Correlation between precipitates evolution and mechanical properties of Al-Sc-Zr alloy with Er additions during isothermal ageing were investigated by microhardness measurements,transmission electron microscopy,atom probe tomography and density functional theory-based *** results demonstrate that the Er additions significantly improve the hardness during elevated temperature ageing,especially at 400℃.This is mainly because Er additions increase the nucleation rate of the Al_(3)(Er,Sc,Zr)precipitates,resulting in a higher density of fine and uniform dispersion of L1_(2)structured ***-principles calculations demonstrate that the second nearest neighboring solute-solute interactions for the species Sc,Zr and Er are energetically favored–a key feature to rationalize the observed precipitate structure and the underlying formation *** sequential formation of the core/shell precipitates in the Er-free alloy and core/double-shell precipitates in the Er-containing alloy arises due to the different solute-solute and solute-vacancy interaction energies,and the relative diffusivities of the Er,Sc and Zr species in *** results shed light on the beneficial effects of Er additions on the agehardening behavior of Al-Sc-Zr alloy and provide guidance for designing the ageing treatments for the Al-Sc-Zr(-Er)alloys.

AIM: To study the interleukin-1(IL-1) pathway as a therapeutic target for liver fibrosis in vitro and in vivo using the ATP-binding cassette transporter b4^(-/-)(Abcb4^(-/-)) mouse ***: Female and male Abcb4^(-/-) mice from 6 to 13 mo of age were analysed for the degree of cholestasis(liver serum tests), extent of liver fibrosis(hydroxyproline content and Sirius red staining) and tissue-specific activation of signalling pathways such as the IL-1 pathway [quantitative polymerase chain reaction(q PCR)]. For in vivo experiments, murine hepatic stellate cells(HSCs) were isolated via pronasecollagenase perfusion followed by density gradient centrifugation using female mice. Murine HSCs were stimulated with up to 1 ng/m L IL-1β with or without 2.5 μg/m L Anakinra, an IL-1 receptor antagonist, respectively. The proliferation of murine HSCs was assessed via the Brd u assay. The toxicity of Anakinra was evaluated via the fluorescein diacetate hydrolysis(FDH) assay. In vivo 8-wk-old Abcb4^(-/-) mice with an already fully established hepatic phenotype were treated with Anakinra(1 mg/kg body-weight daily intraperitoneally) or vehicle and liver injury and liver fibrosis were evaluated via serum tests, q PCR, hydroxyproline content and Sirius red staining. RESuLTS: Liver fibrosis was less pronounced in males than in female Abcb4^(-/-) animals as defined by a lower hydroxyproline content(274 ± 64 μg/g vs 436 ± 80 μg/g liver, respectively; n = 13-15; P u-test) and lower m RNA expression of the profibrogenic tissue inhibitor of metalloproteinase-1(TIMP)(1 ± 0.41 vs 0.66 ± 0.33 fold, respectively; n = 13-15; P u-test). SegVeg: Segmenting RGB Images into Green and Senescent Vegetation by Combining Deep and Shallow Methods

Pixel segmentation of high-resolution RGB images into chlorophyll-active or nonactive vegetation classes is a first step often required before estimating key traits of *** have developed the SegVeg approach for semantic segmentation of RGB images into three classes(background,green,and senescent vegetation).This is achieved in two steps:A u-net model is first trained on a very large dataset to separate whole vegetation from *** green and senescent vegetation pixels are then separated using SVM,a shallow machine learning technique,trained over a selection of pixels extracted from *** performances of the SegVeg approach is then compared to a 3-class u-net model trained using weak supervision over RGB images segmented with SegVeg as groundtruth *** show that the SegVeg approach allows to segment accurately the three ***,some confusion is observed mainly between the background and senescent vegetation,particularly over the dark and bright regions of the *** u-net model achieves similar performances,with slight degradation over the green vegetation:the SVM pixel-based approach provides more precise delineation of the green and senescent patches as compared to the convolutional nature of *** use of the components of several color spaces allows to better classify the vegetation pixels into green and ***,the models are used to predict the fraction of three classes over whole images or regularly spaced *** show that green fraction is very well estimated(R^(2)=0.94)by the SegVeg model,while the senescent and background fractions show slightly degraded performances(R^(2)=0.70 and 0.73,respectively)with a mean 95%confidence error interval of 2.7%and 2.1%for the senescent vegetation and background,versus 1%for green *** have made SegVeg publicly available as a ready-to-use script and model,along with the entire annotated grid-pixels *** thus hope to render segmentation acc

