Amyloid-b-protein(Ab),the key component of senile plaques in Alzheimer’s disease(AD)brain,is produced from amyloid precursor protein(APP)by cleavage of b-secretase and then *** adaptor proteins with phosphotyrosine-binding(PTB)domains,including Dab(gene:DAB)and Numb(gene:NUMB),can bind to and interact with the conserved YENPTY-motif in the APP *** we describe,for the first time,the effects of RNAi knock-down of Dab and Numb expression on APP processing and Ab *** knock-down of Dab and Numb in H4 human neuroglioma cells stably transfected to express either FL-APP(H4-FL-APP cells)or APP-C99(H4-APP-C99 cells)increased levels of APP-C-terminal fragments(APP-CTFs)and lowered Ab levels in both cell lines by inhibiting g-secretase cleavage of ***,RNAi knock-down of APP also reduced levels of Numb in H4-APP *** findings suggest that pharmacologically blocking interaction of APP with Dab and Numb may provide novel therapeutic strategies of *** notion of attenuating g-secretase cleavage of APP via the APP adaptor proteins,Dab and Numb,is particularly attractive with regard to therapeutic potential,given that side effects of gsecretase inhibition owing to impaired proteolysis of other g-secretase substrates,***,might be avoided.

Spatiality of the urban commons:Typo-morphology of the open common spaces in New Belgrade mass housing blocks

The article examines the existing infrastructure of open common spaces within two New Belgrade mass housing blocks(Blocks 23 and 70a)through a typo-morphological *** spaces between the buildings,although the most neglected,underused,and deteriorated components of mass housing neighbourhoods,are at the same time crucial to the quality,vitality and integrated governance of these *** represent the primary tangible commons in cities and *** question of urban commons is increasingly present in scientific literature,urban and architectural ***,approaches exploring the spatiality of the urban commons are scarce,leading to insufficient understanding of the spatial aspect and potentials of the already existing ***,this study includes(1)identification,typological decoding and classification of the common spaces,focusing on the case of New Belgrade blocks,followed by(2)analysis of the spatial patterns and integration of the identified spaces within the *** study confirms the complexity and diverse typology of the common *** finds that the in-between,common spaces contribute to higher integration of different segments of the *** open common spaces have an essential role in humanisation of the blocks,and thus the quality of life in the blocks as integrated *** findings indicate that the spatial setting of the open common spaces in New Belgrade blocks allows for(re)emergence of collective practices,leading to inclusive and integrated rehabilitation of the neighbourhoods.

Plant diversity and community history shift colonization success from early- to mid-successional species

Genetic diversity for mycorrhizal symbiosis and phosphate transporters in rice

Phosphorus （P） is a major plant nutrient and developing crops with higher P-use efficiency is an important breeding goal. In this context we have conducted a comparative study of irrigated and rainfed rice varieties to assess genotypic differences in colonization with arbuscular mycorrhizal （AM） fungi and expression of different P trans- porter genes. Plants were grown in three different soil samples from a rice farm in the Philippines. The data show that AM symbiosis in all varieties was established after 4 weeks of growth under aerobic conditions and that, in soil derived from a rice paddy, natural AM populations recovered within 6 weeks. The analysis of AM marker genes （AM1, AM3, AM14） and P transporter genes for the direct Pi uptake （PT2, PT6） and AM-mediated pathway （PTll, PT13） were largely in agreement with the observed root AM colonization providing a useful tool for diversity studies. Interestingly, delayed AM colonization was observed in the aus-type rice varieties which might be due to their different root structure and might confer an advantage for weed competition in the field. The data further showed that P-starvation induced root growth and expression of the high-affinity P transporter PT6 was highest in the irrigated variety IR66 which a]so maintained grain yield under P-deficient field conditions.

Pathohistological classification systems in gastric cancer:Diagnostic relevance and prognostic value

Several pathohistological classification systems exist for the diagnosis of gastric cancer. Many studies have investigated the correlation between the pathohistological characteristics in gastric cancer and patient characteristics, disease specific criteria and overall outcome. It is still controversial as to which classification system imparts the most reliable information, and therefore, the choice of system may vary in clinical routine. In addition to the most common classification systems, such as the Laurén and the World Health Organization (WHO) classifications, other authors have tried to characterize and classify gastric cancer based on the microscopic morphology and in reference to the clinical outcome of the patients. In more than 50 years of systematic classification of the pathohistological characteristics of gastric cancer, there is no sole classification system that is consistently used worldwide in diagnostics and research. However, several national guidelines for the treatment of gastric cancer refer to the Laurén or the WHO classifications regarding therapeutic decision-making, which underlines the importance of a reliable classification system for gastric cancer. The latest results from gastric cancer studies indicate that it might be useful to integrate DNA- and RNA-based features of gastric cancer into the classification systems to establish prognostic relevance. This article reviews the diagnostic relevance and the prognostic value of different pathohistological classification systems in gastric cancer.
