Connexin 43 (CX43) is one of the major components of gap junctions, the structures responsible for the intercellular communication and transmission of the electrical impulse in the left v.ntricle. There is limited information on the histological changes of CX43 with age and their effect on electrophysiology, especially in humans. Here, we analyzed left v.ntricular biopsies from liv.ng donors starting at midlife to characterize age-related CX43 remodeling. We assessed its quantity, degree of lateralization, and spatial heterogeneity together with fibrotic deposition. We observ.d no significant age-related remodeling of CX43. Only spatial heterogeneity increased slightly with age, and this increase was better explained by biological age than by chronological age. Importantly, we found that CX43 features v.ried considerably among indiv.duals in our population with no relev.nt relationship to age or fibrosis content, in contrast to animal species. We used our experimental results to feed computational models of human v.ntricular electrophysiology and to assess the effects of interindiv.dual differences in specific features of CX43 and fibrosis on conduction v.locity, action potential duration, and arrhythmogenicity. We found that larger amounts of fibrosis were associated with the highest arrhythmic risk, with this risk being increased when fibrosis deposition was combined with a reduction in CX43 amount and/or with an increase in CX43 spatial heterogeneity. These mechanisms underlying high arrhythmic risk in some indiv.duals were not associated with age in our study population. In conclusion, our data rule out CX43 remodeling as an age-related arrhythmic substrate in the population beyond midlife, but highlight its potential as a proarrhythmic factor at the indiv.dual lev.l, especially when combined with increased fibrosis.

Temporomandibular disorders (TmD) negativ.ly affect quality of life, causing pain and restricted jaw mov.ments. This study ev.luates the effectiv.ness of Transcutaneous electrical nerv. Stimulation (TenS) on TmD patients before orthodontic treatment at the Juarez Autonomous Univ.rsity of Tabasco. A quantitativ., quasi-experimental study was conducted with 30 patients who met the inclusion criteria: aged 18 to 65 years, with TmJ pain symptoms, diagnosed with temporomandibular disorders using the Helkimo index, and who had prov.ded informed consent for the study. The range of mandibular mov.ment and the v.sual Analog Scale (v.S) were measured to record v.lues before therapy. The electrodes of the TenS therapy were positioned on both sides of the jaw in the preauricular area and the masseter muscle, the therapy was administered for 40 minutes, utilizing a frequency of 2 - 4 Hz and a pulse duration of 300 microseconds. Following this, the range of mandibular mov.ment and the v.S were reassessed to ev.luate the outcomes., significantly improv.d maximum mouth opening (37.7 ± 6.4 mm to 44.6 ± 5.88 mm), right laterality (3.86 ± 1.57 mm to 5.43 ± 1.21 mm), left laterality (3.13 ± 1.63 mm to 4.53 ± 1.61 mm), protrusion (4.01 ± 1.7 mm to 5.36 ± 1.32 mm), and pain perception (4.56 ± 2.045 to 2.03 ± 1.5) (p enS therapy enhances mandibular mov.ment and reduces pain, making it a well-tolerated, non-inv.siv. complementary treatment for TmD.
