The translocon on the outer membrane of mitochondria （TOM） facilitates the import of nuclear-encoded proteins. The principal machinery of mitochondrial protein transport seems conserved in eukaryotes; however, divergence in the composition and structure of TOM components has been observed between mammals, yeast, and plants. TOM9, the plant homolog of yeast Tom22, is significantly smaller due to a truncation in the cytosolic receptor domain, and its precise function is not understood. Here we provide evidence showing that TOM9.2 from Arabidopsis thaliana is involved in the formation of mature TOM com- plex, most likely by influencing the assembly of the pore-forming subunit TOM40. Dexamethasone- induced RNAi gene silencing of TOM9.2 results in a severe reduction in the mature TOM complex, and the assembly of newly imported TOM40 into the complex is impaired. Nevertheless, mutant plants are fully viable and no obvious downstream effects of the loss of TOM complex, i.e., on mitochondrial import ca- pacity, were observed. Furthermore, we found that TOM9.2 can bind calmodulin （CAM） in vitro and that CaM impairs the assembly of TOM complex in the isolated wild-type mitochondria, suggesting a regula- tory role of TOM9.2 and a possible integration of TOM assembly into the cellular calcium signaling network.

Effect of feeding broilers diets differing in susceptible phytate content

Measurements of total phytate phosphorus content of diets may be deceptive as they do not indicate substrate availability for phytase; it may be that measurements of phytate susceptible to phytase effects are a more accurate measure of phosphorus(P) availability to the bird. To verify this hypothesis, an experiment was conducted to compare diets formulated to contain either high or low susceptible phytate, supplemented with either 0 or 500 FTU/kg phytase. Susceptible phytate was determined by exposing the feed samples to conditions that mimicked the average pH of the proximal gastrointestinal tract(pH 4.5) and the optimum temperature for phytase activity(37℃) and then measuring phytate dissolved. Ross 308 birds(n = 240) were fed one of 4 dietary treatments in a 2 x 2 facto.ial design; 2 diets with high(8.54 g/kg, 57.90% of total phytate) or low(5.77 g/kg, 46.33% of total phytate) susceptible phytate, containing 0 or 500 FTU/kg phytase. Diets were fed to broilers(12 replicate pens of 5 birds per pen) from d 0 to 28 post hatch. Birds fed diets high in susceptible phytate had greater phytate hydrolysis in the gizzard(P ve of phytase presence. Birds fed the high susceptible diets also had greater P solubility in the gizzard and Ca and P solubility in the jejunum and ileum(P vels. As climatic changes and human uses intensify,resource managers and other decision makers are taking actions to either avoid or respond to ecosystem tipping points,or dramatic shifts in structure and function that are often costly and hard to *** indicates that explicitly addressing tipping points leads to improved management *** on theory and examples from marine systems,we distill a set of seven principles to guide effective management in ecosystems with tipping points,derived from the best available *** principles are based on observations that tipping points(1)are possible everywhere,(2)are associated with intense and/or multifaceted human use,(3)may be preceded by changes in earlywarning indicators,(4)may redistribute benefits among stakeholders,(5)affect the relative costs of action and inaction,(6)suggest biologically informed management targets,and(7)often require an adaptive response to *** suggest that early action to preserve system resilience is likely more practical,affordable,and effective than late action to halt or reverse a tipping *** articulate a conceptual approach to management focused on linking management targets to thresholds,tracking early-warning signals of ecosystem instability,and stepping up investment in monitoring and mitigation as the likelihood of dramatic ecosystem change *** approach can simplify and economize management by allowing decision makers to capitalize on the increasing value of precise information about threshold relationships when a system is closer to tipping or by ensuring that restoration effort is sufficient to tip a system into the desired regime.
