High resolution melting real-time PCR detect and identify filarial parasites in domestic cats

Objective: To detect and identify filarial parasites in dried blood spots(DBS) collected from domestic cats using high resolution melting real-time PCR(HRM RT-PCR). Methods: A total of 208 DBS were collected from domestic cats in a brugian filariasis endemic areas in Surat Thani Province, southern Thailand. Microfilariae were found in 9 blood slides using Giemsa-stained thick blood film. The extracted DNA from blood spot volumes of 10 and 20 μL DBS with positive filarial parasites in cats were performed using HRM RT-PCR method. The primers were designed based on the partial mitochondrial 12S rRNA gene for identifying Brugia malayi, Brugia pahangi, Dirofilaria immitis. All purified samples were then detected. Results: Using different volumes of 10 μL and 20 μL DBS could easily distinguish filarial parasites and showed similar results. PCR amplicons of Brugia malayi, Brugia pahangi and Dirofilaria immitis were determined at melting peak(temperature) of 75.70℃, 77.46 ℃, and 73.56 ℃, respectively. All 9 positive DBS samples showed positive Brugia pahangi and similar nucleotide sequences. Conclusions: This HRM RT-PCR method is able to diagnose, identify and discriminate filarial parasites collected from DBS, which is simple and inexpensive compared with other probe-based genotyping methods. Furthermore, this method is useful to survey, prevent and control filariasis.

Background Although freshwater ecosystems cover less than 1% of the earth's surface, they support extremely high levels of biodiversity and provide vital ecosystem services. However, due to the introduction of non-native fishes, aquatic ecosystem functioning has been altered, and in some cases, declined sharply. Quantifying the impacts of invasive species has proven problematic. In this study, we examined the relative trophic position of native piscivorous fishes to estimate the effects of invasive Nile tilapia on food webs in the downstream sections of an invaded large subtropical river, the Pearl River, China. Furthermore, we quantified how native piscivorous fish diets changed as the Nile tilapia invasion *** The trophic position of the widely distributed and locally important economically harvested piscivorous culter fish(Culter recurviceps), mandarinfish(Siniperca kneri), and catfish(Pelteobagrus fulvidraco) lowered significantly in the invaded Dongjiang River compared to an uninvaded reference Beijiang River. The lower trophic position of these piscivorous fishes was reflected by a major reduction in the proportion of prey fish biomass in their diets following the Nile tilapia invasion. Small fishes in the diet of culter fish from the reference river(33% small fishes, 17% zooplankton) shifted to lower trophic level zooplankton prey in the invaded river(36% zooplankton, 25% small fish), possibly due to the presence of Nile tilapia. Additionally, small fishes in the diet of mandarinfish in the reference river(46% small fishes, 11% aquatic insects) declined in the invaded river(20% aquatic insects, 30% small fishes). Similarly, the diet of catfish from the reference river shifted from fish eggs(25% fish eggs, 25% aquatic insects) to aquatic insects in the invaded river(44% aquatic insects, 5% fish eggs).Conclusions The results of this study contributed to a growing body of evidence, suggesting that Nile tilapia can modify trophic interactions in in

This work focuses on the fabrication and characterization of Chemical Field-Effect Transistor(ChemFET)gas nanosensor arrays based on single nanowire(SNW).The fabrication processes include micro and nanofabrication techniques enabled by a combination of ultraviolet(UV)and e-beam lithography to build the ChemFET *** show the integration and connection of SNWs across the multiple pairs of nanoelectrodes in the ChemFET by dielectrophoresis process(DEP)thanks to the incorporation of alignment windows(200-300 nm)adapted to the diameter of the *** of the SNW ChemFET array's output and transfer characteristics prove the influence of gate bias on the drain current *** upon hydrogen(H_(2))and nitrogen dioxide(NO_(2))as analyte models of reducing and oxidizing gases show the ChemFET sensing ***,results demonstrate better response characteristics to H_(2)when the ChemFET operates in the subthreshold *** design concepts and methods proposed for fabricating the SNW-based ChemFET arrays are versatile,reproducible,and most likely adaptable to other systems where SNW arrays are required.

