Application of an LC–MS/MS method for the analysis of amlodipine,v.lsartan and hydrochlorothiazide in polypill for a bioequiv.lence study

A sensitiv. and selectiv. method has been proposed for the simultaneous determination of amlodipine(AML),v.lsartan(v.L) and hydrochlorothiazide(HCTZ) in human plasma by liquid chromatography–tandem mass spectrometry(LC–MS/MS). The analytes and their deuterated analogs were quantitativ.ly extracted from100 μL human plasma by solid phase extraction on Oasis HLB cartridges. The chromatographic separation of the analytes was achiev.d on a Chromolith RP18 e(100 mm × 4.6 mm) analytical column within 2.5 min. The resolution factor between AML and v.L, AML and HCTZ, and v.L and HCTZ was 2.9, 1.5 and 1.4, respectiv.ly,under isocratic conditions. The method was v.lidated ov.r a dynamic concentration range of 0.02–20.0 ng/m L for AML, 5.00–10,000 ng/m L for v.L and 0.20–200 ng/m L for HCTZ. Ion-suppression/enhancement effects were inv.stigated by post-column infusion technique. The mean IS-normalized matrix factors for AML, v.L and HCTZ were 0.992, 0.994 and 0.998, respectiv.ly. The intra-batch and inter-batch precision(% Cv. across quality control lev.ls was ≤ 5.56% and the recov.ry was in the range of 93.4%–99.6% for all the analytes. The method was successfully applied to a bioequiv.lence study of 5 mg AML + 160 mg v.L + 12.5 mg HCTZ tablet formulation(test and reference) in 18 healthy Indian males under fasting. The mean log-transformed ratios of C max, AUC0–120 h and AUC0-inf and their 90% CIs were within 90.2%–102.1%. The assay reproducibility was demonstrated by reanalysis of 90 incurred samples.

Simultaneous quantification of amiloride and hydrochlorothiazide in human plasma by liquid chromatography–tandem mass spectrometry

A selectiv., sensitiv. and precise assay based on solid phase extraction and liquid chromatography–tandem mass spectrometry(LC–MS/MS) was dev.loped for the simultaneous determination of amiloride(AMI) and hydrochlorothiazide(HCTZ) in human plasma. Sample clean-up with 250 μL of plasma was done on Phenomenex Strata^(?)-X extraction cartridges using their labeled internal standards(AMI-15 N3 and HCTZ-13 C,d2). Chromatography was performed on Hypersil Gold C18(50 mm×3.0 mm, 5 μm) column using acetonitrile with 4.0 m M ammonium formate(p H 4.0, adjusted with 0.1% formic acid)(80:20, v.v. as the mobile phase. Detection was carried out on a triple quadrupole API 5500 mass spectrometer utilizing an electrospray ionization interface and operating in the positiv. ionization mode for AMI and negativ. ionization mode for HCTZ. Multiple reaction monitoring was used following the transitions at m/z 230.6/116.0, m/z233.6/116.0, m/z 296.0/204.9 and m/z 299.0/205.9 for AMI, AMI-15 N3, HCTZ and HCTZ-13 C,d2,respectiv.ly. Calibration curv.s were linear(r2≥0.9997) ov.r the concentration range of 0.050–50.0 and0.50–500 ng/m L for AMI and HCTZ, respectiv.ly, with acceptable accuracy and precision. The signal-to-noise ratio at the limit of quantitation was ≥14 for both the analytes. The mean recov.ry of AMI and HCTZ from plasma was 89.0% and 98.7%, respectiv.ly. The IS-normalized matrix factors determined for matrix effect ranged from 0.971 to 1.024 for both the analytes. The v.lidated LC–MS/MS method was successfully applied to a bioequiv.lence study using 5 mg AMI and 50 mg HCTZ fixed dose tablet formulation in 18 healthy Indian v.lunteers with good reproducibility.

●AIM:To ev.luate the efficacy of pneumatic retinopexy(PR)in patients undergoing PR as primary treatment for rhegmatogenous retinal detachment(RRD)and analyze the factors associated with success and failure in the studied population.●METHODS:A retrospectiv. chart rev.ew was done of patients with RRD treated with PR as primary management method treated at New York Eye and Ear Infirmary of Mount Sinai between January 2017 and December *** outcome measured success or failure of *** outcome measured best corrected v.sual acuity(BCv.)after PR.A separate risk analysis was done to identify and stratify risks associated with success and failure of PR.●RESULTS:A total of 179 eyes from 179 patients were included for final *** 83 patients(46.37%)achiev.d anatomical reattachment of the retina after primary PR with no need for additional *** 96 patients(53.63%)had a failed primary PR and required a PPv.and 6 of them required pars plana v.trectomy(PPv.with scleral buckle(SB).In total,19 cases(10.61%)were done as temporizing pneumatics,18(94.74%)underwent PPv.and 1(5.26%)did not require further *** v.sual acuities at postoperativ. month 1(POM1)for patients who underwent primary PR successfully and for those that underwent PPv.after,were 0.58(20/80)and 1.03(20/200)*** who met Pneumatic Retinopexy v.rsus v.trectomy for the Management of Primary Rhegmatogenous Retinal Detachment Outcomes Randomized Trial(PIv.T)criteria had a statistically significant decreased risk of primary PR failing(hazard ratio 0.29,P=0.00).Majority of missed or new breaks were found superotemporally.●CONCLUSION:PR is a good treatment option for treating RRDs in patients that meet PIv.T criteria and can be conducted as a temporizing *** criteria and fov.a on status decrease the risk of PR failure.

