BACKGROUND The utility of D-dimer(DD)as a biomarker for acute aortic dissection(AD)is ***,its predictive value for in-hospital mortality remains uncertain and subject to conflicting *** To conduct a meta-analysis of AD-related in-hospital mortality(ADIM)with elevated DD *** We searched PubMed,Scopus,Embase,and Google Scholar for AD and ADIM literature through May *** was assessed using I2 statistics and effect size(hazard or odds ratio)analysis with random-effects *** size,study type,and patients’mean age were used for subgroup *** significance threshold was Pvalence of ADIM was 20%(95%CI:15%-25%).Despite comparable demographic characteristics and comorbidities,elevated DD values were associated with higher ADIM risk(unadjusted effect size:1.94,95%CI:1.34-2.8;adjusted effect size:1.12,95%CI:1.05-1.19,Pvolving patients with a mean age ofved in studies with a mean age>60 *** and larger sample size studies(n>250)demonstrated a heightened likelihood of ADIM associated with elevated DD levels(effect size:2.57,95%CI:1.30-5.08,Pvs effect size:1.05,95%CI:1.00-1.11,P=0.05,respectively).CONCLUSION Our meta-analysis shows elevated DD increases in-hospital mortality risk in AD patients,highlighting the need for larger,prospective studies to improve risk prediction models.

AIM： To study an inherent effect of insulin on small intestinal transit and to explore involvement of various systems/mechanisms in normal mice. METHODS： Insulin at the doses of 2 μU/kg, 2 mU/kg, 2 U/kg or vehicle was subcutaneously administered to four groups of overnight fasted normal male mice. Blood glucose （BG） levels were measured 2 min before insulin administration and 2 min before sacrificing the animals for the measurement of small intestinal transit （SIT）. Charcoal meal was administered （0.3 mL） intragastrically 20 min after insulin administration and animals were sacrificed after 20 min and SIT was determined. For exploration of the various mechanisms involved in insulin-induced effect on SIT, the dose of insulin which can produce a significant acceleration of SIT without altering BG levels was determined. The following drugs, atropine （1 mg/kg）, clonidine （0.1 mg/kg）, ondansetron （1 mg/kg）, naloxone （5 mg/kg）, verapamil （8 mg/kg） and glibenclamide （10 mg/kg）, were administered intravenously 10 min prior to the administration of insulin （2 μU/kg）. RESULTS： The lower doses of insulin （2 μU/kg and 2 mU/kg） produced a significant acceleration of SIT from 52.0% to 70.7% and 73.5% without lowering blood glucose levels （P〈0.01）, while the highest dose of insulin （2 U/kg） produced a fall in blood glucose levels which was also associated with significant acceleration of SIT （P〈0.01）. After pretreatment of insulin （2 μU/kg） group with atropine, insulin could reverse 50% of the inhibition produced by atropine. In clonidine-pretreated group, insulin administration could reverse only 37% of the inhibition produced by clonidine and inhibition of SIT was significant compared with vehicle ＋ insulintreated group, i.e. from 74.7% to 27.7% （P〈0.01）. In ondansetron-pretreated group, insulin administration could produce only mild acceleration of SIT （23.5%）. AIM: To investigate the inhibitory action of diet-derived phenolic compound gallic acid(GA) against HCT-15 colon cancer ***: The antiproliferative effect of GA against colon cancer cells was determined by performing thiazolyl blue tetrazolium bromide(MTT) assay. The colony forming ability of GA treated colon cancer cells was evaluated using the colony forming assay. The cell cycle changes induced by GA in HCT-15 cells were analyzed by propidium iodide staining. Levels of reactive oxygen species(ROS) and mitochondrial membrane potential of HCT-15 exposed to GA was assessed using 2',7'-dichlorfluorescein-diacetate and rhodamine-123 respectively, with the help of flow cytometry. Morphological changes caused by GA treatment in the colon cancer cells were identified by scanning electron microscope and photomicrograph examination. Apoptosis was confirmed using flow cytometric analysis of GA treated HCT-15 cells after staining with ***: MTT assay results illustrated that GA has an inhibitory effect on HCT-15 cells with IC50 value of 740 μmol/L. A time-dependent inhibition of colony formation was evident with GA treatment. Cell cycle arrest was evident from the accumulation of GA treated HCT-15 cells at sub-G1 phase(0.98 ± 1.03 vs 58.01 ± 2.05)with increasing exposure time. Flow cytometric analysis of GA treated HCT-15 cells depicted early events associated with apoptosis like lipid layer breakage and fall in mitochondrial membrane potential apart from an increase in the generation of ROS which were in a time dependent manner. SEM and photomicrograph images of the GA-treated cells displayed membrane blebbing and cell shrinking characteristics of apoptosis. Further apoptosis confirmation by Yo-Pro-1 staining also showed the time-dependent increase of apoptotic cells after ***: These results show that GA induced ROS dependent apoptosis and inhibited the growth of colon cancer cells.

Withania somnifera (Ashwagandha):a Novel Source of L-asparaginase

Different parts of plant species belonging to Solanaceae and Fabaceae families were screened for L-asparaginase enzyme （E.C.3.***.1.1.）. Among 34 plant species screened for L-asparaginase enzyme. Withania somnifera L. was identified as a potential source of the enzyme on the basis of high specific activity of the enzyme. The enzyme was purified and characterized from W. somnifera, a popular medicinal plant in South East Asia and Southern Europe. Purification was carried out by a combination of protein precipitation with ammonium sulfate as well as Sephadex-gel filtration. The purified enzyme is a homodimer, with a molecular mass of 72 ± 0.5 kDa as estimated by sodium dodecyl sulfate-polyacrylamide gel electrophoresisand size exclusion chromatography. The enzyme has a pH optimum of 8.5 and an optimum temperature of 37 ℃. The Km value for the enzyme is 6.1 × 10^-2 mmol/L. This is the first report for L-asparaginase from W. somnifera, a traditionally used Indian medicinal plant.

Carvacrol Promotes Cell Cycle Arrest and Apoptosis through PI3K/AKT Signaling Pathway in MCF-7 Breast Cancer Cells

Objective:To examine the role of carvacrol in modulating PI3 K/AKT signaling involved in human breast cancer pathogenesis using in vitro experimental model MCF-7 ***:MTT and lactate dehydrogenase assays were performed with cells treated with different doses of carvacrol(0–250μmol/L)at different time points(24 and 48 h).The nuclear morphology was assessed in MCF-7 cells with propidium iodide(PI)and acridine orange/ethidium bromide(AO/EB)staining and analyzed by fluorescence *** like cell cycle arrest and apoptosis were observed by flow cytometric analysis and expressions of p-Rb,cyclin D1,cyclin-dependent kinase 4(CDK4),CDK6,Bax,Bcl-2,PI3 K/p-AKT were analyzed by ***:Carvacrol significantly reduced cell viability with the half maximal inhibitory concentration value of 200μmol/L at 24 and 48 h(Pvacrol in MCF-7 cells(Pvacrol treatment significantly inhibited PI3 K/p-AKT protein expressions leading to induction of apoptosis mediated by decreased Bcl2 and increased Bax protein ***,Annexin v/PI staining by FACS analysis,dual staining by AO/EB and PI staining studies suggested induction of apoptosis by carvacrol through PI3 K/Akt signaling pathway in MCF-7 ***:Carvacrol significantly inhibited the breast cancer MCF-7 cell proliferation and induced apoptosis via suppressing PI3/AKT signaling pathway.
