通过密码子优化改造编码sn-1,3位置专一性疏绵状嗜热丝孢菌脂肪酶(TLL)基因,同时通过重叠PCR技术合成该基因,并将其克隆到巴斯德毕赤酵母的表达载体p PIC9K中。结果表明,TLL脂肪酶被高效胞外表达,在摇瓶中发酵168 h得到的发酵液的上清脂肪酶表达量达到0.1 mg/m L,对应的p NPP水解酶活达到312.72U/m L。与对应的原始编码基因重组菌酶活(179.42 U/m L)相比,密码子优化后酶活提高74.29%,表明密码子优化策略成功提高了TLL在巴斯德毕赤酵母中的重组表达。比较毕赤酵母来源的重组TLL与商业化的米曲霉酶学性质发现它们具有相似的最适p H和温度耐受范围。另外,它们在有机溶剂耐受性、表面活性剂和金属离子效应以及位置选择性方面均较为相似,表明毕赤酵母来源的重组TLL同样具有商业化应用的潜力。

WmG （World masters Games） athletes have either pursued a physically active lifestyle for an extended period of time or have initiated exercise/sport in later life. This unique cohort of middle-aged to older-aged adults remains relatively uninvestigated with regards to various measures of health. With a need for multifaceted solutions to the obesity epidemic, investigating special populations such as those competing in sport at older ages may further the understanding of the nexus between aging, physical activity and obesity. This study aims to investigate the BmI （body mass index） of North American WmG competitors with respect to national health guidelines and demographics. An online survey was utilized to collect demographic information from athletes competing at the Sydney WmG. BmI was derived using the participant＇s height and body mass. A total of 928 （46.7% male, 53.3% female） participants from Canada and the United States （age： 52.6 ± 9.8 years） completed the survey. The top 5 sports in which participants competed were football （25.6%）, track/field （15.4%）, swimming （8.4%）, volleyball （8.2%）, and softball （7.8%）. Female and male BmI （kg/m2） across all sports were：〉 30 （obese： 13.9%）, 25-29.9 （overweight： 34.1%）, 18.5-24.9 （normal： 50.3%）, and 〈18.5 （underweight： 1.7%）. Data indicated that BmI was a health risk factor for 13.9% of the participants and a developing risk factor for 34.1% of the participants. Analysis demonstrated a significantly reduced （P 〈 0.05） classification of obesity of the North American WmG competitors when compared to Canadian and United States national populations. It is believed that adherence to exercise improves indices of general health. A key index of health （obesity） is significantly lower in incidence for North American WmG competitors when compared to Canadian and US populations.

Hydroxyapatite/alumina nanocrystalline composite powders needed for various biomedical applications were successfully synthe- sized by sol-gel process. Structural and morphological investigations of the prepared composite powders were performed using X-ray dif- fractometer （XRD）, scanning electron microscopy （SEm）, X＇pert HighScore software, and Clemex Vision image analysis software. The re- suits show that the crystallite size of the obtained composite powders is in the range of 25 to 90 nm. SEm evaluation shows that the obtained composite powders have a porous structure, which is very useful for biomedical applications. The spherical nanoparticles in the range of 60 to 800 nm are embedded in the agglomerated clusters of the prepared composite powders.

Exocrine pancreatic insufficiency(EPI), an important cause of maldigestion and malabsorption, results from primary pancreatic diseases or secondarily impaired exocrine pancreatic function. Besides cystic fibrosis and chronic pancreatitis, the most common etiologies of EPI, other causes of EPI include unresectable pancreatic cancer, metabolic diseases(diabetes); impaired hormonal stimulation of exocrine pancreatic secretion by cholecystokinin(CCK); celiac or inflammatory bowel disease(IBD) due to loss of intestinal brush border proteins; and gastrointestinal surgery(asynchrony between motor and secretory functions, impaired enteropancreatic feedback, and inadequate mixing of pancreatic secretions with food). This paper reviews such conditions that have less straightforward associations with EPI and examines the role of pancreatic enzyme replacement therapy(pert). Relevant literature was identified by database searches. most patients with inoperable pancreatic cancer develop EPI(66%-92%). EPI occurs in patients with type 1(26%-57%) or type 2 diabetes(20%-36%) and is typically mild to moderate; by definition, all patients with type 3 c(pancreatogenic) diabetes have EPI. EPI occurs in untreated celiac disease(4%-80%), but typically resolves on a gluten-free diet. EPI manifests in patients with IBD(14%-74%) and up to 100% of gastrointestinal surgery patients(47%-100%; dependent on surgical site). With the paucity of published studies on pert use for these conditions, recommendations for or against pert use remain ambiguous. The authors conclude that there is an urgent need to conduct robust clinical studies to understand the validity and nature of associations between EPI and medical conditions beyond those with proven mechanisms, and examine the potential role for pert.

myelin-associated inhibitory factors within the central nervous system（CNS） are considered to be one of the main obstacles for axonal regeneration following disease or injury. The nogo receptor 1（NgR1） has been well documented to play a key role in limiting axonal regrowth in the injured and diseased mammalian CNS. However, the role of nogo receptor in immune cell activation during CNS inflammation is yet to be mechanistically elucidated. microglia/macrophages are immune cells that are regarded as pathogenic contributors to inflammatory demyelinating lesions in multiple sclerosis（mS）. In this study, the animal model of mS, experimental autoimmune encephalomyelitis（EAE） was induced in ngr1^＋/＋ and ngr1^–/– female mice following injection with the myelin oligodendrocyte glycoprotein（mOG_（35–55）） peptide. A fatemap analysis of microglia/macrophages was performed throughout spinal cord sections of EAE-induced mice at clinical scores of 0, 1, 2 and 3, respectively（increasing locomotor disability） from both genotypes, using the CD11 b and Iba1 cell markers. Western immunoblotting using lysates from isolated spinal cord microglia/macrophages, along with immunohistochemistry and flow cytometric analysis, was performed to demonstrate the expression of nogo receptor and its two homologs during EAE progression. myelin protein engulfment during EAE progression in ngr1^＋/＋ and ngr1^–/– mice was demonstrated by western immunblotting of lysates from isolated spinal cord microglia/macrophages, detecting levels of Nogo-A and mOG. The numbers of m1 and m2 microglia/macrophage phenotypes present in the spinal cords of EAE-induced ngr1^＋/＋ and ngr1^–/– mice, were assessed by flow cytometric analysis using CD38 and Erg-2 markers. A significant difference in microglia/macrophage numbers between ngr1^＋/＋ and ngr1^–/– mice was identified during the progression of the clinical symptoms of EAE, in the white versus gray matter regions of the spinal cord. Getting the right people in the right place at the right time has always been a major organizational challenge. In ancient times this process seems to have been accomplished based on the scheme of arrangements being contained in the leader＇s mind and instructions communicated verbally. modem approaches to solving the twin challenges of first thinking through the ＇plan＇ and then communicating the plan to the people who need to do ＇the right work, at the right time, in the right place＇ use sophisticated graphics, charts, diagrams, and computa- tions. This paper traces the development of the concepts most project managers take for granted including bar charts and critical path schedules from their origins （which are far earlier than most people think） through to the modem day. The first section of the paper looks at the development of concepts that allow the visualization of time and other data. The second looks at the shift from static representations to dynamic modeling through the emergence of computers, dynamic calculations and integrated data from the 1950s to the present time.
