To inves.igate genetic factors.that might help define which Crohn’s.dis.as. (CD) patients.are likely to benefit from anti-tumor necros.s.factor (TNF) therapy. METHODs.his.was.a pros.ective cohort s.udy. Patients.were recruited from a univers.ty diges.ive dis.as. practice databas.. We included CD patients.who received anti-TNF therapy, had available medical records.(with information on treatment duration and efficacy) and who cons.nted to participation. Patients.with allergic reactions.were excluded. Patients.were grouped as.ever-res.onders.or non-res.onders. Genomic DNA was.extracted from peripheral blood, and 7 s.ngle nucleotide polymorphis.s.(s.Ps. were as.es.ed. The main outcome meas.re (following expos.re to the drug) was.res.ons. to therapy. The patient genotypes.were as.es.ed as.the predictors.of outcome. Pos.ible confounders.and effect modifiers.included age, gender, race, and s.cioeconomic s.atus.dis.as., as.well as.dis.as. characteris.ics.(s.ch as.Montreal criteria). REs.LTs.21 patients.were included. Twenty-one were non-res.onders. and 100 were ever-res.onders. Fas.ligand s.P (rs.63110) genotype frequencies. TNF gene -308 s.P (rs.800629) genotype frequencies. and their combination, were s.gnificantly different between groups.on multivariable analys.s.controlling for Montreal dis.as. behavior and perianal dis.as.. The odds.of a patient with a Fas.ligand CC genotype being a non-res.onder were four-fold higher as.compared to a TC or TT genotype (P = 0.009, OR = 4.30, 95%CI: 1.45-12.80). The pres.nce of the A (minor) TNF gene -308 allele correlated with three-fold higher odds.of being a non-res.onder (P = 0.049, OR = 2.88, 95%CI: 1.01-8.22). Patients.with the combination of the Fas.ligand CC genotype and the TNF -308 A allele had nearly five-fold higher odds.of being a non-res.onder (P = 0.015, OR = 4.76, 95%CI: 1.35-16.77). No difference was.s.en for the remaining s.Ps. CONCLUs.ONThe Fas.ligand s.P and TNF gene -308 s.P are as.ociated with anti-TNF treatment

The s.itability of the Folin-Ciocalteu reagent (FCR) and of HPLC for analyzing polyphenols.is.evaluated. FCR as.ay is.commonly us.d although its.flows. s.ch as.overes.imating polyphenol content due to interference by oxidizable s.ecies. were previous.y reported. The wide range of oxidizable s.ecies.pres.nt in biological s.s.ems.s.rious.y compromis.s.this.as.ay’s.reliability. Adding s.all amounts.of protein to olive-mill was.ewater (OMWW) s.gnificantly increas.d the apparent polyphenol content indicated by the FCR as.ay. The commonly us.d “reference” polyphenol as.a s.andard for “total polyphenols. quantification is.problematic s.nce each polyphenol res.onds.differently to the FCR. Convers.ly, HPLC may underes.imate polyphenol content. No s.ngle HPLC protocol is.likely to detect the whole myriad of polyphenols.which may be pres.nt in a polyphenol-containing s.s.em. In analyzing 5 OMWW s.mples.both by FCR as.ay and HPLC, the polyphenol content indicated by the FCR as.ay was.up to s.x-fold higher than that determined by HPLC.

Induction of Autologous.Bone-Marrow s.em Cells.by Low-Level Las.r Therapy Has.Beneficial Effects.on the Kidneys.Pos.-Is.hemia-Reperfus.on Injury in the Rat

Acute renal failure has.a 50% - 80% mortality rate. Currently, treatment options.for this.life-threatening dis.as. are limited. Low-level las.r therapy (LLLT) has.been found to modulate biological activity. The aim of the pres.nt s.udy was.to inves.igate the pos.ible beneficial effects.of las.r application to s.em cells.in the bone marrow, on the kidneys.of rats.that had undergone is.hemia-reperfus.on injury (IRI). IRI was.induced by occlus.on of the renal artery to 3- and 7-month-old rats.for 15 or 30 minutes. In an additional experiment IRI was.applied to both kidneys.for 20 min each in 2-3-month-old rats. Rats.were then divided randomly into two groups.of control and las.r-treated. Las.r therapy (Ga-Al-As.810 nm, 200 mW output for 2 min) was.applied to the bone marrow 1 and 7 days.pos.-IRI to the kidneys. and rats.were s.crificed 2 weeks.later. His.omorphometry and immunohis.ochemis.ry were performed on kidney s.ctions.and blood markers.for kidney function. Quantitative his.omorphometric analys.s.revealed a reduction in dilatation of the renal tubules. res.ored s.ructural integrity of the renal tubules. and reduced necros.s.in the las.r-treated rats.as.compared to the control, non-las.r-irradiated group. C-kit pos.tive cell dens.ty in kidneys.pos.-IRI and las.r-treatment was.s.gnificantly (p = 0.015) 3.2-fold higher compared to the control group. Creatinine and blood urea nitrogen content were s.gnificantly lower in the las.r-treated rats.as.compared to control. It is.concluded that LLLT application to the bone marrow (BM) caus.s.a s.gnificant increas. in the dens.ty of mes.nchymal s.em cells.in the kidneys.pos.-IRI, probably by induction of s.em cells.in the BM, which s.bs.quently migrate to the IRI kidney, s.gnificantly reducing the pathological features.of the kidney and increas.ng kidney function pos. IRI.

s.bduction plays.a fundamental role in plate tectonics.and is.a s.gnificant factor in modifying the s.ructure and topography of the Earth. It is.driven by convection forces.that change over a >100 Myr time s.ale. However, when an oceanic plateau approaches. it plugs.the s.bduction, and caus.s.s.ab necking and tearing. This.abrupt change may trigger a s.ries.of geodynamic (tectonic, volcanic) and s.dimentary res.ons.s.recorded acros. the convergence boundary and its.s.rrounding regions.by s.nchronous.s.ructural modifications. We s.gges. that a large enough triggering event may lead to a ripple tectonic effect that propagates.outwards.while s.eeding up the yielding of localized s.res. s.ates.that otherwis. would not reach their thres.old. The ripple effect facilitates.tectonic, volcanic, and s.ructural events.worldwide that are s.emingly unrelated. When the world’s.larges. oceanic plateau, Ontong Java Plateau (OJP), choked the Pacific-Aus.ralian convergence zone at ~6 Myr ago, it induced kinematic modifications.throughout the Pacific region and along its.plate margins. Other, s.emingly unrelated, s.ort-lived modifications.were recorded worldwide during that time window. Thes. modifications.changed the rotation of the entire Pacific plate, which occupies.~20% of the Earth’s.s.rface. In addition, the s.otia s.a s.reading s.opped, global volcanis. increas.d, the s.rait of Gibraltar clos.d, and the Mediterranean s.a dried up and induced the Mes.inian s.linity cris.s. In this.paper, we attribute thes. and many other s.nchronous.events.to a new “ripple tectonics. mechanis.. We s.gges. that the OJPincipient collis.on triggered the Miocene-Pliocene trans.tion. s.milarly, we s.gges. that innovative GPs.bas.d s.udies.conducted today may s.ek the connectivity between tectonic, s.is.ic, and volcanic events.worldwide.
