Bio-scaffolds which are most commonly open celled porous structures are increasingly used for tissue engineering and regenerative medicine. A number of studies have shown that the bulk properties of such irregular structures are poorly modeled using idealized unit cell approaches. The paper therefore uses novel image based meshing techniques to explore both fluid flow and bulk structural properties of a bone scaffold, as accurate modeling of bio-scaffolds with non-uniform cellular structures is very important for the development of optimal scaffolds for tissue engineering application. In this study, a porous hydroxyapatite/tricalcium phosphate (HA/TCP) bone scaffold has been scanned in a Micro-CT scanner, and converted into a volumetric mesh using image processing software developed by the authors. The resulting mesh was then exported to commercial FEA and CFD solvers for analysis. Initial FEA and CFD studies have shown promising results and have highlighted the importance of accurate modeling to understand how microstructures influence the mechanical property of the scaffold, and to analyze flow regimes through the sample. The work highlights the potential use of image based meshing for the ad hoc characterization of scaffolds as well as for assisting in the design of scaffolds with tailored strength, stiffness, and transport properties.

AIM: To evaluate need for and efficacy of a structured gastroenterology didactic session in expanding awareness and understanding of digestive disorders. METHODS: A four-day symposium was developed with didactic sessions (days 1, 2) and practical endoscopy (days 3, 4). Didactic sessions included case presentations highlighting pathophysiology and management. One nurse and four practicing gastroenterologists from the United Kingdom led lectures and supervised work-shops with audience participation. Practical endoscopy focused on diagnostic and therapeutic procedures and their application to diagnosis and treatment of ailments of the gastrointestinal tract. Preand post-workshop questionnaires were distributed to participants during didactic sessions. A pre-workshop questionnaire gauged expectations and identified objectives to be met at thesymposium. Post-workshop questionnaires were administered to assess efficacy of each session. Participants graded sessions from 1 (poor) to 5 (excellent) on quality of case presentations, knowledge, clarity and mode of presentation. We assessed if time allotted to each topic was sufficient, value of sessions, impact on practice and interest in future symposiums. RESULTS: There were 46 attendees on day 1: 41% undergraduates, 41% residents, 11% consultants and 4% unspecified. Day 2 (a Saturday) had 24 participants: 17% undergraduates, 71% residents, 9% consultants, 4% unspecified. Primary pre-workshop symposium expectation was to gain knowledge in: general gastroenterology (55.5%), practical endoscopy (13.8%), pediatric gastroenterology (5%), epidemiology of gastrointestinal disorders specific to Zambia (6%), and interaction with international speakers (6%). The post-symposium questionnaire was answered by 19 participants, of whom 95% felt specific aims were met; all would attend future conferences and recommend to others. The impacts of land-cover composition on urban temperatures, including temperature extremes, are well documented. Much less attention has been devoted to the consequences of land-cover configuration, most of which addresses land surface temperatures. This study explores the role of both composition and configuration—or land system architecture—of residential neighborhoods in the Phoenix metropolitan area, on near-surface air temperature. It addresses two-dimensional, spatial attributes of buildings, impervious surfaces, bare soil/ rock, vegetation and the “urbanscape” at large, from 50 m to 550 m at 100 m increments, for a representative 30-day high sun period. Linear mixed-effects models evaluate the significance of land system architecture metrics at different spatial aggregation levels. The results indicate that, controlling for land-cover composition and geographical variables, land-cover configuration, specifically the fractal dimension of buildings, is significantly associated with near-surface temperatures. In addition, statistically significant predictors related to composition and configuration appear to depend on the adopted level of spatial aggregation.

