Mesenchymal stem cell-derived apoptotic bodies alleviate alveolar bone destruction by regulating osteoclast differentiation and function

Periodontitis is caused by overactive osteoclast activity that results in the loss of periodontal supporting tissue and mesenchymal stem cells(MSCs)are essential for periodontal ***,the hypoxic periodontal microenvironment during periodontitis induces the apoptosis of *** bodies(ABs)are the major product of apoptotic cells and have been attracting increased attention as potential mediators for periodontitis treatment,thus we investigated the effects of ABs derived from MSCs on *** were derived from bone marrows of mice and were cultured under hypoxic conditions for 72 h,after which ABs were isolated from the culture supernatant using a multi-filtration *** results demonstrate that ABs derived from MSCs inhibited osteoclast differentiation and alveolar bone *** array analysis showed that miR-223-3p is highly enriched in those ABs and is critical for their therapeutic *** and luciferase activity results confirmed that Itgb1 is targeted by miR-223-3p,which interferes with the function of ***,DC-STAMP is a key regulator that mediates membrane *** and pre-osteoclasts expressed high levels of DC-STAMP on their membranes,which mediates the engulfment of ABs by *** with knock-down of DC-STAMP failed to be engulfed by ***,MSC-derived ABs are targeted to be engulfed by pre-osteoclasts via DC-STAMP,which rescued alveolar bone loss by transferring miR-223-3p to osteoclasts,which in turn led to the attenuation of their differentiation and bone *** results suggest that MSC-derived ABs are promising therapeutic agents for the treatment of periodontitis.

Cosmological test of dark energy parametrizations within the framework of Horava-Lifshitz gravity via baryon acoustic oscillation

We conduct an investigation to explore late-time cosmic acceleration through various dark energy parametrizations(Wettrich,Efstathiou,and Ma-Zhang)within the Horava-Lifshitz gravity *** an alternative to general relativity,this theory introduces anisotropic scaling at ultraviolet *** primary objective is to constrain the key cosmic parameters and baryon acoustic oscillation(BAO)scale,specifically the sound horizon(rd),by utilizing 24 uncorrelated measurements of BAOs derived from recent galaxy surveys spanning a redshift range from z=0.106 to z=***,we integrate the most recent Hubble constant measurement by Riess in 2022(denoted as R22)as an extra *** the parametrizations of Wettrich,Efstathiou,and Ma-Zhang,our analysis of BAO data yields sound horizon results of r_(d)=148.1560±2.7688 Mpc,r_(d)=148.6168±10.2469 Mpc,and r_(d)=147.9737±10.6096 Mpc,*** the R22 prior into the BAO dataset results in r_(d)=139.5806±3.8522 Mpc,r_(d)=139.728025±2.7858 Mpc,and r_(d)=139.6001±2.7441 *** outcomes highlight a distinct inconsistency between early and late observational measurements,analogous to the H_(0) tension.A notable observation is that,when we do not include the R22 prior,the outcomes for rd tend to be in agreement with Planck and SDSS *** this,we conducted a cosmography test and comparative study of each parametrization within the Lambda Cold Dark Matter *** diagnostic analyses demonstrate that all models fit seamlessly within the phantom *** dark energy parametrizations predict an equation of state parameter close ω=-1,indicating a behavior similar to that of a cosmological *** statistical analysis indicates that neither of the two models can be ruled out based on the latest observational measurements.

SRG1,Encoding a Kinesin-4 Protein,Is an Important Factor for Determining Grain Shape in Rice

Grain shape is one of the important agronomic traits, which is closely related to the yield of rice.A new rice mutant, named small and round grain(srg1), was isolated from an indica cultivar Zhenong 34 by ethyl methane sulfonate(EMS) mutagenesis. The microscopic analysis showed that the cell length of spikelet in srg1 was decreased compared with that in wild type(WT), which caused the grain length ***, the grains of srg1 were wider than those of WT because of the increased cell layers in spikelet in the lateral direction. Therefore, the inhibition of cell expansion and increased cell proliferation collectively led to the small and round grain. By map-based cloning, the gene SRG1 was located on the short arm of chromosome 9, which encodes a kinesin-4 protein, represented by the gene LOC_Os09 g02650. A single nucleotide polymorphism, occurred in the 16 th exon of SRG1, led to premature translation stop in mutant. The cell cycle-related genes were up-regulated in srg1, which conferred that SRG1 controlled grain width through the cell proliferation. Since the role of SRG1 in regulating grain shape was not clarified well before, it is valuable to explore the mechanism of grain development. This study could, hence, provide a morphogenesis and molecular basis for elucidating the function of SRG1, as well as a new germplasm resource for the further study of grain development.

