Gargantulides B and C,two new and highly complex 52-membered glycosylated macrolactones,were isolated from Amycolatopsis sp.strain CA-230715 during an antibacterial screening campaign.The structures of these giant macrolides were elucidated by 2D NMR spectroscopy and shown to be related to gargantulide A,although containing additional β-glucopyranose and/orα-arabinofuranose monosaccharides separately attached to their backbones.Genome sequencing allowed the identification of a strikingly large 216 kbp biosynthetic gene cluster,among the largest type Ⅰ PKs clusters described so far,and the proposal of a previously unreported biosynthetic pathway for gargantulides A-C.The absolute configurations of gargantulides B and C were assigned based on a combination of NMR and bioinformatics analysis of ketoreductase and enoylreductase domains within the multimodular type Ⅰ PKs.In addition,the absolute stereochemistry of gargantulide A has now been revised and completed.Gargantulides B and C display potent antibacterial activity against a set of drug-resistant Gram-positive bacteria and moderate activity against the clinically relevant Gram-negative pathogen Acinetobacter baumannii.

Concrete structures may suffer considerable restraint stresses during their hardening period. This is caused by several deformation impacts, especially temperature field changings due to hydration heat and volume changes due to autogenous shrinkage. Mainly affected are massive concrete members, but also the application of new concrete types or the erection of outstanding constructions requires further investigations in this context. 3D-FEM analyses of hydration heat induced temperature development in combination with the well known shrinkage give sufficient results for the deformation impact. The according elastic restraint stresses can be determined with consideration of the concrete’s rising elastic modulus and the restraint degree of the system. But due to duration of the heat flow process, the height of restraint stresses is strongly dependent from the viscoelasticity of the concrete. The viscoelastic effects consist of many components constituted by changing material properties influencing themselves. In practice, different simplified approaches are available for considering this in calculations. Their implementation in time step analyses is not generally admitted and requires expertise. In contrast, present research develops material models needing specific input parameters for every use case. This contribution focuses on a practicable approach considering the superposition of the viscoelastic behaviour of every stress increment in time step FEM analysis. The differentiation between the pure viscoelastic material behaviour (as it is given in the codes for idealistic conditions like creep or relaxation) and the according viscoelastic system response (addicted to the systems variable restraint degree) allows the transfer of this model into practice. As one application example of this approach, the compatibility check and the FEM-based recalculation of the monitoring program of a massive power plant slab will be presented.

Unambiguous diagnosis of the two main forms of inflammatory bowel diseases(IBD):Ulcerative colitis(UC)and Crohn’s disease(CD),represents a challenge in the early stages of the diseases.The diagnosis may be established several years after the debut of symptoms.Hence,protein biomarkers for early and accurate diagnostic could help clinicians improve treatment of the individual patients.Moreover,the biomarkers could aid physicians to predict disease courses and in this way,identify patients in need of intensive treatment.Patients with low risk of disease flares may avoid treatment with medications with the concomitant risk of adverse events.In addition,identification of disease and course specific biomarker profiles can be used to identify biological pathways involved in the disease development and treatment.Knowledge of disease mechanisms in general can lead to improved future development of preventive and treatment strategies.Thus,the clinical use of a panel of biomarkers represents a diagnostic and prognostic tool of potentially great value.The technological development in recent years within proteomic research(determination and quantification of the complete protein content)has made the discovery of novel biomarkers feasible.several IBD-associated protein biomarkers are known,but none have been successfully implemented in daily use to distinguish CD and UC patients.The intestinal tissue remains an obvious place to search for novel biomarkers,which blood,urine or stool later can be screened for.When considering the protein complexity encountered in intestinal biopsysamples and the recent development within the field of mass spectrometry driven quantitative proteomics,a more thorough and accurate biomarker discovery endeavor could today be performed than ever before.In this review,we report the current status of the proteomics IBD biomarkers and discuss various emerging proteomic strategies for identifying and characterizing novel biomarkers,as well as suggesting future targets for analysis.