This paper discussed effects of lowland-associated influences on upland ecology, food security and biocultural diversity in the Sarangani farming communities of the Philippines. In the uplands of Sarangani Province, the conservation of traditional rice varieties, the centrality of rice in tribal life, and the continued observance of planting rituals attest to its cultural significance and convey a common desire for cultural preservation and community solidarity. Economic and socio-political pressures had transformed tribal communities, although vestiges of traditional farming systems are still being practiced in remote sitios（villages）. Changing land use patterns had also resulted in shrinking farm sizes and consequently in food insecurity in the Sarangani uplands. Extractive industries（i.e. logging, mining and charcoal making） and swidden farming were observed to cause widespread environmental degradation, while modern agriculture had undermined the capacity of indigenous peoples to survive because of their complete dependence on lands and resources. With the reality that cultural and biological diversities are inextricably linked, trans-disciplinary strategies coupling indigenous knowledge systems with scientific knowledge should, therefore, be instituted to save the Sarangani upland ecosystem, the indigenous peoples and their tribal resources.

Predicting an Athlete’s Physiological and Haematological Response to Live High-Train High Altitude Training Using a Hypoxic Sensitivity Test

Purpose Elite endurance runners frequently utilise live high-train high(LHTH)altitude training to improve endurance per-formance at sea level(SL).Individual variability in response to the hypoxic exposure have resulted in contradictory *** the present case study,changes in total haemoglobin mass(tHbmass)and physiological capacity,in response to 4-weeks of LHTH were *** tested if a hypoxic sensitivity test(HST)could predict altitude-induced adaptations to *** Fifteen elite athletes were selected to complete 4-weeks of LHTH(~2400 m).Athletes visited the laboratory for preliminary testing(PRE),to determine lactate threshold(LT),lactate turn point(LTP),maximal oxygen uptake VO_(2max)and *** LHTH,athletes completed daily physiological measures[arterial oxygen saturation(SpO2)and body mass]and subjective wellbeing *** was repeated,for those who completed the full camp,post-LHTH(POST).Additionally,athletes completed the HST prior to *** A difference(P<0.05)was found from PRE to POST in average tHbmass(1.8%±3.4%),VO_(2max)(2.7%±3.4%),LT(6.1%±4.6%)and LTP(5.4%±3.8%),after 4-weeks *** revealed a decrease in oxygen saturation at rest(ΔSp_(r))and higher hypoxic ventilatory response at rest(HVRr)predicted individual changes *** hypoxic cardiac response at rest(HCRr)and higher HVRr predicted individual changes VO_(2max).Conclusion Four weeks of LHTH at~2400 m increased tHbmass and enhanced physiological capacity in elite endurance *** was no observed relationship between these changes and baseline characteristics,pre-LHTH serum ferritin levels,or reported incidents of musculoskeletal injury or *** HST did however,estimate changes in tHbmass and VO_(2max).HST prior to LHTH could allow coaches and practitioners to better inform the acclimatisation strategies and train-ing load application of endurance runners at altitude.

Genomic assemblies of the unicellular green alga Chlamydomonas reinhardtii have provided important resources for ***,assembly errors,large gaps,and unplaced scaffolds as well as strain-specific variants currently impede many types of *** combining PacBio HiFi and Oxford Nanopore long-read technologies,we generated a de novo genome assembly for strain CC-5816,derived from crosses of strains CC-125 and *** methods of evaluating genome completeness and base-pair error rate suggest that the final telomere-to-telomere assembly is highly *** CC-5816 assembly enabled previously difficult analyses that include characterization of the 17 centromeres,rDNA arrays on three chromosomes,and 56 insertions of organellar DNA into the nuclear *** Nanopore sequencing,we identified sites of cytosine(CpG)methylation,which are enriched at *** analyzed CRISPR-Cas9 insertional mutants in the PF23 *** of the three alleles produced progeny that displayed patterns of meiotic inviability that suggested the presence of a chromosomal *** Nanopore reads from pf23-2 and pf23-3 onto the CC-5816 genome showed that these two strains each carry a translocation that was initiated at the PF23 gene locus on chromosome 11 and joined with chromosomes 5 or 3,*** translocations were verified by demonstrating linkage between loci on the two translocated chromosomes in meiotic *** three pf23 alleles display the expected short-cilia phenotype,and immunoblotting showed that pf23-2 lacks the PF23 *** CC-5816 genome assembly will undoubtedly provide an important tool for the Chlamydomonas research community.
