Infectious Spondylitis-Associated Staphylococcus aureus with Virulence Gene pvl or tst Causes More Necrosis than Apoptosis in Human Alveolar Basal Epithelial Cell Line A549

Methicillin-sensitive and resistant Staphylococcus aureus (MSSA and MRSA, respectively) can cause non-tuberculosis infectious spondylitis. In 43 cases of bacterial infectious spondylitis, Mycobacterium tuberculosis and S. aureus were the two major causative pathogens. MRSA caused more anterior operations and thoracic infections, while MSSA caused more posterior infections and lumbar infections. Differences between six S. aureus isolates from infectious spondylitis were characterized. MLST and staphylococcal cassette chromosome mec (SCCmec) analysis identified MSSA ST959 and ST30 isolates, MRSA ST239/SCCmec IIIA isolates 2 and 3, ST59/SCCmec IIIA-like isolate 6, and ST30/SCCmec IV isolate 5. While all of the isolates were resistant to penicillin and ampicillin, the MRSA isolates were more resistant than the MSSA isolates. Carbapenem-resistant MRSA ST239/SCCmec IIIA and ST59/SCCmec IIIA-like isolates of the agr1 type were also resistant to clindamycin and erythromycin. Leukocidin genes (pvl or lukED) and hemolysin genes (hla, hld and hlg) were present in all of the isolates. All six isolates caused more necrosis than apoptosis in the human alveolar basal epithelial cell line A549;however, ST59/SCCmec IIIA-like isolate 6, ST30/ SCCmec IV isolate 5 with pvl genes, and MSSA ST30 isolates with tst caused greater than 40% cell death after the 4-h incubation. Regardless of the MRSA isolate and its SCCmec type or the MSSA isolate, the infectious spondylitis-associated S. aureus isolates differed genetically, and the pvl and tst genes may be important genes for cell necrosis.

Temperature-dependent electrical properties of β-Ga_2O_3Schottky barrier diodes on highly doped single-crystal substrates

Beta-phase gallium oxide(β-Ga_2O_3) Schottky barrier diodes were fabricated on highly doped single-crystal substrates,where their temperature-dependent electrical properties were comprehensively investigated by forward and reverse current density – voltage and capacitance – voltage characterization. Both the Schottky barrier height and the ideality factor showed a temperature-dependence behavior, revealing the inhomogeneous nature of the Schottky barrier interface caused by the interfacial defects. With a voltage-dependent Schottky barrier incorporated into thermionic emission theory, the inhomogeneous barrier model can be further examined. Furthermore, the reverse leakage current was found to be dominated by the bulk leakage currents due to the good material and surface quality. Leakage current per distance was also obtained. These results can serve as important references for designing efficient β-Ga_2O_3 electronic and optoelectronic devices on highly doped substrates or epitaxial layers.

Platelet-rich plasma enhances adipose-derived stem cell-mediated angiogenesis in a mouse ischemic hindlimb model

AIM To evaluate the angiogenic effect of platelet-rich plasma(PRP)-preconditioned adipose-derived stem cells(ADSCs) both in vitro and in a mouse ischemic hindlimb *** ADSCs were divided based on culture medium: 2.5% PRP, 5% PRP, 7.5% PRP, and 10% PRP. Cell proliferation rate was analyzed using the MTS assay. The gene expression of CD31, vascular endothelial growth factor, hypoxia-inducible factors, and endothelial cell nitric oxide synthase was analyzed using reverse transcription polymerase chain reaction. Cell markers and structural changes were assessed through immunofluorescence staining and the tube formation assay. Subsequently, we studied the in vivo angiogenic capabilities of ADSCs by a mouse ischemic hindlimb *** The proliferation rate of ADSCs was higher in the 2.5%, 5%, and 7.5% PRP groups. The expression of hypoxia-inducible factor, CD31, vascular endothelial growth factor, and endothelial cell nitric oxide synthase in the 5% and 7.5% PRP groups increased. The 5%, 7.5%, and 10% PRP groups showed higher abilities to promote both CD31 and vascular endothelial growth factor production and tubular structure formation in ADSCs. According to laser Doppler perfusion scan, the perfusion ratios of ischemic limb to normal limb were significantly higher in 5% PRP, 7.5% PRP, and human umbilical vein endothelial cells groups compared with the negative control and fetal bovine serum(FBS) groups(0.88 ± 0.08, 0.85 ± 0.07 and 0.81 ± 0.06 for 5%, 7.5% PRP and human umbilical vein endothelial cells compared with 0.42 ± 0.17 and 0.54 ± 0.14 for the negative control and FBS, P < 0.01).CONCLUSION PRP-preconditioned ADSCs presented endothelial cell characteristics in vitro and significantly improved neovascularization in ischemic hindlimbs. The optimal angiogenic effect occurred in 5% PRP-and 7.5% PRPpreconditioned ADSCs.

