Prognostic significance and clinical relevance of Sprouty 2 protein expression in human hepatocellular carcinoma

BACKGROUND:In vitro experiments and mice models have confirmed the importance of Sprouty 2(Spry2) in inhibiting tumorigenesis and the progression of human ***,the prognostic value of Spry2 in cancer patients remains *** study is aimed to investigate the clinical relevance and prognostic significance of Spry2 expression in patients with hepatocellular carcinoma(HCC).METHODS:With samples from 240 randomly-selected HCC patients who underwent surgery,immunohistochemistry was used to investigate Spry2 expression on tissue *** correlation of Spry2 expression with survival was estimated by the Kaplan-Meier method and univariate/multivariate Cox proportional hazard regression ***2,ERK and phospho-ERK expression in HCC cell lines was detected by Western ***:Among the patients,86.3%(207 of 240) exhibited down-regulation of Spry2 *** negative for Spry2 showed poorer survival(P=0.002) and increased recurrence(P=0.003).Multivariate analysis further established Spry2 as an independent predictor of postoperative recurrence in HCC patients(HR=1.47;95% CI,1.02-2.08;P=0.037).Downregulation of Spry2 was associated with highly malignant phenotypes like vascular invasion and advanced tumor stages,and was positively correlated with the metastatic potential of HCC cell ***:In the era of molecular targeted therapy,the expression of Spry2 in HCC may have relevant clinical significance and turn out to be a key factor in prognostic assessment and in treatment planning.

CCAAT/enhancer binding protein α(C/EBPα)is a transcriptional regulatory factor that inhibits cell proliferation,andalternative translational initiation produces two polypeptides,C/EBPcαp30 and C/EBPα*** expression profiling,it was revealed that C/EBPαp30 specifically inhibited a unique set of genes,including MPP11,p84N5 and SMYD2,which were not affected by C/EBPαp42 in both QSG-7701 hepatocyte cell line and QGY-7703 hepatoma ***-quantitative RT-PCR analysis independently confirmed these *** immunoprecipitation assay showed thatC/EBPαp30 bound to the promoters of these genes more strongly than C/EBPα*** clinical hepatoma samples inwhich C/EBPα was downregulated,all three genes specifically inhibited by C/EBPαp30 were ***,repression of MPP11,p84N5 and SMYD2 genes might not be directly involved in C/EBPαp30-mediated growth *** data suggest that C/EBPαp30 regulates a unique set of target genes and is more than a dominant-negativeregulator of C/EBPαp42.

目的制订《临床试验框架下噬菌体治疗多重耐药菌感染患者护理专家共识》(以下简称《共识》),规范临床试验框架下噬菌体治疗多重耐药菌感染患者的护理实践流程。方法成立《共识》构建小组,通过查阅文献和开展利益相关者访谈,形成《共识》初稿;通过2轮德尔菲专家咨询和4轮小组会议形成《共识》终稿。结果《共识》围绕多重耐药菌感染患者治疗开始前、治疗期间、治疗结束后3个阶段展开,包括临床研究护士职责与资质、患者评估与入组、治疗环境准备、噬菌体制剂准备、依据治疗方案执行给药、感控措施执行与督查、观察监测、追踪随访、数据记录与管理等。结论《共识》综合了文献报道、临床经验和专家建议,具有一定的科学性和实用性,可为在临床试验框架下护理噬菌体治疗多重耐药菌感染患者方面提供指导。

Analysis of HLA alleles polymorphism in Chinese patients with primary biliary cirrhosis

BACKGROUND: A familial dustering of patients with primary biliary cirrhosis (PBC) and the presence of immunological abnormalities in family members suggest a genetic component involved in the pathogenesis of PBC. The aims of this study are to investigate the frequencies of human leukocyte antigen HLA-A, -B, and -DRB1 alleles in Chinese patients with PBC by polymerase chain reaction (PCR)-based techniques, and to assess the correlation of the above-mentioned HLA with some clinical and laboratory features. METHODS: Genotyping of HLA alleles were performed in 65 well-characterized PBC patients and 431 healthy controls with sequence-specifc primers PCR amplification. RESULTS: HLA-DRB1~*07 allele detected in 19 of the 65 (29.2%) PBC patients was subtyped as DRB1~*0701, as well as in 13.9% of controls (P_C<0.05, OR=2.55, 95% CI: 1.4-4.6). An increased frequency of DRB1~*03 (18.4% vs. 7.2% in healthy controls) and a decreased frequency of DRB1~*12 (16.9% vs. 28.8%) in PBC patients were statistically significant. There was no association with HLADRB1~*08 reported. The frequencies for HLA-A, B and the other DRB1 alleles were similar between patients and healthy controls. CONCLUSIONS: The susceptibility to PBC in Chinese individuals is associated with DRB1~*0701 allele. This association differs from that in North Americans, South Americans, North Europeans and even Japanese, but it is not restricted to any particular subgroup of patients.