Osteoclasts are bone-specific multinucleated cells generated by the differentiation of monocyte/macrophage lineage precursors. Regulation of osteoclast differentiation is considered an effective therapeutic approach to the treatment of bone-lytic diseases. Periodontitis is an inflammatory disease characterized by extensive bone resorption. In this study, we investigated the effects of sodium fluoride （NaF） on osteoclastogenesis induced by Porphyromonas gingivalis, an important colonizer of the oral cavity that has been implicated in periodontitis. NaF strongly inhibited the P. gingivalis-induced alveolar bone loss. That effect was accompanied by decreased levels of cathepsin K, interleukin （IL）-1β, matrix metalloproteinase 9 （MMP9）, and tartrate-resistant acid phosphatase, which were up-regulated during P. gingivalis-induced osteoclastogenesis. Consistent with the in vivo anti-osteoclastogenic effect, NaF inhibited osteoclast formation caused by the differentiation factor RANKL （receptor activator of nuclear factor KB ligand） and macrophage colony-stimulating factor （M-CSF）. The RANKL-stimulated induction of the transcription factor nuclear factor of activated T cells （NFAT） cl was also abrogated by NaF. Taken together, our data demonstrate that NaF inhibits RANKL-induced osteoclastogenesis by reducing the induction of NFATcl, ultimately leading to the suppressed expression of cathepsin K and MMP9. The in vivo effect of NaF on the inhibition of P. gingivalis-induced osteoclastogenesis strengthens the potential usefulness of NaF for treating periodontal diseases.

An Adaptive Time-Step Backward Differentiation Algorithm to Solve Stiff Ordinary Differential Equations: Application to Solve Activated Sludge Models

A backward differentiation formula (BDF) has been shown to be an effective way to solve a system of ordinary differential equations (ODEs) that have some degree of stiffness. However, sometimes, due to high-frequency variations in the external time series of boundary conditions, a small time-step is required to solve the ODE system throughout the entire simulation period, which can lead to a high computational cost, slower response, and need for more memory resources. One possible strategy to overcome this problem is to dynamically adjust the time-step with respect to the system’s stiffness. Therefore, small time-steps can be applied when needed, and larger time-steps can be used when allowable. This paper presents a new algorithm for adjusting the dynamic time-step based on a BDF discretization method. The parameters used to dynamically adjust the size of the time-step can be optimally specified to result in a minimum computation time and reasonable accuracy for a particular case of ODEs. The proposed algorithm was applied to solve the system of ODEs obtained from an activated sludge model (ASM) for biological wastewater treatment processes. The algorithm was tested for various solver parameters, and the optimum set of three adjustable parameters that represented minimum computation time was identified. In addition, the accuracy of the algorithm was evaluated for various sets of solver parameters.

Identification, occurrence, and validation of DRE and ABRE Cis-regulatory motifs in the promoter regions of genes of Arabidopsis thaliana

Plants posses a complex co-regulatory network which helps them to elicit a response under diverse adverse conditions. We used an in silico approach to identify the genes with both DRE and ABRE motifs in their promoter regions in Arabidopsis thaliana. Our results showed that Arabidopsis contains a set of 2,052 genes with ABRE and DRE motifs in their promoter regions. Approximately 72~o or more of the total predicted 2,052 genes had a gap distance of less than 40o bp between DRE and ABRE motifs. For positional orientation of the DRE and ABRE motifs, we found that the DR form （one in direct and the other one in reverse orientation） was more prevalent than other forms. These predicted 2,o52 genes include 155 transcription factors. Using microarray data from The Arabidopsis Information Resource （TAIR） database, we present 44 transcription factors out of 155 which are upregulated by more than twofold in response to osmotic stress and ABA treatment. Fifty-one transcripts from the one predicted above were validated using semiquantitative expression analysis to support the microarray data in TAIR. Taken together, we report a set of genes containing both DRE and ABRE motifs in their promoter regions in A. thaliana, which can be useful to understand the role of ABA under osmotic stress condition.
