Corn Leaf Water Retention as Affected by OrganicFertilizations and Effective Microbes Applications

Effects of organic fertilizers and effective microbes on leaf water retention of sweet corn (Zea mays L. ***-Bantam) were studied. Sweet corns were grown with organic or chemical fertilizers with or without effective microbes (EM). A water retention curve was obtained by drying the excised leaves under a light of 500 μmol (m2·s)-1. The curve shows two distinct phases. The initial steep slope indicates the water loss speed by stomatal transpiration (Est) and the gentle slope of the second phase indicates water loss speed by cuticular transpiration (Ecu). Both Est and Ecu were lower for leaves of plants grown with organic materials than for those with chemical fertilizers. Addition of EM to both organic and chemical fertilizers decreased Est but showed no effect on Ecu. The water retention ability of the excised leaves was proportional to photosynthetic maintenance ability under soil water deficit conditions as well as the solute concentration in leaves. The results suggested that organic fertilization and EM application increased water stress resistance both under in situ conditions and in excised leaves of sweet corn plants.

Beam-ion losses induced by fast-ion-driven toroidal Alfven eigenmodes （TAE） were measured with a scintillator-based lost fast-ion probe （SLIP） in the large helical device （LHD）. The SLIP gave simultaneously the energy E and the pitch angle X=arccos（v///v） distribution of the lost fast ions. The loss fluxes were investigated for three typical magnetic configurations of Rax-vac=3.60 m, 3.75 m. and 3.90 m, where Rax-vac is the magnetic axis position of the vacuum field. Dominant losses induced by TAEs in these three configurations were observed in the E/X regions of 50-190 keV/40°, 40-170 keV/25°, and 30-190 keV/30°, respectively. Lost-ion fluxes induced by TAEs depend clearly on the amplitude of TAE magnetic fluctuations, Rax-vac and the toroidal field strength Bt. The increment of the loss fluxes has the dependence of （bTAE/Bt）s. The power s increases from s = 1 to 3 with the increase of the magnetic axis position in finite beta plasmas.

Metabolomic Screening Applied to Rice FOX Arabidopsis Lines Leads to the Identification of a Gene-Changing Nitrogen Metabolism

Plant metabolomics developed as a powerful tool to examine gene functions and to gain deeper insight into the physiology of the plant cell. In this study, we screened Arabidopsis lines overexpressing rice full-length （FL） cDNAs （rice FOX Arabidopsis lines） using a gas chromatography-time-of-flight mass spectrometry （GC-TOF/MS）-based technique to identify rice genes that caused metabolic changes. This screening system allows fast and reliable identification of candi- date lines showing altered metabolite profiles. We performed metabolomic and transcriptomic analysis of a rice FOX Ara- bidopsis line that harbored the FL cDNA of the rice ortholog of the Lateral Organ Boundaries （LOB） Domain （LBD）/ Asymmetric Leaves2-1ike （ASL） gene of Arabidopsis, At-LBD37/ASL39. The investigated rice FOX Arabidopsis line showed prominent changes in the levels of metabolites related to nitrogen metabolism. The transcriptomic data as well as the results from the metabolite analysis of the Arabidopsis At-LBD37/ASL39-overexpressor plants were consistent with these findings. Furthermore, the metabolomic and transcriptomic analysis of the Os-LBD37/ASL39-overexpressing rice plants indicated that Os-LBD37/ASL39 is associated with processes related to nitrogen metabolism in rice. Thus, the combination of a metabolomics-based screening method and a gain-of-function approach is useful for rapid characterization of novel genes in both Arabidopsis and rice.

Subacute Toxicological Analysis of Excessive <i>Staphylococcus epidermidis</i>Administration in Mice

We performed subacute toxicological analysis of mice that received repeated oral administration of excess Staphylococcus epidermidis every day for 4 weeks. No mouse died and no abnormalities of specific internal organs were induced by excessive S. epidermidis administration. Furthermore, acute toxicity analysis following oral and intraperitoneal administration of excess S. epidermidis showed no toxicity in the test mice.

Prevalence and Antimicrobial Susceptibility Pattern of Streptococcus pneumoniae at General Hospital in the Central Region of Japan from December 2013 to February 2014

Streptococcus pneumonia infection is important cause of morbidity and mortality. This study was conducted to find out the prevalence and antimicrobial susceptibility pattern of Streptococcus pneumoniae isolates at general hospitalin the central region of Japan from December 2013 to February 2014. Streptococcus pneumoniae was identified by standard laboratory procedure. Antimicrobial susceptibility testing was performed by micro dilution assay according to CLSI recommendation. One hundred fifty-three Streptococcus pneumoniae were isolated among which 80 (52.2%) were males and 73 (47.8%) were females. Nasal discharge (134%/87.6%) contributed more than other biological materials. The age incidence of (0 - 1) years, (1 - 10) years, (11 - 40) years, (41 - 60) years and >60 years age groups were 26 (17.0%), 110 (71.9%), 3 (2.0%), 10 (6.5%), and 4 (2.6%) respectively. Positive samples were received mostly from the pediatrics (137%/89.5%), respiratory medicine (12%/7.8%) and lowest from gastroenterology (1%/0.6%) and neurology (1/ 0.6%) department. Vancomycin and rifampicin were the most active antibiotics with 100% susceptibility rates. The next best were levofloxacin, penicillin G and ceftriaxone. Our study revealed that 82 Streptococcus pneumonia isolates had multidrug resistant ability (53.6%). Streptococcus pneumoniae infection spreads among community easily and inappropriate use of antibiotics contributes to their resistance. Continuous antimicrobial susceptible surveys are essential to guide policy on the adequate use of antibiotics to reduce the morbidity and mortality and reduce the emergency of antimicrobial resistance.

