Preclinical and clinical evidence of the association of colibactinproducing Escherichia coli with anxiety and depression in colon cancer

BACKGROUND The association between the intestinal microbiota and psychiatric disorders is becoming increasingly apparent.The gut microbiota contributes to colorectal carcinogenesis(CRC),as demonstrated with colibactin-producing Escherichia coli(CoPEC).AIM To evaluate the association between CoPEC prevalence and anxiety-and depressive-like behaviors with both preclinical and clinical approaches.METHODS Patients followed after a CRC surgery and for whom the prevalence of CoPEC has been investigated underwent a psychiatric interview.Results were compared according to the CoPEC colonization.In parallel C57BL6/J wild type mice and mice with a CRC susceptibility were chronically infected with a CoPEC strain.Their behavior was assessed using the Elevated Plus Maze test,the Forced Swimming Test and the Behavior recognition system PhenoTyper®.RESULTS In a limited cohort,all patients with CoPEC colonization presented with psychiatric disorders several years before cancer diagnosis,whereas only one patient(17%)without CoPEC did.This result was confirmed in C57BL6/J wildtype mice and in a CRC susceptibility mouse model(adenomatous polyposis colimultiple intestinal neoplasia/+).Mice exhibited a significant increase in anxiety-and depressive-like behaviors after chronic infection with a CoPEC strain.CONCLUSION This finding provides the first evidence that CoPEC infection can induce microbiota-gut-brain axis disturbances in addition to its procarcinogenic properties.

Real-time Volume Preserving Constraints for Volumetric Model on GPU

This paper presents a parallel method for simulating real-time 3D deformable objects using the volume preservation mass-spring system method on tetrahedron meshes.In general,the conventional mass-spring system is manipulated as a force-driven method because it is fast,simple to implement,and the parameters can be controlled.However,the springs in traditional mass-spring system can be excessively elongated which cause severe stability and robustness issues that lead to shape restoring,simulation blow-up,and huge volume loss of the deformable object.In addition,traditional method that uses a serial process of the central processing unit(CPU)to solve the system in every frame cannot handle the complex structure of deformable object in real-time.Therefore,the first order implicit constraint enforcement for a mass-spring model is utilized to achieve accurate visual realism of deformable objects with tough constraint error.In this paper,we applied the distance constraint and volume conservation constraints for each tetrahedron element to improve the stability of deformable object simulation using the mass-spring system and behave the same as its real-world counterparts.To reduce the computational complexity while ensuring stable simulation,we applied a method that utilizes OpenGL compute shader,a part of OpenGL Shading Language(GLSL)that executes on the graphic processing unit(GPU)to solve the numerical problems effectively.We applied the proposed methods to experimental volumetric models,and volume percentages of all objects are compared.The average volume percentages of all models during the simulation using the mass-spring system,distance constraint,and the volume constraint method were 68.21%,89.64%,and 98.70%,respectively.The proposed approaches are successfully applied to improve the stability of mass-spring system and the performance comparison from our experimental tests also shows that the GPU-based method is faster than CPU-based implementation for all cases.

Parallel Cloth Simulation Using OpenGL Shading Language

The primary goal of cloth simulation is to express object behavior in a realistic manner and achieve real-time performance by following the fundamental concept of physic.In general,the mass–spring system is applied to real-time cloth simulation with three types of springs.However,hard spring cloth simulation using the mass–spring system requires a small integration time-step in order to use a large stiffness coefficient.Furthermore,to obtain stable behavior,constraint enforcement is used instead of maintenance of the force of each spring.Constraint force computation involves a large sparse linear solving operation.Due to the large computation,we implement a cloth simulation using adaptive constraint activation and deactivation techniques that involve the mass-spring system and constraint enforcement method to prevent excessive elongation of cloth.At the same time,when the length of the spring is stretched or compressed over a defined threshold,adaptive constraint activation and deactivation method deactivates the spring and generate the implicit constraint.Traditional method that uses a serial process of the Central Processing Unit(CPU)to solve the system in every frame cannot handle the complex structure of cloth model in real-time.Our simulation utilizes the Graphic Processing Unit(GPU)parallel processing with compute shader in OpenGL Shading Language(GLSL)to solve the system effectively.In this paper,we design and implement parallel method for cloth simulation,and experiment on the performance and behavior comparison of the mass-spring system,constraint enforcement,and adaptive constraint activation and deactivation techniques the using GPU-based parallel method.