目的本研究旨在研制并验证针对转移性乳腺癌(Metastatic Breast Cancer,MBC)患者的快速心理社会健康筛查工具(MBC-PsySoc-Well-being)。方法采用混合研究方法,分两个阶段进行。第1阶段,采用焦点小组访谈法,与患者、护理人员和医疗专业人员分别进行小组讨论,制订筛查工具的条目内容。第2阶段,进行信效度检验。由5名专家审查工具的内容效度。在53例MBC患者中进行信效度检验,包括结构效度、效标关联效度、内部一致性系数和重测信度检验。结果根据焦点小组访谈结果,对6个主题进行分析,基于这些主题制订了8个条目。专家评判条目目标一致性指数为0.6~1.0。探索性因子分析提取出3个因子:好奇并主动寻求信息,热切希望回归正常生活,以及调整为积极的生活方式。该工具与欧洲癌症研究与治疗组织研制的生活质量核心调查问卷得分呈正相关(r=0.404,P=0.003),Cronbach?蒺sα系数为0.686,相隔2周测量的条目得分相关系数为0.410~0.673。结论本研究期望引起对MBC患者心理社会健康快速筛查工具研发的重视。仍需进一步进行更大样本量的深入研究,以确定该评估工具的信效度。

Effect of storage conditions on long-term stability of Ag nanoparticles formed via green synthesis

Spherical Ag nanoparticles(AgNPs) with a diameter of 20 nm or smaller were biologically synthesized using algae Parachlorella kessleri. The effect of storage conditions on the long-term stability of AgNPs was investigated. UV/Vis spectrophotometry, transmission electron microscopy, and dynamic light scattering measurements revealed that the long-term stability of AgNPs was influenced by light and temperature conditions. The most significant loss of stability was observed for the AgNPs stored in daylight at room temperature. The AgNPs stored under these conditions began to lose their stability after approximately 30 d; after 100 d, a substantial amount of agglomerated particles settled to the bottom of the Erlenmeyer flask. The AgNPs stored in the dark at room temperature exhibited better long-term stability. Weak particle agglomeration began at approximately the 100 th day. The AgNPs stored in the dark at about 5℃ exhibited the best long-term stability; the AgNPs stored under such conditions remained spherical, with a narrow size distribution, and stable(no agglomeration) even after 6 months. Zeta-potential measurements confirmed better dispersity and stability of AgNPs stored under these conditions.

公共组织中多次出现影响巨大的操作风险管理失败案例,让人们对公共组织理论中的操作风险管理产生了质疑。这些重复的操作风险失败案例源自于人为操作,属于内部流程和系统,以及外部事件引起的范畴。针对这一问题,本论文分析了创新能力、领导包容行为、感知到的工作团队多样性以及工作控制点因素之间的关系,同时考察这些因素对操作风险管理的影响。本论文采用横断式调查法,对中国大陆公共组织人员进行数据抽样调查。本文的理论假设已通过459个有效案例得到验证。本文发现,创新能力对操作风险管理有正面的影响。领导包容行为和感知到的工作团队多样性的介入进一步加强了操作风险管理。此外,工作控制点对领导包容行为、感知到的工作团队多样性和操作风险管理之间的关系具有正面影响。类似于之前的研究,本论文同样支持公共组织理论通过战略上定位创新型员工在保持高度包容的工作环境里所带来的操作风险管理的预期结果,从而进一步避免创新风险的能力。本研究主要关注在创新能力对操作风险管理的直接影响。本文展示了领导包容行为、感知到的工作团队多样性以及工作控制点在加强对公共组织操作风险管理的创新能力所起到的积极作用。

Snags are an important component of beech forests that promote ***,their occurrence is completely marginal in managed *** snags in these stands would greatly enhance *** investigated whether snag dimensions were important for saproxylic beetle richness since they were easily transferable parameters to forest management and assessed the presence of other snag microhabitats affecting beetle *** collection was performed using passive flight traps placed on thirty snags in a recent beech reserve.A total of 6706 adults belonging to 231 saproxylic species(53 Red List species,23%)were *** results showed that the most important snag parameters were the diameter(thickness)and canopy openness of the surrounding *** occurrence of Fomes fomentarius,the volume of snag and decay class 3 were marginally significant in terms of the preference of all saproxylic *** diversity was reduced by an advanced degree of decay and a surprisingly deep stem *** dividing snag thickness into categories(70 cm DBH),we found that categories with snag diameter greater than 35 cm showed little differences in all saproxylic and Red List species richness and diversity indices and exhibited the highest similarity in beetle *** recommendations to forest managers in terms of optimization and simplification of practical procedures,we suggest actively creating high stumps to act as snags greater than 35 cm in DBH diameter to promote biodiversity in beech management stands.