LicT Modulates Biofilm Formation of Streptococcus pyogenes

Streptococcus pyogenes frequently causes purulent infections in humans. Biofilm formation is an important virulence property of S. pyogenes because of decreased susceptibility of bacteria to antibiotic treatment. Biofilm is composed of various types of matrix including glycocalyx which is an important exocellular matrix material related to bacterial sugar metabolism. A putative antiterminator protein, LicT (Spy0571), is one of the components of the glucose-independent β-gluco-side-specific phosphotransferase system (PTS). Although the PTS, a carbohydrate metabolic system, may play a role in biofilm formation, the relationship between LicT and biofilm formation has not yet been elucidated. Here, we evaluated whether LicT affected biofilm formation in modified chemically defined medium (CDMM) supplemented with glucose or β-glucoside:salicin. We created licT- and licT-complemented mutant strains from S. pyogenes 1529. Although the licT mutant strain tended to have higher growth rate than wild-typestrain in CDMM with glucose, it had a significant lower growth rate than the wild-type strain in CDMM with salicin. In addition, the licT mutant exhibited lower biofilm formation in CDMM containing salicin than the wild-type strain by 96 well plate analysis and confocal laser scanning microscopic analysis. Our results suggest that LicT plays an important role in biofilm formation of S. pyogenes.

Clinical Characteristics of Vaginal Discharge Associated Streptococcus pyogenes at General Japanese Hospital

Streptococcus pyogenes (S. pyogenes) is popular pathogen caused various infectious disease. This study was conducted to find out the recent clinical characteristics of S. pyogenes isolates from vaginal discharge at Japanese hospital. S. pyogenes was identified by standard laboratory procedure. Antimicrobial susceptibility testing was performed by microdilution assay according to CLSI recommendation. Total 61 S. pyogenes were isolated from vaginal discharge. The major age incidence among 0 - 10 years age group was 14 (23%), among 21 - 30 years age group, 7 (11.5%), among 31 - 40 years age group, 13 (21.3%), among 51 - 60 years age group, 8 (13.1%), and among 61 - 70 years age group, 11 (18%). The numbers of ciprofloxacin and clarithromycin no-susceptible S. pyogenes from 0 - 10, 31 - 40, and 61-70 years age group were higher than those from other groups. The numbers of clindamycin and minocycline no susceptible S. pyogenes from 0 - 10, and 21 - 40 years age group were higher than those from other group. Furthermore, our study revealed that clarithromycin was completely ineffective in 21 - 30 years age group (p S. pyogenes because the trend of infectious disease epidemiology is always changing drastically.

Stimulation of Root Growth Induced by Aluminum in <i>Quercus serrata</i>Thunb. Is Related to Activity of Nitrate Reductase and Maintenance of IAA Concentration in Roots

Aluminum (Al) is the most abundant metal in the earth’s crust. Excess Al3+ released by soil acidification in soil solution is thought to be a growth limiting factor to many cultivated plant species, but it has been reported to stimulate plant growth in some crop and tree species in certain concentration of Al3+. Previously, we had reported that Al treatment enhanced root development, uptake from growth media and in vivo nitrate reductase (NR) activity of roots. NR is one of the key enzymes in nitrogen metabolism and acts at the first step of nitrate assimilation in plants. In this study, we investigated the process of Al-induced root development in an early stage, focusing on the change in in vitro NR activity, and indole-3-acetic acid (IAA) and cytokinins concentration in roots of Quercus serrata seedlings, which were treated for 1 h with Al or Ca. In Al-treated roots, NR activity increased and IAA concentration was maintained at the same level as pretreatment, and indole-3-acetyl-L-aspartic acid (IA-Asp), which is a metabolic intermediate of IAA degradation, was not detected in roots. In Ca-treated roots, NR activity increased, but IAA concentration decreased as IA-Asp concentration increased. Thus, the maintenance of IAA concentration in Al-treated roots seems to result from suppression in the process of IAA decomposition. Al treatment increased the length and number of second lateral roots but Ca treatment did not. We concluded that root development induced by Al in the early stage was related to NR activity and maintenance of IAA concentration.

探讨阻塞性睡眠呼吸暂停（Ｏｂｓｔｒｕｃｔｉｖｅｓｌｅｅｐａｐｎｅａ，ＯＳＡ）患者上气道狭窄的程度、头姿改变情况，以及这些形态学指标与病理生理学参数之间的关系。研究对象为８６例男性ＯＳＡ患者及３７名健康男性。经整夜多导睡眠仪检查诊断或排除ＯＳＡ；采用数字化技术摄取受试者头颅侧位片，并用ＮＩＨＩｍａｇｅ软件测定上气道口径、面积及头姿指标共１５项。ＯＳＡ患者各水平上气道前后径均显著小于对照组；经年龄及身高体重指数校正后，上述结果不变。重症ＯＳＡ患者呈显著头颅前倾。头姿变化指标与呼吸暂停指数及多个水平的气道前后径呈显著相关。亚洲人种ＯＳＡ患者存在明显及广泛的上气道狭窄；随ＯＳＡ程度加重及上气道阻塞程度加重。
