Background: Current understanding of T cell epitopes in coeliac disease (CD) largely derives from intestinal T cell clones in vitro. T cell clones allow identification of gluten peptides that stimulate T cells but do not quantify their contribution to the overall gluten specific T cell response in individuals with CD when exposed to gluten in vivo. Aims: To determine the contribution of a putative dominant T cell epitope to the overall gliadin T cell response in HLA- DQ2 CD in vivo. Patients: HLA- DQ2+ individuals with CD and healthy controls. Methods: Subjects consumed 20 g of gluten daily for three days. Interferon γ (IFN- γ ) ELISPOT was performed using peripheral blood mononuclear cells (PBMC) to enumerate and characterise peptide and gliadin specific T cells before and after gluten challenge. Results: In 50/59 CD subjects, irrespective of homo- or heterozygosity for HLA- DQ2, IFN- γ ELISPOT responses for an optimal concentration of A- gliadin 57- 73 Q- E65 were between 10 and 1500 per million PBMC, equivalent to a median 51% of the response for a “ near optimal" concentration of deamidated gliadin. Whole deamidated gliadin and gliadin epitope specific T cells induced in peripheral blood expressed an intestinal homing integrin (α 4β 7) and were HLA- DQ2 restricted. Peripheral blood T cells specific for A- gliadin 57- 73 Q- E65 are rare in untreated CD but can be predictably induced two weeks after gluten exclusion. Conclusion: In vivo gluten challenge is a simple safe method that allows relevant T cells to be analysed and quantified in peripheral blood by ELISPOT, and should permit comprehensive high throughput mapping of gluten T cell epitopes in large numbers of individuals with CD.

Several MoS2 catalysts of different structure, prepared by in situ decomposition of ammonium heptamolybdate （AHM） and molybdenum naphthenate （MoNaph）, and by MoS2 exfoliation （TDM）, were characterized by BET, X-ray diffraction （XRD）, Energy Dispersive X-ray （EDX） and transmission electron microscopy （TEM）. The analysis showed that MoS2 structure was dependant upon the preparation procedure. The activity of the catalysts was determined by measuring the hydrodeoxygenation （HDO） of phenol, 4-methylphenol and 4-methoxyphenol using a batch autoclave reactor operated at 2.8 MPa of hydrogen and temperatures ranging from 320-370℃. By comparing the conversion, the reactivity order of the catalysts was： AHM〉TDM-D〉MoNaph〉thermal〉MoS2 powder〉 TDM-W. Also, the effect of reaction temperature on the HDO conversion was explained in terms of equilibrium of reversible reaction kinetics. The main products of the HDO for phenolic compounds were identified by gas chromatography/mass spectrometry （GC/MS）. The results showed that the product distribution and the HDO selectivity were correlated with the reaction temperature. Two parallel reaction routes, direct hydrogenolysis and combined hydrogenation-hydrogenolysis, were confirmed by the analysis of the product distribution. High temperature favored hydrogenolysis over hydrogenation for HDO of phenol and 4-methoxyphenol, whereas for 4-methylphenol the reverse was true.

An electrochemically pretreated silver macroporous(Ag MP) multiwalled carbon nanotube modified glassy carbon electrode(PAN-Ag MP-MWCNT-GCE) was fabricated for the selective determination of an antihyperlipidimic drug, pitavastatin(PST). The fabricated electrochemical sensor was characterized by cyclic voltammetry(CV) and electrochemical impedance spectroscopy(EIS). The fabricated electrode was employed in quantifying and determining PST through differential pulse adsorptive stripping voltammetry(DPAdSV) and CV. The electrode fabrication proceeded with remarkable sensitivity to the determination of PST. The effect of various optimized parameters such as pH, scan rate(ν), accumulation time(t_(acc)), accumulation potential(U_(acc))and loading volumes of Ag MP-MWCNT suspension were investigated to evaluate the performance of synthesized electrochemical sensor and to propose a simple, accurate, rapid and economical procedure for the quantification of PST in pharmaceutical formulations and biological fluids. A linear response of PST concentration in the range 2.0×10^(-7)–1.6×10^(-6)M with low detection(LOD) and quantification(LOQ) limits of 9.66 ± 0.04 nM and 32.25 ± 0.07 nM, respectively, were obtained under these optimized conditions.

Peach-like ZnO microstructures are synthesized using vapor phase transport on MgO(001)substrates with a copper oxide(60 nm)buffer *** structure and morphology of the product are investigated using an x-ray diffractometer(XRD)and a field-emission scanning electron *** peaches have an average diameter of 3μm and a wurtzite *** study the optical properties,photoluminescence(PL)and Raman spectroscopy are employed.A strong UV emission at 380 nm in the PL spectra is observed,and a sharp and dominant peak at 437 cm^(-1) in the Raman spectrum can be assigned to the good crystallization of obtained *** addition,the growth mechanism of the peach-like ZnO structure is tentatively investigated based on the EDX analysis and growth time.

Magnesium hydride(MgH_(2))is the most feasible and effective solid-state hydrogen storage material,which has excellent reversibility but initiates decomposing at high temperatures and has slow kinetics ***,zinc titanate(Zn_(2)TiO_(4))synthesised by the solid-state method was used as an additive to lower the initial temperature for dehydrogenation and enhance the re/dehydrogenation behaviour of MgH_(2).With the presence of Zn_(2)TiO_(4),the starting temperature for the dehydrogenation of MgH_(2)was remarkably lowered to around 290℃–305℃.In addition,within 300 s,the MgH_(2)–Zn_(2)TiO_(4)sample absorbed 5.0 wt.%of H_(2)and 2.2–3.6 wt.%H_(2)was liberated from the composite sample in 30 min,which is faster by 22–36 times than as-milled MgH_(2).The activation energy of the MgH_(2)for the dehydrogenation process was also downshifted to 105.5 k j/mol with the addition of Zn_(2)TiO_(4)indicating a decrease of 22%than as-milled MgH_(2).The superior behaviour of MgH_(2)was due to the formation of Mg Zn_(2),MgO and MgTiO_(3),which are responsible for ameliorating the re/dehydrogenation behaviour of MgH_(2).These findings provide a new understanding of the hydrogen storage behaviour of the catalysed-MgH_(2)system.

Growth and characterization of ZnO multipods on functional surfaces with different sizes and shapes of Ag particles

Three-dimensional ZnO multipods are successfully synthesized on functional substrates using the vapor transport method in a quartz tube. The functional surfaces, which include two different distributions of Ag nanoparticles and a layer of commercial Ag nanowires, are coated onto silicon substrates before the growth of ZnO nanostructures. The structures and morphologies of the ZnO/Ag heterostructures are investigated using X-ray diffraction and field emission scanning electron microscopy. The sizes and shapes of the Ag particles affect the growth rates and initial nucleations of the ZnO structures, resulting in different numbers and shapes of multipods. They also influence the orientation and growth quality of the rods. The optical properties are studied by photoluminescence, UV-vis, and Raman spectroscopy. The results indicate that the surface plasmon resonance strongly depends on the sizes and shapes of the Ag particles.