目的　了解儿科感染性疾病中肺炎克雷伯菌和大肠埃希菌产超广谱 β内酰胺酶 (ESBLs)的发生率及其对 16种抗生素的耐药情况。方法　应用纸片扩散确证法检测ESBLs,Kirby Bauer(KB)法测定细菌对抗菌药物的敏感性。结果　肺炎克雷伯菌产酶率为 2 5 .3 % ,大肠埃希菌为 14.6 %。产ESBLs株对 16种抗生素的耐药性显著高于非产ESBLs ;产ESBLs菌对头孢类和青霉素类抗生素的耐药率较高 ,除头孢他啶以外 ,其耐药率均在6 0 %以上。产ESBLs菌对亚胺培南的敏感率为 10 0 %。结论　儿科中产ESBLs的发生率已相当高 ,实验室应积极开展产ESBLs的检测。

Diagnosis and differentiation of rejection after liver transplantation: report of 5 cases

BACKGROUND: The most common complications after liver transplantation are acute and chronic rejection. Al- though special changes can be found clinically and patho- logically in both of them, sometimes other complications such as preservation injury, recurrent disease ( HBV) and hepatic artery thrombosis should be differentiated from them. METHODS: Five patients with various complications after liver transplantation were synthetically analyzed according to their clinical information, laboratory tests and liver biop- sy findings so as to find out the clinical rules and the histo- logical features. RESULTS: Special changes and diagnostic standards were found clinically and pathologically in both acute and chro- nic rejection. Other complications such as fibrosing choles- tatic hepatitis and hepatic artery thrombosis also had then- own pathological features. Evaluation of donor liver should be done before liver transplantation. CONCLUSION: It is very important to understand and master the diagnostic standards of rejection and the major points for its differentiation.

Solid-pseudopapillary tumors (SPTs) of the pancreas have been reported as rare lesions with' low malignant potential' occurring mainly in young women. This study was designed to define the clinicopathological characteristics and the effect of surgical intervention. METHODS:A retrospective clinical analysis was made of 21 patients with solid-pseudopapillary tumor of the pancreas admitted from June 1994 to December 2004. RESULTS:Abdominal pain as the major complaint was found in 13 patients, and palpable abdominal mass in 7. Imaging examination showed the abdominal mass clearly, but diagnosis was not defined. In 12 patients, tumors were located in the pancreatic head and in 9 patients, in the pancreatic tail. The average diameter of the tumor was 9.5 cm (range, 3-20 cm). One patient had liver metastasis. The diagnosis of the tumor was proved pathologically during operation in 14 patients, and other 3 patients who had been diagnosed as having insulinoma and other malignant tumors were confirmed by paraffin section and enzyme labeling after operation. Eighteen patients (85.7% ) were followed up with a median period of 24 months, 1-60 months without tumor recurrence. CONCLUSION:Huge pancreatic masses of the pancreas in young women are suggestive of solid-pseudopapillary tumors. Aggressive surgical therapy can result in good prognosis in these patients.

Background Tuberous sclerosis (TS) is an autosomal dominant disorder with a significant range of clinical expressions. The involvement of vital organs, such as the brain, kidney, heart and lung is the main cause of death in patients with TS. The aim of this study is to summarize the charateristic cutaneous features and common extracutaneous involvement of TS, which are helpful to the early detection of visceral involvement.

Receptor tyrosine kinases positively regulate BACE activity and Amyloid-β production through enhancing BACE internalization

Amyloid-β (AP) peptide, the primary constituent of senile plaques in Alzheimer's disease (AD), is generated byβ-secretase- and γ-secretase-mediated sequential proteolysis of the amyloid precursor protein (APP). The aspartic pro-tease, β -site APP cleavage enzyme (BACE), has been identified as the main P-secretase in brain but the regulation of itsactivity is largely unclear. Here, we demonstrate that both BACE activity and subsequent Aβ production are enhancedafter stimulation of receptor tyrosine kinases (RTKs), such as the receptors for epidermal growth factor (EGF) and nervegrowth factor (NGF), in cultured cells as well as in mouse hippocampus. Furthermore, stimulation of RTKs also inducesBACE internalization into endosomes and Golgi apparatus. This enhancement of BACE activity and Aβ production uponRTK activation could be specifically inhibited by Src family kinase inhibitors and by depletion of endogenous c-Src withRNAi, and could be mimicked by over-expressed c-Src. Moreover, blockage of BACE internalization by a dominantnegative form of Rab5 also abolished the enhancement of BACE activity and Aβ production, indicating the requirementof BACE internalization for the enhanced activity. Taken together, our study presents evidence that BACE activity andAβ production are under the regulation of RTKs and this is achieved via RTK-stimulated BACE internalization, andsuggests that an aberration of such regulation might contribute to pathogenic Aβ production.

Formation and Characterization of Bone-like Nanoscale Hydroxyapatite in Glass Bone Cement

Glass based bone cement (GBC) was synthesized by mixing CaO-SiO2-P2O5 based glass powder with ammonium phosphate liquid medium. Bone-like hydroxyapatite (HAP, Ca10(PO4)6(OH)2) was found to form after GBC was immersed in simulated body fluid (SBF). HAP crystal grew with an increasing time along c axle and reached about 200 nm in length after 30 days, however, the end plane granularity remained 30-50 nm. The chemical composition, crystal structure and morphology of HAP formed from GBC were proved to have great resemblance with living HAP. It is believed that GBC was a desirabie biomedicai material with high bioactivity. Furthermore, the high compressive strength guaranteed the possibility of GBC in clinical application.