Inappropriate e-waste-processing operation is a serious issue in developing countries since the early 2000's.Field research was conducted to clarify its influences and diffuse toxic substances in the environment at e-waste-processing sites in a village in northern Vietnam from 2012 to 2014.We investigated surface soil samples collected from e-waste-processing sites,open burning sites,and rice paddy sites.sediment samples and water samples were also collected from a river that runs through the village.The hazardous metal concentration in soil and river sediment samples indicated that 7 elements(Ca,Cu,Mo,Ni,Pb,sn,and Zn)had high concentrations as a result of e-waste-processing.Most notably,Pb and Cu had high concentrations in soil samples(Pb:26e2200 mg/kg,Cu:22e3000 mg/kg).The results of a multivariate analysis indicated that these 7 elements were closely correlated with e-waste-processing.Concentrations of hazardous metals showed an upward trend where e-waste-processing occurred during the 3-year study period,although it was not significant statistically at present.Open burning activity had a great influence on Cu accumulation,and maximum Cu concentrations increased from 340 mg/kg in 2012e3000 mg/kg in 2014 in soil samples from open-burning sites.There were large differences in the dominant hazardous metal depending on the type of e-waste-processing workshop because hazardous metal composition was influenced by the types of e-waste and the processing procedures.It is necessary to avoid e-waste processing in open-air environment to control emissions of hazardous substances.Especially,open burning should be prohibited.

HIV testing among persons with tuberculosis (TB) results in high-yield identification of persons infected with HIV. To evaluate differences in HIV testing among children versus adults with TB in Vietnam, we collected and analyzed age-disaggregated facility and aggregated provincial data from the National Tuberculosis Program. HIV testing was incompletely documented for >70% of children, whereas adult testing data were >90% complete. standardized training of personnel for universal HIV testing and documentation for children with TB could improve HIV case-detection and permit linking of children with HIV to antiretroviral treatment to prevent morbidity and mortality.

目的探讨一氧化氮(NO)与心肌缺血预调置作用的相关性。方法将50只体质量300～350 g雄性健康Wistar大鼠分为对照组、缺血/再灌注损伤(I/R)组、预调置组、L-硝基精氨酸(L-NNA组)和L-精氨酸(L-Arg)+L-NNA等5组,分别观察冠脉灌注压(CPP)和心肌蛋白、乳酸脱氢酶(LDH)、环一磷酸鸟苷(cGMP)和丙二醛(MDA)含量等心肌损伤指标的变化。结果与I/R组比较,预调置组心肌蛋白漏出量明显降低、LDH漏出量显著下降、心肌MDA含量明显降低、cGMP水平则明显升高(P均<0.01),CPP降低(P<0.05);L-NNA灌流组可以阻断预调置的心肌保护作用,L-NNA+L-Arg灌流组可逆转L-NNA阻抑预调置的心肌保护作用。结论心肌缺血预调置明显提高缺血心肌对缺血缺氧的耐受力,与改善心肌I/R损伤时NO缺陷密切相关。

strong>Introductionstrong>: Rheumatoid arthritis (RA) is the most common chronic inflammatory rheumatism. The increasing use of bone and joint ultrasound as a tool to aid in the detection of unrefined synovitis in developed countries, has led us to study the performance of ultrasound in the diagnosis of RA in subjects consulting in rheumatology at Cotonou. strong>study Method:strong> Descriptive cross-sectional study of patients with RA, selected on the basis of ACR/ EULAR 2010 criteria. A complete clinical examination with imaging including X-rays and ultrasounds of the hands and feet were performed in all patients who have given their consent. The data collected was analyzed using EPI data 3.1 and sPss 24.0 software. The significance rate was defined with a p-value s Kappa (k) test was used to assess agreement. strong>Results:strong> All 53 patients included had lesions on ultrasound. synovitis was found in 98.1% and bone erosions in 88.7%. There was no agreement in the wrists between clinical examination and ultrasound (k = -0.116), MCP (k = -0.097) and MTP (k = -0.031). Agreement was very low at the PPI level (k = 0.03). It was low at the different sites, between ultrasound and radiographic detection of lesions (k between 0.022 - 0.22). Age, positive immunologic RA factor and biological inflammatory syndrome were associated with ultrasound erosions (p = 0.0001) and only age was associated with active synovitis (p = 0.022). strong>Conclusion:strong> Bone and articular ultrasound is a good complementary alternative to clinical examination and radiography diagnostic in early diagnosis of RA in our developing countries, in the absence of MRI which remains a luxury.

虽然总体价格水平止跌回稳 ,但是反映消费需求的零售商品的价格指数却仍是负增长 ,消费不足仍是当今制约经济走出低谷的桎梏。从收入层次来看 ,富裕阶层消费在其收入中所占比例极低 ,中层阶层预期支出不稳定及消费供给滞后 ,贫困阶层低收入是制约消费需求的最根本原因。因此 ,必须加强对富裕阶层收入的宏观调控及税收管理 ,稳定中间阶层支出 ,改善供给结构 ,提高贫困阶层的收入 ,以期达到全面提高消费需求。
