Skeletal muscle plays a significant role in both local and systemic energy metabolism. The current investigation aims to explore the role of the Bambi gene in skeletal muscle, focusing on its implications for muscle hypertrophy and systemic metabolism. We hypothesize that skeletal muscle-specific deletion of Bambi induces muscle hypertrophy, improves metabolic performance, and activates thermogenic adipocytes via the reprogramming of progenitor of i WAT, offering potential therapeutic strategies for metabolic syndromes. Leveraging the chromatin immunoprecipitation(chIP)-seq and bioinformatics analysis, Bambi gene is shown to be a direct target of HIF2α, which is further confirmed by chIP-qPcR and promoter luciferase assay. Skeletal muscle-specific Bambi deletion led to significant muscle hypertrophy and improved metabolic parameters, even under high-fat diet conditions. This deletion induced metabolic reprogramming of stromal vascular fractions(SVFs) into thermogenic adipocytes, contributing to systemic metabolic improvements, potentially through the secretory ***, mice with skeletal muscle-specific Bambi deletion demonstrate resistance to high-fat diet-induced metabolic disorders, highlighting a potential therapeutic pathway for metabolic syndrome management. Thus, skeletal muscle-specific deletion of Bambi triggers muscle growth, enhances metabolic performance, and activates thermogenic adipocytes. These findings suggest Bambi as a novel therapeutic target for metabolic syndromes, providing new insights into the interaction between muscle hypertrophy and systemic metabolic improvement. The study underscores the potential of manipulating muscle physiology to regulate whole-body metabolism, offering a novel perspective on treating metabolic disorders.

Genotypic diversity of a dominant c_(4) grass across a long-term fire frequency gradient

Aims and Methods Diversity-disturbance research has focused on community diversity,but disturbance frequency could impact diversity within species as well,with important consequences for community diversity and ecosystem *** examined patterns of genetic diversity of a dominant grass species,Andropogon gerardii,in native North American tallgrass prairie sites located in eastern Kansas that have been subjected to a gradient of fire frequency treatments(burned every 1,2,4 or 20 years)since the *** addition,we were able to assess the relationships between genetic diversity of ***,species diversity and productivity across this range of fire *** Findings We found no significant relationships between genetic diversity of *** at the local scale(1 m^(2) plot level)and disturbance frequency(burned 2 to 32 times over a 38-year period).However,at the site level(*** all plots sampled within a site,~100 m^(2))there were differences in genotype richness and composition,as well as genomic dissimilarity among individuals of *** richness was greatest for the site burned at an intermediate(4-year)frequency and lowest for the infrequently(20-year)burned *** addition,genotypes found in the frequently burned sites were more similar from each other than expected by random chance than those found in the infrequently burned *** composition of *** was not significantly different between the frequently burned sites(annual vs.2 year)but did differ between frequently burned and infrequently burned sites(1 and 2 year vs.4 and 20 year,etc.).Together,these results suggest site-level ecological sorting of genotypes in intact prairie across a broad gradient of disturbance frequencies,likely driven by alterations in environmental *** fire promotes the abundance of dominant grass species,reduces plant community diversity and impacts ecosystem processes such as *** study suggests that

Detection of Breaking Waves Using X-Band Pulse Radar in the Nearshore Region

An X-band pulsed Doppler microwave radar has been used to determine the characteristics of breaking waves. Field experiments were conducted at the Shuang-Si estuary in the north of Taiwan in the winter of 2005. Analyses on maxima radar cross section and Doppler frequency shift are done to characterize wave breaking zones. Based on observations of breaking waves, the wave breaking zones are shown to be located at water depths of 1.8 to 2.2 m in the experimental site. In general, the results indicate that a radar system has the potential to delineate the spatial variation of breaking waves clearly and that this is sufficient to achieve a measurement operation for near-shore air-sea interaction events.

Oxidation Protective c/Sic/Si-Sic Multilayer coating for carbon/carbon composites Applying at 1873 K

A c/Sic/Si-Sic multilayer coating for protecting carbon/carbon （c/c） composites against oxidation was prepared by slurry and pack cementation. X-ray diffraction （XRD） and energy dispersive spectroscopy （EDS） analysis showed that the inner coating obtained from the slurry and pack cementation was a c/Sic gradient layer acting as bonding layer, and the exterior coating formed in the second pack cementation was a Si-Sic double phase coating. Oxidation tests at 1873 K in air showed that the coating exhibited excellent oxidation protective ability and thermal shock resistance. The coating could effectively protect c/c composites from oxidation at 1873 K in air for 170 h and undergo the thermal cycling between 1873 K and room temperature for 8 cycles. The weight loss of the coated c/c composites is due to the formation of cracks and holes in the coating and the volatilization of SiO2 glass at high temperature.

