironment represent a paradigmatic condition in which the hematopoietic niche and the immune systems,thought to properly sustain blood cell production and distinguish between friend and foe,can actively sustain a corrupted neighborhood within a chronic aberrant inflamed *** bone marrow niche hijacks the physiologic *** interactions between the hematopoietic stem cells and the niche in the bone marrow are critical determinants of *** examined several approaches to confront the available evidence;three key points emerged,pointing to the chronic inflammation process,especially the chronic infection and systemic inflammatory states,as leading causes of hematopoietic stem cell *** hematopoiesis,defined as a relative expansion of individual clones,is caused by somatic alterations in essential hematopoietic genes,which increase stem cell ***,terminal differentiation plays a significant role in progenitor loss and inflammatory signaling,promoting clonal selection and clonal hematopoiesis *** myeloid malignancies as paradigmatic examples are discussed as a condition associated with inflammation,including the 5q-syndrome,Philadelphia negative myeloproliferative neoplasms,and chronic myeloid *** with increased fitness and hematopoietic stem cell attrition,extrinsic stress,enhanced stressor-specific fitness,and intrinsic defect across the hematopoietic process represent the route for novel insights in defective *** discussion in this review also points out that the hematopoietic niches’inflammatory stimulation may affect differentiation patterns and the function of downstream cells.