Xylariomycetidae(Ascomycota)is a highly diversified group with variable stromatic characters.Our research focused on inconspicuous stromatic xylarialean taxa from China,Italy,Russia,Thailand and the United Kingdom.Detailed morpho-logical descriptions,illustrations and combined ITS-LSU-rpb2-tub2-tef1 phylogenies revealed 39 taxa from our collections belonging to Amphisphaeriales and Xylariales.A new family(Appendicosporaceae),five new genera(Magnostiolata,Mela-nostictus,Neoamphisphaeria,Nigropunctata and Paravamsapriya),27 new species(Acrocordiella photiniicola,Allocryp-tovalsa sichuanensis,Amphisphaeria parvispora,Anthostomella lamiacearum,Apiospora guiyangensis,A.sichuanensis,Biscogniauxia magna,Eutypa camelliae,Helicogermslita clypeata,Hypocopra zeae,Magnostiolata mucida,Melanostictus longiostiolatus,M.thailandicus,Nemania longipedicellata,N.delonicis,N.paraphysata,N.thailandensis,Neoamphispha-eria hyalinospora,Neoanthostomella bambusicola,Nigropunctata bambusicola,N.nigrocircularis,N.thailandica,Occul-titheca rosae,Paravamsapriya ostiolata,Peroneutypa leucaenae,Seiridium italicum and vamsapriya mucosa)and seven new host/geographical records are introduced and reported.Divergence time estimates indicate that Delonicicolales diverged from Amphisphaeriales+Xylariales at 161(123-197)MYA.Amphisphaeriales and Xylariales diverged 154(117-190)MYA with a crown age of 127(92-165)MYA and 147(111-184)MYA,respectively.Appendicosporaceae(Amphisphaeriales)has a stem age of 89(65-117)MYA.Ancestral character state reconstruction indicates that astromatic,clypeate ascomata with aseptate,hyaline ascospores that lack germ slits may probably be ancestral Xylariomycetidae having plant-fungal endo-phytic associations.The Amphisphaeriales remained mostly astromatic with common septate,hyaline ascospores.Stromatic variations may have developed mostly during the Cretaceous period.Brown ascospores are common in Xylariales,but they first appeared in Amphisphaeriaceae,Melogrammataceae and Sporocadaceae during the early Cretaceous.The asco

Analyzing Changes in Maize Leaves Orientation due to GxExM Using an Automatic Method from RGB Images

The sowing pattern has an important impact on light interception efficiency in maize by determining the spatial distribution of leaves within the canopy.Leaves orientation is an important architectural trait determining maize canopies light interception.Previous studies have indicated how maize genotypes may adapt leaves orientation to avoid mutual shading with neighboring plants as a plastic response to intraspecific competition.The goal of the present study is 2-fold:firstly,to propose and validate an automatic algorithm(Automatic Leaf Azimuth Estimation from Midrib detection[ALAEM])based on leaves midrib detection in vertical red green blue(RGB)images to describe leaves orientation at the canopy level;and secondly,to describe genotypic and environmental differences in leaves orientation in a panel of 5 maize hybrids sowing at 2 densities(6 and 12 plants.m^(−2))and 2 row spacing(0.4 and 0.8 m)over 2 different sites in southern France.The ALAEM algorithm was validated against in situ annotations of leaves orientation,showing a satisfactory agreement(root mean square[RMSE]error=0.1,R^(2)=0.35)in the proportion of leaves oriented perpendicular to rows direction across sowing patterns,genotypes,and sites.The results from ALAEM permitted to identify significant differences in leaves orientation associated to leaves intraspecific competition.In both experiments,a progressive increase in the proportion of leaves oriented perpendicular to the row is observed when the rectangularity of the sowing pattern increases from 1(6 plants.m^(−2),0.4 m row spacing)towards 8(12 plants.m^(−2),0.8 m row spacing).Significant differences among the 5 cultivars were found,with 2 hybrids exhibiting,systematically,a more plastic behavior with a significantly higher proportion of leaves oriented perpendicularly to avoid overlapping with neighbor plants at high rectangularity.Differences in leaves orientation were also found between experiments in a squared sowing pattern(6 plants.m^(−2),0.4 m row spacing),indicating a possible contribution

