User-friendly tools for robust transcriptional activation of endogenous genes are highly demanded in plants. We previously showed that a dCas9-VP64 system consisting of the deactivated CRISPR- associated protein 9 （dCasg） fused with four tandem repeats of the transcriptional activator VP16 0/1=64） could be used for transcriptional activation of endogenous genes in plants. In this study, we developed a second generation of vector systems for enhanced transcriptional activation in plants. We tested multiple strategies for dCasg-based transcriptional activation, and found that simultaneous recruitment of VP64 by dCas9 and a modified guide RNA scaffold gRNA2.0 （designated CRISPR-Act2.0） yielded stronger transcrip- tional activation than the dCas9-VP64 system. Moreover, we developed a multiplex transcription activator- likeeffector activation （mTALE-Act） system for simultaneous activation of up to four genes in plants. Our results suggest that mTALE-Act is even more effective than CRISPR-Act2.0 in most cases tested. In addition, we explored tissue-specific gene activation using positive feedback loops. Interestingly, our study revealed that certain endogenous genes are more amenable than others to transcriptional activation, and tightly regulated genes may cause target gene silencing when perturbed by activation probes. Hence, these new tools could be used to investigate gene regulatory networks and their control mechanisms. Assembly of multiplex CRISPR-Act2.0 and mTALE-Act systems are both based on streamlined and PCR-independent Golden Gate and Gateway cloning strategies, which will facilitate transcriptional activation applications in both dicots and monocots.

The authors propose a new "three-layer" conceptual model for the air-sea exchange of organic gases, which includes a dynamic surface microlayer with photochemical and biological processes. A parameterization of this three-layer model is presented, which was used to calculate the air-sea fluxes of acetone over the Pacific Ocean. The air-sea fluxes of acetone calculated by the three-layer model are in the same direction but possess half the magnitude of the fluxes calculated by the traditional two-layer model in the absence of photochemical and biological processes. However, photochemical and biological processes impacting acetone in the microlayer can greatly vary the calculated fluxes in the three-layer model, even reversing their direction under favorable conditions. Our model may help explain the discrepancies between measured and calculated acetone fluxes in previous studies. More measurements are needed to validate our conceptual model and provide constraints on the model parameters.

Atmospheric deposition of particulate organic nitrogen(ONp) is a significant process in the global nitrogen cycle and may be pivotally important for N-limited ***,past models largely overlo oked the spatial and chemical inhomogeneity of atmospheric ONpand were thus deficient in assessing global *** constructed a comprehensive global model of atmospheric gaseous and particulate organic nitrogen(ON),including the latest knowledge on emissions and secondary *** this model,we simulated global atmospheric ONpabundances consistent with *** estimated global atmospheric ON deposition was 26 Tg N yr-1,predominantly in the form of ONp(23 Tg N yr-1)and mostly from wildfires(37%),oceans(22%) and aqueous productions(17%).Globally,ONpcontributed as much as 40% to 80% of the total N deposition downwind of biomass-burning *** ONpdeposition thus constituted the dominant external N supply to the N-limited boreal forests,tundras and the Arctic Ocean,and its importance maybe amplified in a future warming climate.

This work mainly indicates that information transparency and accounting quality are not highly related, although they are more related in the electronics industry than in other industries. The empirical findings suggest that it may be important to include more accounting-quality related items into the proxies of financial reporting when building an information evaluation system.

A 42-year-old man presented with a two-year history of progressive dysphagia and hoarseness. Esophagogram and endoscopy revealed submucosal mass effect on the upper esophagus. Computed tomography and magnetic resonance imaging revealed an elongated mass in the retrotracheal region of the lower neck with extension to the posterior mediastinum. Partial tumor resection and histopathological evaluation revealed a WHO type B2 thymoma. Adjuvant radiation and chemotherapy were subsequently administered resulting in complete tumor regression. To our knowledge, this is the first report of ectopic retrotracheal thymoma with clinical and imaging manifestations mimicking those for esophageal submucosal tumor.

根据κ-Khler理论,利用2006年中国境内气溶胶浓度的数值模拟结果,计算总体气溶胶活化为云凝结核的数浓度,并评估有机气溶胶对云凝结核数量的贡献。中国境内云凝结核数浓度总体呈东部高西部低的分布,这与东部受到更多人为源排放影响、气溶胶浓度高有关。假设气溶胶各类化学组分彼此外混合,则各季节云凝结核数浓度为0.9×10^3~1.2×10^3 cm^-3,有机气溶胶对云凝结核数量贡献全年平均为30%。假设气溶胶中有机组分彼此内混合并与其他组分外混合,则各季节云凝结核数浓度为0.9×10^3~1.1×10^3 cm^-3,有机气溶胶对云凝结核数量的贡献全年平均28%。尽管有机气溶胶对云凝结核数量的年平均贡献相差不大,但夏季和冬季的贡献有较大差异,原因在于夏季有更多的二次有机气溶胶生成,而冬季更多的是一次有机气溶胶排放。因此,有机气溶胶是中国云凝结核的重要来源,并且有机气溶胶的混合状态对云凝结核数量有很大的影响。
