CYP1A1 , GSTM1 , GSTT1 and NQO1 polymorphisms and colorectal adenomas in Japanese men

AIM: To investigate the role of functional genetic poly-morphisms of metabolic enzymes of tobacco carcinogens in the development of colorectal adenomas. METHODS: The study subjects were 455 patients with colorectal adenomas and 1052 controls with no polyps who underwent total colonoscopy in a preretirement health examination at two Self Defense Forces hospitals. The genetic polymorphisms studied wereCYP1A1*2A (rs 4646903), CYP1A1*2C (rs 1048943), GSTM1 (null or non-null genotype), GSTT1 (null or non-null genotype) and NQO1 C609T (rs 1800566). Genotypes were determined by the polymerase chain reaction (PCR)-restriction fragment length polymorphism or PCR method using genomic DNA extracted from the buffy coat. Cigarette smoking and other life-style factors were ascertained by a self-administered questionnaire. The associations of the polymorphisms with colorectal adenomas were examined by means of OR and 95%CI, which were derived from logistic regression analysis. Statistical adjustment was made for smoking, alcohol use, body mass index and other factors. The gene-gene interaction and effect modification of smoking were evaluated by the likelihood ratio test. RESULTS: None of the five polymorphisms showed a significant association with colorectal adenomas, nor was the combination of GSTM1 and GSTT1 . A borderline significant interaction was observed for the combination of CYP1A1*2C and NQO1 (P = 0.051). The OR associated with CYP1A1*2C was significantly lower than unity among individuals with the NQO1 609CC genotype. The adjusted OR for the combination of the CYP1A1*2C allele and NQO1 609CC genotype was 0.61 (95%CI: 0.42-0.91). Although the interaction was not statistically significant (P = 0.24), the OR for individuals carrying the CYP1A1*2C allele and GSTT1 null genotype decreased significantly compared with those who had neither CYP1A1*2C allele nor GSTT1 null genotype (adjusted OR: 0.69, 95%CI: 0.49-0.97). AIM: To retrospectively study pancreatic cancer patients with respect to their ABO blood type and diabetes. METHODS: Our analysis included a cohort of 1017 patients with pancreatic ductal cancer diagnosed at our hospital in Tokyo. They were divided into two groups: 114 patients with long-standing type 2 diabetes (DM group, defined as diabetes lasting for at least three years before the diagnosis of pancreatic cancer) and 903 patients without diabetes (non-DM group). Multivariate analysis was performed to identify factors that are associated with long-standing diabetes. The DM group was further divided into three subgroups according to the duration of diabetes (3-5 years, 5.1-14.9 years, and 15 years or more) and univariate analyses were performed. RESULTS: Of the 883 pancreatic cancer patients with serologically assessed ABO blood type, 217 (24.6%) had blood type O. Compared with the non-DM group, the DM group had a higher frequency of blood type B [odds ratio (OR) = 2.61, 95%CI: 1.24-5.47; reference group: blood type A]. Moreover, male (OR = 3.17, 95%CI: 1.67-6.06), older than 70 years of age (OR = 2.19, 95%CI: 1.20-3.98) and presence of a family history of diabetes (OR = 6.21, 95%CI: 3.38-11.36) were associated with long-standing type 2 diabetes. The mean ages were 64.8 ± 9.2 years, 67.1 ± 9.8 years, and 71.7 ± 7.0 years in the subgroups with the duration of diabetes, 3-5 years, 5.1-14.9 years, and 15 years or more, respectively (P = 0.007). A comparison of ABO blood type distribution among the subgroups also showed a significant difference (P = 0.03). CONCLUSION: The association of pancreatic cancer with blood type and duration of diabetes needs to be further examined in prospective studies.

Oxygen uptake(V O_(2))was measured during a non-exhaustive high-intensity intermittent cross-exercise(HIICE)protocol consisting of four alternating bouts of 20 s running(R)and three bouts of bicycle exercise(BE)at~160%and~170%maximal oxygen uptake(V O_(2)max),respectively,with 10 s between-bout rests(sequence R-BE-R-BER-BE-R).The V O_(2) during the last BE([52.2±5.0]mL⋅kg^(-1)⋅min^(-1))was significantly higher than the V O_(2)max of the BE([48.0±5.4]mL⋅kg^(-1)⋅min^(-1),n=30)and similar to that of *** clarifying the underlying mechanisms,a corresponding HIICE-protocol with BE and arm cranking ergometer exercise(AC)was used(sequence AC-AC-BE-AC-BE-AC-AC-BE).In some experiments,thigh blood flow was occluded by a cuff around the upper *** occlusion,the V O_(2) during the AC([39.2±7.1]mL⋅kg^(-1)⋅min^(-1)[6th bout])was significantly higher than the V O_(2)max of AC([30.2±4.4]mL⋅kg^(-1)⋅min^(-1),n=7).With occlusion,the corresponding V O_(2)([29.8±3.9]mL⋅kg^(-1)⋅min^(-1))was reduced to that of the V O_(2)max of AC and significantly less than the V O_(2) without *** findings suggest that during the last bouts of HIICE may exceed the of the specific exercise,probably because it is a summation of the V O_(2) for the ongoing exercise plus excess post-oxygen consumption(EPOC)produced by the previous exercise with a higher V O_(2)max.

The results of near-infrared photometric observations of a transit event of an extrasolar planet HAT-P-54b are presented herein. Precise near-infrared photometry was carried out using the Nayuta 2 m telescope at Nishi-Harima Astronomical Observatory, Japan and Nishi-Harima Infrared Camera (NIC). 170 J-, H-, and Ks-band images were taken in each band in 196 minutes. The flux of the planetary system was observed to decrease during the transit event. While the Ks-band transit depth is similar to that in the r-band, the J- and H-band transits are deeper than those in the Ks-band. We constructed simple models of the planetary atmosphere and found that the observed transit depths are well reproduced by inflated atmosphere containing H2S molecule.

