Striatins and STRIPAK complex partners in clinical outcomes of patients with breast cancer and responses to drug treatment

Objective: Striatins(STRNs) family, which contains three multi-domain scaffolding proteins, are cornerstones of the striatins interacting phosphatase and kinase(STRIPAK) complex. Although the role of the STRIPAK complex in cancer has become recognized in recent years, its clinical significance in breast cancer has not been fully ***: Using a freshly frozen breast cancer tissue cohort containing both cancerous and adjacent normal mammary tissues, we quantitatively evaluated the transcript-level expression of all members within the STRIPAK complex along with some key interacting and regulatory proteins of STRNs. The expression profile of each molecule and the integrated pattern of the complex members were assessed against the clinical-pathological factors of the patients. The Cancer Genome Atlas(TCGA) dataset was used to evaluate the breast cancer patients’ response to chemotherapies. Four human breast cancer cell lines, MDA-MB-231, MDA-MB-361, MCF-7, and SKBR-3, were subsequently adopted for in vitro ***: Here we found that high-level expressions of STRIP2, calmodulin, CCM3, MINK1 and SLMAP were respectively associated with shorter overall survival(OS) of patients. Although the similar pattern observed for STRN3, STRN4 and a contrary pattern observed for PPP2CA, PPP2CB and PPPR1A were not significant, the integrated expression profile of STRNs group and PPP2 group members constitutes a highly significant prognostic indicator for OS [P<0.001, hazard ratio(HR)=2.04, 95% confidence interval(95% CI), 1.36-3.07] and disease-free survival(DFS)(P=0.003, HR=1.40, 95% CI, 1.12-1.75). Reduced expression of STRN3 has an influence on the biological functions including adhesiveness and migration. In line with our clinical findings, the breast cancer cells responded to STRN3 knockdown with changes in their chemo-sensitivity, of which the response is also breast cancer subtype ***: Our results suggest a possible role of the STRIPAK complex

jadomycin is an antibiotic that has shown activities against bacteria, yeasts and fungi as well as cytotoxic properties to cancer cells. Because of the wide range of its inhibitory actions, jadomycin shows promise as a novel antibiotic and cancer treatment drug. Streptomyces venezuelae are aerobic bacteria that produce jadomycin and the size of bacterial population can significantly affect the yield of jadomycin. Therefore, the bacterial population must be accurately measured in order to standardize the reproducibility of jadomycin production process. In this study, a dehydrogenase activity measurement test, using triphenyl tetrazolium chloride (TTC), was used to measure the dehydrogenase activity of Streptomyces venezuelae during growth in maltose-yeast extract-malt extract (MYM) broth. The aims were to evaluate the effectiveness of the test for measuring microbial growth and to study the effects of the test conditions (incubation time, incubation temperature and medium pH) on triphenyl formazan (TF) yield. The results showed that the TF yield was highly correlated to the optical density. The highest TF yield was observed at a pH of 6 at all incubation times and temperature. Lower TF yields were obtained at higher temperature (40 and 50oC) compared to those obtained at lower temperatures (22 and 30oC). The difference between the yields obtained at 22oC and 30oC were not significant. The differences between incubation time were also not significant. The recommended test conditions are an incubation time of 1 hour at a temperature of 30oC and a pH of 6 followed by three extractions using methanol.

It has been documented in prior studies that Chinese companies issuing both A-shares （domestic investors） and B-shares （foreign investors） often report earnings numbers under Chinese GAAP that are notably different from their IAS *** has been cited as evidence of failure in China’s attempts to harmonize their accounting standards with IAS,and that Chinese GAAP earnings numbers may have been ’managed’ upwards because Chinese managers’ incentives are primarily built around Chinese GAAP data,rather then IAS data （***,et al.2002）.However,since the start of 2001,China’s new Accounting Standard for Business Enterprises has been in effect: we provide some of the first empirical evidence on how this new regulation has impacted the China-IAS earnings *** employ both graphical analysis and multivariate regression models to a set of 198 firm-year observations for B-share firms on the Shanghai Stock Exchange for the period *** find evidence of the earnings gap being notably reduced in the years immediately following 2001,consistent with improved de facto *** also find evidence that the earnings gap is significantly greater in those cases where we may expect there to be incentives for earnings management,following US evidence （Burgstahler and Dichev,1997）.That is to say,having controlled for a declining time trend in the mean earnings gap during 2000-03 we find evidence of a significant positive bias in Chinese GAAP earnings in cases where firms make ’manageable’ losses or where earnings are declining.

Objective: To report our experience of attempting a randomised controlled trial of an intensive lifestyle intervention for early type 2 diabetes delivered in a residential setting. Methods: We established a trial requiring 84 participants (46 standard care and 38 intervention) to detect a 1% difference in HbA1c between intervention and control groups at 12 months, allowing for attrition. Ethics approval was obtained from Monash University. Results: The study was abandoned after five months of consistent promotion due to recruitment failure (four subjects recruited). Conclusion: It appears to be difficult for patients with diabetes to commit to a live-in period of education regarding lifestyle modification as a means of treating the illness. We recommend better education of patients and their doctors about the potential health benefits of lifestyle change to manage type 2 diabetes, and further research into novel methods of delivering lifestyle advice which are both effective and sustainable.

旨在修订个人球形职业兴趣量表简版（PGI-S）以形成中国版本（PGI-SC）,用于我国的职业兴趣研究及职业测评。首先,采用个人球形职业兴趣量表简版（PGI-S）中文版本,对197名大学生和高中生进行预测验,对获得的预测验数据进行统计分析,以此进行量表的修订,获得正式施测问卷。然后,对全国的高中生（一~三年级）和在校大学生（一~四年级）发放正式施测问卷3300份,回收问卷2950份,获得有效问卷2567。采用RANDALL软件和SPSS 15.0进行统计分析,获得了个人球形职业兴趣量表简版中国版（PGI-SC）。该量表包含八种基本职业兴趣类型,后者构成八边形的环形结构,数据与结构拟合,CI在0.72~0.86之间（p〈0.01）。该量表的Cronbachα系数在0.73~0.94之间（p〈0.01）,重测信度在0.54~0.78之间（p〈0.01）,与Holland职业兴趣类型关联系数在0.27~0.6之间（p〈0.05）。因此,PGI-S中文修订版（PGI-SC）显示出良好的信效度,可作为我国职业兴趣的测量工具。
