hronic rejection is the main factor to result in the loss of renal allograft In order to look for a potential therapy chronic rejection, we investigated the efficacy of estradiol on preventing renal chronic rejection The kidneys of female F344 rats were orthotopically transplanted into ovatiectomized female Lewis rats and treated for 16 weeks with either estradiol or vehicle Compared with controls treated with vehicle, estradiol treatment reduced urinary protein excretion, glomerular sclerosis, interstitial infiltration and fibrosis, vascular lesions, in parallel to a reduced ICAM 1 and TGF β mRNA expression Our results suggested that estrodiol could significantly decrease the progression of chronic rejection, at least in female recipients, and the reduced adhesion molecule and TGF β gene expression may be involved in the mechanism for estradiol to prevent chronic rejection

Background Increased risk of bladder cancer has been reported in diabetic patients. This study was to investigate the roles of mitogen-activated protein kinase kinase （MEK） 1 and 2 in the regulation of human insulin- and insulin glargine-induced proliferation of human bladder cancer T24 cells. Methods In the absence or presence of a selective inhibitor for MEK1 （PD98059） or a specific siRNA for MEK2 （siMEK2）, with or without addition of insulin or glargine, T24 cell proliferation was evaluated by cell counting kit （CCK）-8 assay. Protein expression of MEK2, phosphorylation of ERK1/2 and Akt was analyzed by Western blotting. Results T24 cell proliferation was promoted by PD98059 at 5-20 IJmol/L, inhibited by siMEK2 at 25-100 nmol/L. PD98059 and siMEK2 remarkably reduced phosphorylated ERKI/2. Insulin- and glargine-induced T24 cell proliferation was enhanced by PD98059, suppressed while not blocked by siMEK2. Insulin- and glargine-induced ERKI/2 activation was blocked by PD98059 or siMEK2 treatment, whereas activation of Akt was not affected. Conclusion MEK1 inhibits while MEK2 contributes to normal and human insulin- and insulin glargine-induced human bladder cancer T24 cell proliferation.

Background Increased levels of plasma lipopolysaccharide （LPS） have been found in obesity and diabetes patients. This study was to investigate the effect of LPS on pancreatic beta-cell viability and the involvement of caspase 3 in NIT-1 cell line. Methods Mouse insulinoma NIT-1 cells were treated with LPS for the indicated time and dose. Cell viability was measured by cell counting kit-8 reagent. Toll-like receptor 4 （TLR4）, caspase 3 and cleaved caspase 3 were detected by Western blotting. Insulin was determined by radioimmunoassay （RIA）. Results LPS promoted NIT-1 cell proliferation at 1 μg/ml, peaked at 72 hours of incubation. A reduction in cleavage of caspase 3 was observed upon LPS treatment. Bay11-7082, a specific inhibitor of nuclear factor （NF）-κB, blunted LPS-induced inhibition of caspase 3 cleavage. Reduction in chronic insulin secretion was observed after treatment with LPS at 1 μg/ml for 48 and 72 hours, not for 24 hours. TLR4 protein was upregulated when NIT-1 cells were treated with LPS at 1 μg/ml for 24 hours. Conclusions LPS promotes early NIT-1 cell proliferation in association with NF-KB-mediated inhibition of caspase 3 cleavage. LPS exerts a time-dependent inhibitory effect on chronic insulin secretion from NIT-1 cells.

The safety and durability of lithium-ion batteries under mechanical constraints depend significantly on electrochemical,thermal,and mechanical fields in *** and quantifying the multi-field coupling behaviors requires interdisciplinary ***,we design experiments under mechanical constraints and introduce an in-situ analytical framework to clarify the complex interaction mechanisms and coupling degrees among multi-physics *** proposed analytical framework integrates the parameterization of equivalent models,in-situ mechanical analysis,and quantitative assessment of coupling *** results indicate that the significant impact of pressure on impedance at low temperatures results from the diffusion-controlled step,enhancing kinetics when external pressure,like 180 to 240 k Pa at 10℃,is *** diversity in control steps for the electrochemical reaction accounts for the varying impact of pressure on battery performance across different *** thermal expansion rate suggests that the swelling force varies by less than 1.60%per unit of elevated temperature during the lithiation *** introducing a composite metric,we quantify the coupling correlation and intensity between characteristic parameters and physical fields,uncovering the highest coupling degree in electrochemical-thermal *** results underscore the potential of analytical approaches in revealing the mechanisms of interaction among multi-fields,with the goal of enhancing battery performance and advancing battery management.

Monolithic refractory castables comprising a hydraulic bond are still used in a vast majority of cases because of their flexibility and robustness,despite many developments for chemical as well as non-cement castable *** drying can however be a challenge,in particular for deflocculated dense castables of the low cement castable *** publications have been released on this topic for the last ten years,but they often focused on the drying mechanisms or on the addition of drying *** paper presents some experimental results on the effect of the composition on the drying properties,especially on the effect of silicon carbide,used for its high thermal conductivity,and on the matrix *** also introduces two laboratory tests to study and iteratively improve the drying schedule of a given castable *** results show that the spalling resistance and the vapor pressure build-up are significantly influenced by the *** is also shown that the castable properties after drying can be altered if the heating rate is very high.

Mescaline,among the earliest identified natural hallucinogens,holds great potential in psychotherapy ***,despite the existence of a postulated biosynthetic pathway for more than half a century,the specific enzymes involved in this process are yet to be *** this study,we investigated the cactus Lophophora williamsii(Peyote),the largest known natural producer of the phenethylamine *** employed a multi-faceted approach,combining de novo whole-genome and transcriptome sequencing with comprehensive chemical profiling,enzymatic assays,molecular modeling,and pathway engineering for pathway *** identified four groups of enzymes responsible for the six catalytic steps in the mescaline biosynthetic pathway,and an N-methyltransferase enzyme that N-methylates all phenethylamine intermediates,likely modulating mescaline levels in ***,we reconstructed the mescaline biosynthetic pathway in both Nicotiana benthamiana plants and yeast cells,providing novel insights into several challenges hindering complete heterologous mescaline *** together,our study opens up avenues for exploration of sustainable production approaches and responsible utilization of mescaline,safeguarding this valuable natural resource for future generations.

Implementation of the Flood Risk Management Directive in Selected European Countries

Flood events are a recurrent threat to economic developments and can jeopardize human existence. In order to reduce the flood risks in Europe on a sustainable basis, the European Union adopted the Flood Risk Management Directive in 2007. The implementation of the directive in the national laws of all member states has laid the foundation for intergovernmental integrated flood risk management on an European scale. The following article is intended to compare and briefly assess the implementation process on inland waters at the technical level in selected European countries in the middle of the process. It covers the different basic structural, methodological, and data conditions for preparing the preliminary flood risk assessment, the flood hazard maps, the flood risk maps, and the flood risk management plans as a result of two EU projects. The technical differences in the various European countries need to be reduced in the next cycles of implementation of the Flood Risk Management Directive.

Necessary and sufficient conditions are obtained for the boundedness of Berezin transformation on Lebesgue space Lp(B,dVβ) in the real unit ball B in Rn. As an application, we prove that Gleason type problem is solvable in hyperbolic harmonic Bergman spaces. Furthermore we investigate the boundary behavior of the solutions of Gleason type problem.