I INTRODUCTIONTissue engineering has increasingly attracted as a promising new technology to assist tissue regeneration at body defects as well as biological functions of damaged or injured organs. There are three factors necessary for tissue engineering: growth factors, cells, and materials for scaffolding. Since among them is growth factor of protein or glycoprotein, which is susceptible to proteolysis and denaturation, if the growth factor is administered in solution form into the body, one cannot always expect the biological function. Therefore, it is of prime necessity to develop dosage forms for in vivo prolongation of the biological activity. One possible way is to incorporate a growth factor into an appropriate matrix for achieving controlled release of the factor at the site of action over a long time period. Numerous studies have been performed on protein release by taking advantage of polymer matrices[1-5],but there is a problem before us in the protein release technology, i.e., loss in the biological activity of protein released. It has been demonstrated that this activity loss mainly results from protein denaturation and deactivation during a formulation process with a polymer matrix. When exposed to harsh environmental changes, such as heating and exposure to sonication and organic solvents, protein is generally denatured, losing its biological activity[6-8]. Therefore, it is required to contrive a new formulation method for growth factor release under mild conditions to minimize protein deactivation. From this viewpoint, hydrogel is a preferable candidate for a release matrix because of its biosafety and inertness toward protein[9]. However, it should be noted that the period of protein release from hydrogels is mostly as short as a day because of their diffusion-controlled characteristics[1,2,9,10].

Characterization of the Temperature Conditions of Inside Narrow Rocky Outcrops that Serve as a Habitat for Semi-Fossorial Mammals

We characterized the temperature conditions inside narrow rocky outcrops that served as habitats for semi-fossorial small mammals in a mountainous locality on the Japanese Islands. Usually, it is considered that the narrow rocky outcrops have poorer resources than the soil ground of forest floors, which have rich vegetation and nutrition. On the basis of this tendency, it is considered that ecologically dominant species occupy the rich soil habitats and subordinate species are chased away to the narrow rocky outcrops by ecological species competitions, resulting in habitat segregation. However, the present temperature data revealed that the temperatures inside rocky terrains were more stable than the shaded ambient temperatures in the forest. The rocky habitats were apparently colder in summer and warmer in winter, in both daily maximum and minimum temperatures, than the ambient temperatures in the forest during the research period. In addition, the daily difference between maximum and minimum temperatures in the rocky habitats was apparently smaller than that of the ambient ones. These temperature conditions in the narrow rocky outcrops are advantageous to the small mammalian metabolic system. Namely, we estimate that the semi-fossorial small mammals are not chased out by the dominant species through ecological competitions and that the semi-fossorial small mammals may occupy the narrow rocky outcrops as a more advantageous habitat than the forest floor.

Human-induced pluripotent stem cell-derived cardiomyocytes(hiPSC-CMs)have been widely used in therapy of ischemic heart ***,there are still remaining issues that limit the therapeutic efficacy,such as immune rejection and low retention of *** adipose mesenchymal stromal cells(hADSCs)have been reported to be able to regulate the immune response,promote angiogenesis and promote the maturation of *** this study,we co-cultured these two types of cells on fiber scaffold made of biodegradable poly(D,L-lactic-co-glycolic acid)(PLGA)polymer for several days to develop a composited 3D cardiac tissue *** expected,the cells formed 231.00±15.14μm thickness tissue,with improved organization,alignment,ECM condition,contractile ability,and paracrine function compared to culture hiPSC-CMs only on PLGA ***,the composited 3D cardiac tissue sheet significantly promoted the engraftment and survival after *** composited 3D cardiac tissue sheet also increased cardiac function,attenuated ventricular remodeling,decreased fibrosis,and enhanced angiogenesis in rat myocardial infarction model,indicating that this strategy wound be a promising therapeutic option in the clinical scenario.

Efficacy and Safety of Basal-Supported Prandial GLP-1 Receptor Agonist Therapy

Aim: To assess the safety and efficacy of basal-supported prandial GLP-1 receptor agonist therapy (BPT)* in type 2 diabetes mellitus (T2DM). Methods: Patients with T2DM, who had previously received insulin injection therapy and who had had their treatment switched to BPT (liraglutide), were retrospectively recruited. The efficacy of BPT was assessed by determining changes in HbA1c, body weight and total daily insulin dose from baseline to 4 months after BPT initiation. Safety was assessed by comparing the frequency of hypoglycemic episodes at baseline and after 4 months. The Wilcoxon test was used to analyze changes in parameters throughout the study period. Results: Twenty-nine patients, previously treated with basal-supported oral therapy (BOT), basal-bolus insulin, or pre-mixed insulin, were recruited. When analyzed together, there was no change in HbA1c throughout the study period, but body weight decreased (baseline 68.8 ± 13.2 kg vs. month 4 67.3 ± 13.1 kg;p < 0.001). Total daily insulin dose decreased after 4 months (baseline 24.4 ± 15.5 U/day vs. month 4 14.7 ± 9.2 U/day;p < 0.001), and there was no change in the frequency of hypoglycemic episodes. Analysis was conducted within sub-groups based on previous treatment modality. In the BOT group, HbA1c decreased from baseline after 2 months and body weight did not change throughout the study period. In both the basal-bolus insulin group and the pre-mixed insulin group, HbA1c remained steady throughout and there was a decrease in body weight. No change in the frequency of hypoglycemia was observed in any of the sub-groups. Conclusion: BPT in T2DM was associated with weight loss without changes in glycemic control over 4 months, suggesting that it may be an effective and safe therapy.