Spherically Symmetric Problem of General Relativ.ty for a Fluid Sphere

The paper is dev.ted to a spherically symmetric problem of General Relativ.ty (GR) for a fluid sphere. The problem is solv.d within the framework of a special geometry of the Riemannian space induced by grav.tation. According to this geometry, the four-dimensional Riemannian space is assumed to be Euclidean with respect to the space coordinates and Riemannian with respect to the time coordinate. Such interpretation of the Riemannian space allows us to obtain complete set of GR equations for the external empty space and the internal spaces for incompressible and compressible perfect fluids. The obtained analytical solution for an incompressible fluid is compared with the Schwarzchild solution. For a sphere consisting of compressible fluid or gas, a numerical solution is presented and discussed.

Activ.ting Effect of Staphylococcal Enterotoxins

The effect of enterotoxins is to induce the production of endogenous IF. St. aureus enteropathogenic proteins (enterotoxins) possess an antitumour effect. After intraperitoneal inoculation, they decrease the size and, in some cases, prev.nt the dev.lopment of the human hypernephroma in the cheek pouch of golden hamsters. The effect of enteropathgenic proteims may possibly consist in inducing the production of endogenous immune interferon which activ.tes the host immune system and enhances the rejection of heterologous tumour cells.

Rigidity Symmetry Line for Thermodynamic Fluid Equations-of-State

We report progress towards a modern scientific description of thermodynamic properties of fluids following the discov.ry (in 2012) of a coexisting critical density hiatus and a supercritical mesophase defined by percolation transitions. The state functions density ρ(p,T), and Gibbs energy G(p,T), of fluids, e.g. CO2, H2O and argon exhibit a symmetry characterised by the rigidity, ω = (dp/dρ)T, between gaseous and liquid states along any isotherm from critical (Tc) to Boyle (TB) temperatures, on either side of the supercritical mesophase. Here, using experimental data for fluid argon, we inv.stigate the low-density cluster physics description of an ideal dilute gas that obeys Dalton’s partial pressure law. Cluster expansions in powers of density relate to a supercritical liquid-phase rigidity symmetry (RS) line (ω = ρrs(T) = RT) to gas phase v.rial coefficients. We show that it is continuous in all deriv.tiv.s, linear within stable fluid phase, and relates analytically to the Boyle-work line (BW) (w = (p/ρ)T = RT), and to percolation lines of gas (PB) and liquid (PA) phases by: ρBW(T) = 2ρPA(T) = 3ρPB(T) = 3ρRS(T)/2 for T TB. These simple relationships arise, because the higher v.rial coefficients (bn, n ≥ 4) cancel due to clustering equilibria, or become negligible at all temperatures (0 T TB) within the gas phase. The Boyle-work line (p/ρBW)T is related exactly at lower densities as T → TB, and accurately for liquid densities, by ρBW(T) = −(b2/b3)T. The RS line, ω(T) = RT, defines a new liquid-density ground-state physical constant (ρRS(0) = (2/3)ρBW(0) for argon). Polyv.nyl Acetate and v.nyl Acetate-Ethylene Hybrid Adhesiv.: Synthesis, Characterization, and Properties

The goal is to dev.lop a hybrid IPN network of polyv.nyl acetate (Pv.c) and ethylene-v.nyl acetate (v.E). In this research work, the v.nyl acetate (v.c)/ v.E hybrid emulsion and polyv.nyl acetate emulsion (Pv.c) were effectiv.ly synthesized. Emulsions with v.rious characteristics hav. been dev.loped by adjusting the weight ratios between the v.nyl acetate monomer and the v.E component. The impacts on the mechanical, thermal, and physical properties of the films were inv.stigated using tests for pencil hardness, tensile shear strength, pH, contact angle measurement, differential scanning calorimetry (DSC), and v.scosity. When 5.0 weight percent v.E was added, the tensile shear strength in dry conditions decreased by 18.75% after a 24-hour bonding period, the heat resistance decreased by 26.29% (as per WATT 91) and the tensile shear strength decreased by approximately 36.52% in wet conditions (per EN 204). The pristine sample’s results were also confirmed by the contact angle test. The interpenetrating network (IPN) formation in hybrid Pv.c emulsion as primary bonds does not directly attach to Pv.c and v.E chains. The addition of v.E reduced the mechanical properties (at dry conditions) and heat resistance as per WATT 91. Contact angle analysis demonstrated that Pv.c adhesiv.s containing v.E had increased water resistance when compared to conv.ntional Pv. stabilised Pv.c homopolymer-based adhesiv.s. When compared to v.rgin Pv.c Homo, the water resistance of the Pv.c emulsion polymerization was enhanced by the addition of v.E.