Analytical Result on the Supercurrent Through a Superconductor/Quantum-Dot/Superconductor Junction

We present an analytical result for the supercurrent across a superconductor/quantum-dot/superconductor junction. By converting the current integration into a special contour integral, we can express the current as a sum of the residues of poles. These poles are real and give a natural definition of the Andreev bound states. We also use the exact result to explain some features of the supercurrent transport behavior.

Effect of Intra-Dot Coulomb Interaction on Andreev Reflection in Normal-Metal/Quantum-Dot/Superconductor System

We investigate the effect of intra-dot Coulomb interaction on the Andreev reflection in a normalmetal/quantum-dot/superconductor (N-QD-S) system with multiple levels in the quantum dot, in the regime where the intra-dot interacting constant is comparable to the energy gap of superconducting lead. By using nonequilibrium Green function method, the averaged occupation of electrons in the quantum dot and the Andreev reflection (AR) current are studied. Comparing to the case of non-interacting quantum dot, the system shows significant changes for the a two-step-like behavior; and the I-Vg shows two groups of peaks, separated by U and with equal heights, where Vg is the gate voltage and U denotes the intra-dot Coulomb interaction constant. (ii) For finite bias voltage, dips, superposed V ≥ U/2, extra AR current peaks occur between the two groups of the peaks. Besides, the properties of the heights of the AR current peaks are more complicated.

We study Andreev tunneling through a ferromagnet/quantum-dot (QD)/superconductor system. By usingnonequilibrum Green function method, the averaged occupation of electrons in QD and the Andreev tunneling currentare studied. Comparing to the norma-metal/quantum-dot/superconductor, the system shows significant changes: (i)The averaged occupations of spin-up and spin-down electrons are not equal. (ii) With the increase of the polarizationof ferromagnetic lead, the Andreev reflection current decreases. (iii) However, even the ferromagnetic lead reaches fullpolarization, the averaged occupation of spin-down electrons is not zero. The physics of these changes is discussed.

Validation of a steel dual-core self-centering brace （DC-SCB） for seismic resistance： from brace member to one-story one-bay braced frame tests

A steel dual-core self-centering brace （DC-SCB） is an innovative structural member that provides both energy dissipation and self-centering properties to structures, reducing maximum and residual drifts of structures in earthquakes. The axial deformation capacity of the DC-SCB is doubled by a parallel arrangement of two inner cores, one outer box and two sets of tensioning elements. This paper presents cyclic test results of a DC-SCB component and a full- scale one-story, one-bay steel frame with a DC-SCB. The DC-SCB that was near 8 m-long was tested to evaluate its cyclic behavior and durability. The DC-SCB performed well under a total of three increasing cyclic loading tests and 60 low- cycle fatigue loading tests without failure. The maximum axial load of the DC-SCB was near 1700 kN at an interstory drift of 2.5%. Moreover, a three-story dual-core self-centering braced frame （DC-SCBF） with a single-diagonal DC-SCB was designed and its first-story, one-bay DC-SCBF subassembly specimen was tested in multiple earthquake-type loadings. The one-story, one-bay subassembly frame specimen performed well up to an interstory drift of 2% with yielding at the column base and local buckling in the steel beam; no damage of the DC-SCB was found after all tests. The maximum residual drift of the DC-SCBF caused by beam local buckling was 0.5% in 2.0% drift cycles.

The choice that a solid system“makes”when adopting a crystal structure(stable or metastable)is ultimately governed by the interactions between electrons forming chemical *** we analyze six prototypical binary transition metal compounds and shed light on the connection between Mott physics and the behavior of the energy as a function of the spatial arrangement of the atoms in these ***,we find that the main qualitative features of this complex behavior in the Mott phase of these systems can be traced back to the fact that the strong d-electron correlations influence substantially the charge transfer mechanism,which,in turn,controls the electrostatic *** result advances our understanding of the influence of strong correlations on the crystal structure,opens a new avenue for extending structure prediction methodologies to strongly correlated materials,and paves the way for predicting and studying metastability and polymorphism in these systems.

BACKGROUND Spontaneous spinal epidural hematoma is a rare neurosurgical *** SUMMARY A 53-year-old healthy woman suffered from complete paraplegia in both legs and loss of all sensation below the xiphoid *** was diagnosed as acute spontaneous thoracic epidural hematoma caused by an intraspinal *** primary lab survey showed all within normal *** of a posteriorly epidural space-occupying lesion at the T4-T8 level of the spinal canal was confirmed on magnetic resonance imaging.A decompressive laminectomy was performed from the T4 to T7 levels at the sixth hour following abrupt onset of complete *** lesion was confirmed as *** patient recovered well within one *** This study reports a case of acute spontaneous thoracic epidural hematoma caused by an intraspinal lymphangioma with well recovery after surgical intervention.
