Multi-clustering study on the association between human leukocyte antigen-DP-DQ and hepatitis B virus-related hepatocellular carcinoma and cirrhosis in Viet Nam

BACKGROUND Human leukocyte antigen(HLA)class II molecules are cell surface receptor proteins found on antigen-presenting *** and mutations in the HLA gene can affect the immune system and the progression of hepatitis *** To study the relation between rs2856718 of HLA-DQ,rs3077,and rs9277535 of HLA-DP,hepatitis B virus(HBV)-related cirrhosis,and hepatocellular carcinoma(HCC).METhoDS In this case-control study,the genotypes of these single nucleotide polymorphisms(SNPs)were screened in 315 healthy controls,471 chronic hepatitis B patients,250 patients with HBV-related liver cirrhosis,and 251 patients with HCC using TaqMan real-time *** conducted Hardy-Weinberg equilibrium and linkage disequilibrium tests on the genotype distributions of rs2856718,rs3077,and rs9277535 before hierarchical clustering analysis to build the complex interaction between the markers in each patient *** The physical distance separating these SNPs was 29816 kB with the disequilibrium(D’)values ranging from 0.07 to *** close linkage between rs3077 and rs9277535 was attributed to a distance of 21 *** D’value decreased from moderate in the healthy control group(D’=0.50,P<0.05)to weak in the hepatic disease group(D’<0.3,P<0.05).In a combination of the three variants rs2856718,rs3077,and rs9277535,the A allele decreased hepatic disease risk[A-A-A haplotype,risk ratio(RR)=0.44(0.14;1.37),P<0.05].The G allele had the opposite effect[G-A/G-G haplotype,RR=1.12(1.02;1.23),P<0.05].In liver cancer cases,the A-A-A/G haplotype increased the risk of HCC by 1.58(P<0.05).CONCLUSION Rs9277535 affects liver fibrosis progression due to HBV infection,while rs3077 is associated with a risk of HBVrelated *** link between rs2856718,rs3077,and rs9277535 and disease risk was determined using a multiclustering analysis.

Bioengineered miR-27b-3p and miR-328-3p modulate drug metabolism and disposition via the regulation of target ADME gene expression

Drug-metabolizing enzymes, transporters, and nuclear receptors are essential for the absorption, distribution, metabolism, and excretion(ADME) of drugs and xenobiotics. MicroRNAs participate in the regulation of ADME gene expression via imperfect complementary Watson–Crick base pairings with target transcripts. We have previously reported that Cytochrome P450 3A4(CYP3A4) and ATP-binding cassette sub-family G member 2(ABCG2) are regulated by miR-27b-3p and miR-328-3p,respectively. Here we employed our newly established RNA bioengineering technology to produce bioengineered RNA agents(BERA), namely BERA/miR-27b-3p and BERA/miR-328-3p, via fermentation. When introduced into human cells, BERA/miR-27b-3p and BERA/miR-328-3p were selectively processed to target miRNAs and thus knock down CYP3A4 and ABCG2 mRNA and their protein levels,respectively, as compared to cells treated with vehicle or control RNA. Consequently, BERA/miR-27b-3p led to a lower midazolam 10-hydroxylase activity, indicating the reduction of CYP3A4 activity. Likewise,BERA/miR-328-3p treatment elevated the intracellular accumulation of anticancer drug mitoxantrone, a classic substrate of ABCG2, hence sensitized the cells to chemotherapy. The results indicate that biologic miRNA agents made by RNA biotechnology may be applied to research on miRNA functions in the regulation of drug metabolism and disposition that could provide insights into the development of more effective therapies.

Influence of CeO2 on Properties and Activity of Oxide Catalysts in Carbon Monoxide Oxidation

A series of catalysts on the basis of 10 wt.% CuO/y-AI203, 10 wt.% CuO ＋ 10 wt.% Cr2O3/y-AI203 and 15 wt.% MnO2/y -A1203 have been prepared and modified by CeO2 with contents up to 20 wt.%. Physico-chemical properties of the catalysts were determined by the methods of BET Adsorption, XRD, and TPR. Oxidative activity of the catalysts was studied at the temperature range 90-220 ℃and CO concentration of 3 mol.%. Addition of CeO2 led to changes in physico-chemical properties of the catalysts and formation of novel active centres that increased the activity of CuO and Cr203 containing catalysts, but decreased the activity of those, containing MnO2. The catalyst sample containing 10 wt.% CuO and 15 wt.% CeO2 has been shown to be the best one for complete conversion of CO. At the given conditions on this catalyst the complete oxidation of CO to CO2 occurred at 130 ~C during more than 500 h.

Upconversion luminescence of Gd2O3:Er3+and Gd2O3:Er3+/silica nanophosphors fabricated by EDTA combustion method

Gd2O3:Er^3+nanophosphors were fabricated by the combustion method in presence of Na2 ethylene diamine tetra acetic acid(EDTA-Na2)as fuel at not high temperature(≤350℃)within a very short time of 5 *** added concentration of Er^3+ions in Gd2O3 matrix was changed from 0.5 mol%to 5.0 mol%.The X-ray diffraction pattern of samples indicates the monoclinic structure of Gd2O3:Er^3+.The morphology and chemical composition analysis of the Gd2O3:Er^3+samples are characterized by a field emission scanning electron microscope(FESEM)and a Fourier-transform infrared spectrometer(FTIR).The photoluminescence(PL),photo luminescence excitation(PLE)and upconversion(UC)at room temperature of the prepared materials with different concentrations of Er^3+were *** PL of Gd2O3:Er^3+nanomaterials are shown in visible at 545,594,623,648,688 nm under excitation at 275 *** emission bands from transitions of Er^3+from 2P3/2 to 4F9/2 are observed,UC luminescent spectra of the Gd2O3:Er^3+/silica nanocomposites under 976 nm excitation show the bands at 548 and 670 *** influence of excitation power at 980 nm for transitions were measured and *** results indicate that the upconversion process of Gd2O3:Er^3+/silica is two photons absorption *** low temperature dependence of UC luminescent intensities of the main bands of Gd2O3:Er^3+was investigated towards development of a nanotemperature sensor in the range of 10-300 K.

Trastuzumab resistance is one of the causes of poor prognosis in patients with human epidermal growth factor receptor 2(HER2)-positive(HER2+)breast cancer(BC).The truncated isoform of dopamine-and camPregulated phosphoprotein(t-DARPP)has been reported to be involved in trastuzumab therapy resistance and promoting tumor *** evaluate the t-DARPP expression in BC,paired tumors and surrounding normal tissues were analyzed by real-time polymerase chain reaction and confirmed higher DARPP-32 kDa family mRNA expression in HER2+BC tumor *** established 2 patient-derived xenografts(PDX)mice models to test the efficacy of trastuzumab,named model 1(non-responder)and model 2(responder).t-DARPP and p95-HER2 protein-protein interactions were detected in PDX tumor tissue from non-responders using Förster resonance energy transfer ***,there is no response from the ***,mechanistic studies using transwell and western blot assays demonstrated that t-DARPP could upregulate epithelial-mesenchymal transition signaling proteins,enhance p95-HER2 expression and promote cell *** found that quercetin effectively reduced t-DARPP expression in HER2+BC *** t-DARPP ShRNA-suppressed cells,quercetin synergistically enhanced trastuzumab-induced apoptotic cell death and G2/M phase *** conclusion,the combination of quercetin and trastuzumab treatment by targeting t-DARPP in HER2+BC patients has the potential as a biomarker for mitigating drug resistance.