Anthostomella has long been regarded as a large,but polyphyletic genus in the family Xylariaceae,but species in this group generally lack phylogenetic data.In this study,14 anthostomella-like taxa collected from Italy,were studied using both morphology and molecular data.Single ascospore isolates were obtained and the asexual morphs of five taxa established.The phylogenetic relationships of the xylariaceous taxa were inferred using combined ITS,RPB2,βtubulin and LSU gene regions.We introduce new sequence data for 24 with included the 14 new anthostomella-like taxa.The subfamilies Hypoxyloideae and Xylarioideae within Xylariaceae were recognized as the two major clades with high bootstrap support.Within the two clades 21 subclades were resolved and the anthostomella-like taxa clustered in five of these subclades indicating that the genus is polyphyletic.Anthostomella sensu stricto comprised A.forlicesenica,A.formosa,A.helicofissa,A.rubicola and A.obesa.The A.formosa and A.rubicola collections morphologically closely resemble the type specimens and therefore we designate reference specimens.Three new species Anthostomella helicofissa,A.forlicesenica and A.obesa are also introduced.Four distinct lineages of anthostomella-like taxa correspondent to four new genera,Anthocanalis,Brunneiperidium,Lunatiannulus and Pyriformiascoma,which are also introduced,while one clustered in Astrocystis and is introduced as a new species.Keys to the new anthostomella-like genera and species examined in this study are provided.

Default or profit scoring credit systems?Evidence from European and US peer-to-peer lending markets

For the emerging peer-to-peer(P2P)lending markets to survive,they need to employ credit-risk management practices such that an investor base is profitable in the long run.Traditionally,credit-risk management relies on credit scoring that predicts loans’probability of default.In this paper,we use a profit scoring approach that is based on modeling the annualized adjusted internal rate of returns of loans.To validate our profit scoring models with traditional credit scoring models,we use data from a European P2P lending market,Bondora,and also a random sample of loans from the Lending Club P2P lending market.We compare the out-of-sample accuracy and profitability of the credit and profit scoring models within several classes of statistical and machine learning models including the following:logistic and linear regression,lasso,ridge,elastic net,random forest,and neural networks.We found that our approach outperforms standard credit scoring models for Lending Club and Bondora loans.More specifically,as opposed to credit scoring models,returns across all loans are 24.0%(Bondora)and 15.5%(Lending Club)higher,whereas accuracy is 6.7%(Bondora)and 3.1%(Lending Club)higher for the proposed profit scoring models.Moreover,our results are not driven by manual selection as profit scoring models suggest investing in more loans.Finally,even if we consider data sampling bias,we found that the set of superior models consists almost exclusively of profit scoring models.Thus,our results contribute to the literature by suggesting a paradigm shift in modeling credit-risk in the P2P market to prefer profit as opposed to credit-risk scoring models.

Potential prospects of nanomedicine for targeted therapeutics in inflammatory bowel diseases

Inflammatory bowel diseases (IBDs) such as Crohn's disease are highly debilitating. There are inconsistencies in response to and side effects in the current conventional medications, failures in adequate drug delivery, and the lack of therapeutics to offer complete remission in the presently available treatments of IBD. This suggests the need to explore beyond the horizons of conventional approaches in IBD therapeutics. This review examines the arena of the evolving IBD nanomedicine, studied so far in animal and in vitro models, before comprehensive clinical testing in humans. The investigations carried out so far in IBD models have provided substantial evidence of the nanotherapeutic approach as having the potential to overcome some of the current drawbacks to conventional IBD therapy. We analyze the pros and cons of nanotechnology in IBD therapies studied in different models, aimed at different targets and mechanisms of IBD pathogenesis, in an attempt to predict its possible impact in humans.
