感染性心内膜炎很少引起二尖瓣瓣周脓肿,一旦累及瓣周则预后较差。病例报告病史患者,女,68岁,因“发热3个月,一般情况差”治疗1周,疑为心内膜炎。曾静脉使用过青霉素、庆大霉素、速尿并且口服过强心剂。有胰岛素依赖性Ⅱa型糖尿病史8年。

Nuclear mechanics during constricted migration

Cell migration through narrow interstitial space in tissues is commonly seen in both physiological and pathological conditions. For example, immune cells have to squeeze through vasculature and extracellular matrix to reach the wound or diseased sites, and similarly cancer cells need to crawl through porous space in tissues to invade surrounding tumor-free regions. Therefore, the bulky stiff nucleus in the cell becomes a barrier during constricted migration making such process pore-size dependent with smaller pores exponentially alleviate the passage capability. However, in some cases, cells can actively deform their nuclei to squeeze through constrictions although some price needs to be paid. In this review, we focus on:(1) nuclear structures and morphological regulations,(2) proposed mechanisms that drive constricted migration,(3) short-term consequences such as nuclear envelope (NE) rupture and DNA damage during such process,(4) biophysical factors that facilitate NE rupture, and (5) long term consequences such as genomic variation caused by repetitive NE rupture. Both experimental results and modeling are provided with the intention to better understand the constricted migration.

Novel fiber-optical interferometer with miniaturized probe for in-hole measurements

Today, micro-system technology and the development of new MEMS (Micro-Electro-Mechanical Systems) are emerging rapidly. In order for this development to become a success in the long run, measurement systems have to ensure product quality. Most often, MEMS have to be tested by means of functionality or destructive tests. One reason for this is that there are no suitable systems or sensing probes available which can be used for the measurement of quasi inaccessible features like small holes or cavities. We present a measurement system that could be used for these kinds of measurements. The system combines a fiber optical, miniaturized sensing probe with low-coherence interferometry, so that absolute distance measurements with nanometer accuracy are possible.

Examination of a person who has been a victim of a physical or sexual assault may be very important for upcoming legal *** the context of a clinical forensic examination,physical findings are recorded and biological trace material is gathered and ***,all forensic findings are documented in a detailed report combined with photographic docu-mentation,which employs a forensic scale to depict the size of the ***,the integrity of such forensic findings depends particularly on two ***,the examination needs to be conducted professionally to ensure that the findings are properly admissible as court ***,the examination should take place as soon as possible because the opportunity to successfully secure biological samples declines rapidly with *** to low-threshold clinical forensic examinations is not evenly provided in all member states of the European union(Eu);in some states,they are not available at *** part of the JuST_(e)u!(Juridical standards for clinical forensic examinations of victims of violence in Europe)pro-ject,the Ludwig Boltzmann Institute for Clinical Forensic Imaging in Graz,Austria created(in cooperation with its international partner consortium)a questionnaire:the purpose was to collect information about support for victims of physical and/or sexual assault in obtaining a low-threshold clinical forensic examination in various countries of the *** paper pro-vides a summary of the responses and an overview of the current situation concerning pro-vided clinical forensic services.

Acremonium species are prolific producers of therapeutic molecules which include the widely used beta-lactam antibiotic,*** light of their significant medical value,an efficient gene disruption method is required for the physiological and biochemical studies on this genus of ***,the number of selection markers that can be used for gene targeting is limited,which constrain the genetic analysis of multiple functional *** this study,we established a uridine auxotrophy based marker recycling system which achieves scarless gene deletion,and allows the use of the same selection marker in successive transformations in a deep seaderived fungus Acremonium *** identified one homologue of Acremonium chrysogenum pyrG(also as a homologous gene of the yeast uRA3)from HDN16-126,designated as pyrG-A1,which can be used as a selection marker on uridine free *** then removed pyrG-A1 from HDN16-126 genome via homologous recombination(HR)on MM medium with 5-fluoroortic acid(5-FOA),a chemical that can be converted into a toxin of 5-flurouracil by pyrG-A1 activity,thus generating the HDN16-126-△pyrG mutant strain which showed auxotrophy for uridine but insensitivity to 5-FOA and enabled the use of exogenous pyrG gene as both positive and negative selection marker to achieve the scarless deletion of target DNA *** further applied this marker recycling system to successfully disrupt two target genes pepL(encodes a putative 2OG-Fe(Ⅱ)dioxygenase)and pepM(encodes a putative aldolase)identified from HDN16-126 genome,which are proposed to be functional genes related to 2-aminoisobutyric acid metabolism in *** work is the first application of uridine auxotrophy based scarless gene deletion method in Acremonium species and shows promising potential in assisting sequential genetic analysis of filamentous fungi.