Identification and characterization of Heparan Sulphate Binding Proteins of <i>Entamoeba histolytica</i>

A large number of microbial pathogens bind to heparan sulphate on eukaryotic cell surfaces. Heparan Sulphate Binding Proteins (HSBPs) from Entamoeba histolytica culture lysates were obtained by sequential ammonium sulphate precipitation and Protein purify. SDS-PAGE and immunoblotting experiments indicated the presence of two major extracellular proteins in E. histolytica (51.2 kDa and 61.0 kDa). characterization of HSBPs by 2D Gel electrophoresis of 40% (NH4)2SO4 precipitated lysate of E. histolytica revealed that the isoelectric point of 51.2 kDa HSBP was at pH3.0. The protein of 61.0 kDa HSBP showed three spots in 40% (NH4)2SO4 lysate which had isoelectric point between pH 4.0 - 7.0. While in 80% (NH4)2SO4 precipitated lysate, 51.2 kDa HSBP showed only one spot which had isoelectric point at pH 3. However, 61.0 kDa HSBP revealed 2 spots which had isoelectric point between pH 4 and 5. The result showed that this parasite has proteins which interact with heparan sulphate whose molecular formula is c14H23NO21S-23. These proteins may have a role in binding of parasite to heparan sulphate on host cells. Further characterization by MALDI-TOF analysis of HSBPs from E. histolytica demonstrated HSBPs to be novel protein in this parasite which has been isolated, purified and characterized first time by our group in the present study.

New method for rekindling the explosive waves in Maxwellian space plasmas

Our interest is to study the nonlinear wave phenomena in complex plasma constituents with Maxwellian electrons and ions. The main aim is to use a new method known as the(G′/G)method to exhibit the effects of dust charge fluctuations on the evolution of nonlinear waves. The coherent features of the shock and solitary waves along with the generation of high-energy waves have been amplified through the solution of the Korteweg–de Vries–Burgers equation,and the different natures of the waves were found successfully. Results are discussed graphically with the thoughtful choice of typical plasma parameters from different space plasma environments, exactly those found in cosmic dusty plasmas laden in ionospheric auroral region,radial spokes of Saturn's rings, planetary nebulae and solar F-corona region. All conclusions are in good accordance with the actual occurrences and could be of interest to further investigations of space. Moreover, the applicability of the present method is hoped to be a great enhancement by its use as ingenious mechanism used to evaluate the soliton dynamics and Burgers shock waves.

Pushover analysis of reinforced concrete frames considering shear failure at beam-column joints

Since most current seismic capacity evaluations of reinforced concrete （Rc） frame structures are implemented by either static pushover analysis （PA） or dynamic time history analysis, with diverse settings of the plastic hinges （PHs） on such main structural components as columns, beams and walls, the complex behavior of shear failure at beam-column joints （BcJs） during major earthquakes is commonly neglected. This study proposes new nonlinear PA procedures that consider shear failure at BcJs and seek to assess the actual damage to Rc structures. Based on the specifications of FEMA-356, a simplified joint model composed of two nonlinear cross struts placed diagonally over the location of the plastic hinge is established, allowing a sophisticated PA to be performed. To verify the validity of this method, the analytical results for the capacity curves and the failure mechanism derived from three different full-size Rc frames are compared with the experimental measurements. By considering shear failure at BcJs, the proposed nonlinear analytical procedures can be used to estimate the structural behavior of Rc frames, including seismic capacity and the progressive failure sequence of joints, in a precise and effective manner.

Roles of soil-structure interaction and damping in base-isolated structures built on numerous soil layers overlying a half-space

This study examines the roles of soil-structure interaction （SSI）, higher modes, and damping in a base-isolated structure built on multiple layers of soil overlying a half space. closed-form solutions for the entire system, including a superstructure, seismic isolator, and numerous soil layers overlying a half-space, were obtained. The formulations obtained in this study simply in terms of well-known frequencies and mechanical impedance ratios can explicitly interpret the dynamic behavior of a base-isolated structure interacting with multiple soil layers overlying a half-space. The key factors influencing the performance of the isolation system are the damping ratio of the isolator and the ratio of the natural frequency of the fixed-base structure to that of the isolated structure by assuming that the superstructure moves as a rigid body. This study reveals that higher damping in the base isolator is unfavorable to higher mode responses that usually dominate the responses of the superstructure and that the damping mechanism plays an important role in transmitting energy in addition to absorbing energy. It is also concluded that it is possible to design a soft soil layer as an isolation system for isolating vibration energy.

Equivalent series system to model a multiple friction pendulum system with numerous sliding interfaces for seismic analyses

current structural analysis software programs offer few if any applicable device-specifi c hysteresis rules or nonlinear elements to simulate the precise mechanical behavior of a multiple friction pendulum system(MFPS) with numerous sliding *** on the concept of subsystems,an equivalent series system that adopts existing nonlinear elements with parameters systematically calculated and mathematically proven through rigorous derivations is *** aim is to simulate the characteristics of sliding motions for an MFPS isolation system with numerous concave sliding interfaces without prior knowledge of detailed information on the mobilized forces at various sliding *** MFPS with numerous concave sliding interfaces and one articulated or rigid slider located between these interfaces is divided into two subsystems: the fi rst represents the concave sliding interfaces above the slider,and the second represents those below the *** equivalent series system for the entire system is then obtained by connecting those for each subsystem in *** equivalent series system is validated by comparing numerical results for an MFPS with four sliding interfaces obtained from the proposed method with those from a previous study by Fenz and ***,these numerical results demonstrate that an MFPS isolator with numerous concave sliding interfaces,which may have any number of sliding interfaces,is a good isolation device to protect structures from earthquake damage through appropriate designs with controllable mechanisms.