There have been significant breakthroughs over the past decade in the development and use of pluripotent stem cells as a potential source of cells for applications in regenerative medicine. It is likely that this methodology will begin to play an important role in human clinical medicine in the years to come. This review describes the plasticity of one type of pluripotent cell, spermatogonial stem cells （SSCs）, and their potential therapeutic applications in regenerative medicine and male infertility. Normally, SSCs give rise to sperm when in the testis. However, both human and murine SSCs can give rise to cells with embryonic stem （ES） cell-like characteristics that can be directed to differentiate into tissues of all three embryonic germ layers when placed in an appropriate inductive microenvironment, which is in contrast to other postnatal stem cells. Previous studies have reported that SSCs expressed an intermediate pluripotent phenotype before differentiating into a specific cell type and that extended culture was necessary for this to occur. However, recent studies from our group using a tissue recombination model demonstrated that SSCs differentiated rapidly into another tissue, in this case, prostatic epithelium, without expression of pluripotent ES cell markers before differentiation. These results suggest that SSCs are capable of directly differentiating into other cell types without going through an intermediate ES cell-like stage. Because SSCs do not require reprogramming to achieve a pluripotent state, they are an attractive source of pluripotent cells for use in regenerative medicine.

Genetic transformation is a useful technique to complement conventional breeding in crop improvement. Although carrot has been a model organism for in vitro embryogenesis study, genetic transformation of carrot is still lengthy and labor intensive. An efficient transformation and detection system is desirable. Direct infection of Agrobacterium to carrot calli has provided an easy way for carrot genetic transformation. To improve the efficiency of antibiotic selection in this method, we report the combined use of an improved green-fluorescent protein, referred to as smGFP, to establish a versatile selection method for carrot callus transformation system. By combining antibiotic selection with the bright fluorescence observed in the callus tissue, we were able to easily identify stable transformants in early stage of the transformation process. In addition to the GFP expression of the callus cells, the transgenic nature of callus cells was confirmed with Southern and Western analysis. We found we can link the simplicity of carrot-callus-cell transformation, early detection of stable transformants with antibiotic selection, visualization of GFP fluorescence, and molecular analysis （Southern and Western） of callus tissue （non-photosynthetic tissue） to provide a more efficient way in identifying stable transformants at early stage of carrot transformation.

This paper presents a novel consensus clustering(CC)approach for a document repository concerning power substations(PSD)and contributes to the intangible asset management of power systems.A domain ontology model,i.e.,substation ontology(SONT),is applied to modify the traditional vector space model(VSM)for document representation,which is concerned with the semantic relationship between terms.A new document representation is generated using a term mutual information matrix with the aid of *** addition,compared with two other novel CC algorithms,i.e.,non-negative matrix factorisation-based CC(NNMF-CC)and information theory-based CC(INT-CC),weighted partition via kernel-based CC algorithm(WPK-CC)is utilised to solve the CC issue for ***,genetic algorithms(GA)were applied to WPK-CC for PSD,as there are limitations in the original WPK-CC for document ***,selected mechanisms in each GA’s procedure are compared and improved,resulting in comprehensive parameter settings for the PSD *** simulation studies have been designed,in which the results are evaluated by purity validation method and show that the SONT-based document representation and improved WPK-CC,via modified GA,significantly improve the performance of the PSD CC.

Magnetocontrollable droplet mobility on surfaces of both solids and simple fluids have been widely used in a wide range of ***,little is understood about the effect of the magnetic field on the wettability and mobility of droplets on structured ***,we report the manipulation of the dynamic behaviors of water droplets on a film of thermotropic liquid crystals(LCs).We find that the static wetting behavior and static friction of water droplets on a 4’-octyl-4-biphenylcarbonitrile(8CB)film strongly depend on the LC mesophases,and that a magnetic field caused no measurable change to these ***,we find that the droplet dynamics can be affected by a magnetic field as it slides on a nematic 8CB film,but not on isotropic 8CB,and is dependent on both the direction and strength of the magnetic *** measuring the dynamic friction of a droplet sliding on a nematic 8CB film,we find that a magnetic field alters the internal orientational ordering of the 8CB which in turn affects its *** support this interpretation with a scaling argument using the LC magnetic coherence length that includes(i)the elastic energy from the long-range orientational ordering of 8CB and(ii)the free energy from the interaction between 8CB and a magnetic ***,these results advance our understanding of droplet mobility on LC films and enable new designs for responsive surfaces that can manipulate the mobility of water droplets.
