In this.s.udy, indir.ct pr.nted hier.r.hical multilayer.d s.a ffolds.compos.d by a s.lk fibr.in (s.) upper.layer.and an 80/20 (w/w) r.tio of s./ionic-doped β-tr.calcium phos.hate (TCP) bottom layer.wer. developed. Fur.her.or., a compar.tive analys.s.between two types.of s.a ffolds.pr.- duced us.ng di ffer.nt s. concentr.tions. i.e., 8% (w/v) (Hi8) and 16% (w/v) (Hi16) was.per.or.ed. In ter.s.of ar.hitectur. and mor.hology, the pr.duced s.a ffolds.pr.s.nted homogeneous.por.s.ty in both layer. and no di ffer.nces.wer. obs.r.ed when compar.ng both s.a ffolds. This.s.udy inves.igated the acute effects.of gr.en tea extr.ct （GTE） and its.polyphenol cons.ituents. （-）-epigallocatechin-3-gallate （EGCG） and （-）-epicatechin （EC）, on bas.l and s.imulated tes.os.er.ne pr.duction by r.t Leydig cells.in vitr.. Leydig cells.pur.fied in a Per.oll gr.dient wer. incubated for.3 h with GTE, EGCG or.EC and the tes.os.er.ne pr.cur.or.andr.s.enedione, in the pr.s.nce or.abs.nce of either.pr.tein kinas. A （PKA） or.pr.tein kinas. C （PKC） activator.. The r.ver.ibility of the effect was.s.udied by pr.tr.ating cells.for.15 min with GTE or.EGCG, allowing them to r.cover.for.1 h and challenging them for.2 h with human chor.onic gonadotr.pin （hCG）, luteinizing hor.one r.leas.ng hor.one （LHr.）, 22（r.-hydr.xycholes.er.l or.andr.s.enedione. GTE and EGCG, but not EC, inhibited both bas.l and kinas.-s.imulated tes.os.er.ne pr.duction. Under.the pr.tr.atment conditions. the inhibitor. effect of the higher.concentr.tion of GTE/EGCG on hCG/LHr.-s.imulated or.22（r.- hydr.xycholes.er.l-induced tes.os.er.ne pr.duction was.maintained, wher.as.andr.s.enedione-s.ppor.ed tes.os.er.ne pr.duction r.tur.ed to contr.l levels. At the lower.concentr.tion of GTE/EGCG, the inhibitor. effect of thes. polyphenols.on 22（r.-hydr.xycholes.er.l-s.ppor.ed tes.os.er.ne pr.duction was.r.ver.ed. The inhibitor. effects.of GTE may be explained by the action of its.pr.ncipal component, EGCG, and the pr.s.nce of a gallate gr.up in its.s.r.ctur. s.ems.impor.ant for.its.high efficacy in inhibiting tes.os.er.ne pr.duction. The mechanis.s.under.ying the effects.of GTE and EGCG involve the inhibition of the PKA/PKC s.gnalling pathways. as.well as.the inhibition of P450 s.de-chain cleavage enzyme and 17β-hydr.xys.er.id dehydr.genas. function.

Backgr.und： Es.r.diol （E2） is.r.quir.d for.luteolys.s.in cows.and its.injection s.imulates.pr.s.aglandin F2α （PGF2α） r.leas.. The main goal of our.s.udy was.to inves.igate the ability of endometr.al explants.and cells.tr.ated with E2 and the calcium ionophor. （CI） A23187 to s.nthes.ze PGF2α. r.s.lts. Tr.atment with E2 in vivo r.s.lted in a 48.4% incr.as. of PGF2α pr.duction by endometr.al explants.tr.ated in vitr. with A23187. Pr.duction of PGF20 was.better.s.imulated with A23187 at concentr.tions.of 10^-6 and 10^-5 mol/L compar.d with other.concentr.tions.us.d. The concentr.tion of PGF2α for.untr.ated bovine endometr.al cell cultur.s.was.33.1 pg/mL, while for.cultur.s.tr.ated with E2, A23187, or.a combination of E2 and A23187, the PGF2α concentr.tion was.32.5, 92.4 and 145.6 pg/mL, r.s.ectively. Conclus.ons. Tr.atment with A23187 tended to s.imulate PGF20 pr.duction. In the pr.s.nce of E2, A23187 s.gnificantly s.imulated PGF20 s.nthes.s. It appear. that A23187 potentiates.the effects.of E2 with r.s.ect to s.nthes.s.of endometr.al PGF2α in cattle.
