Synapses utilize a network of proteins, the cytomatrix at the active zone(CAZ) to tightly regulate synaptic strength and SV release kinetics to match the changing needs of the neuronal circuit. The CAST/ELKS family, central components of the CAZ, have been implicated as key proteins that regulate synaptic function. However, their functional roles in the regulation of neuronal circuits remain unresolved. Using a newly created CAST KO/ELKS conditional KO mouse line which overcomes the embryonic lethality associated with constitutive KO of ELKS proteins, we analyzed the role of the CAST/ELKS protein family in regulation of presynaptic function to match the needs of the neuronal circuit. To do so, we utilized the calyx of Held, a large excitatory presynaptic terminal located in the auditory brainstem and the dominant input for driving postsynaptic spiking in the Medial Nucleus of the trapezoid body(MNTB) to help encode sound information. We carried out electrophysiological and ultrastructural analysis in both the immature(prehearing postnatal day,(P9-11) and functionally mature(hearing, P16-P21) calyx to elucidate the roles of the CAST/ELKS proteins. Here we identify for the first time, that the CAST/ELKS protein family regulates voltage gated Ca channel abundance in the presynaptic terminal. In addition, loss of CAST/ELKS proteins did not affect basal synaptic transmission. However, loss of CAST/ELKS resulted in increased release probability while decreasing the size of the RRP available for action potential mediated release in the functionally mature calyx. Therefore, we conclude that the CAST/ELKS proteins are critical molecules that differentially control key parameters of synaptic function in response to the changing neuronal circuit demand.

Influence of combustion-originated dioxins in atmospheric deposition on water quality of an urban river in Japan

Bulk（wet and dry） deposition samples were collected in Saitama Prefecture, Japan throughout a year（February 8, 2012 to February 7, 2013） to estimate the influence of dioxins emitting from waste incinerators on river water quality. The annual deposition flux of dioxins was 3.3 ng-toxic equivalent（TEQ）/m^2/year. Source identification using indicative congeners estimated that 82% of dioxin TEQ in the bulk deposition（2.7 ng-TEQ/m^2/year）was combustion-originated, indicating that most of the dioxins in the deposition were derived from waste incinerators. In Saitama prefecture the annual flux of combustionoriginated dioxins in depositions was apparently consistent with that of dioxin emission into the air from waste incinerators. The TEQ of combustion-originated dioxins in the deposition per rainfall was 2.4 pg-TEQ/L on annual average, exceeding the environmental quality standard（EQS） for water in Japan of 1 pg-TEQ/L. This suggests there is a possibility that dioxins in atmospheric deposition have a significant influence on the water quality of urban rivers which rainwater directly flows into because of many paved areas in the *** influence of combustion-originated dioxin in the deposition on the water quality of Ayase River, an urban river heavily polluted with dioxins, was estimated at 0.29 pg-TEQ/L on annual average in 2015. It seems that dioxins in atmospheric deposition from waste incinerators have a significant influence on water quality of some urban rivers via rainwater though the dioxins in the ambient air have achieved the EQS for atmosphere at all monitoring sites in Japan.

Ultraviolet and visible upconversion luminescence of Tm(0.1)Yb(5):FOV oxyfluoride nanophase vitroceramics

The ultraviolet upconversion luminescence of Tm3+ ions sensitized by Yb3+ ions in oxyfluoride nanophase vitroceramics when excited by a 975 nm diode laser was studied. An ultraviolet upconversion luminescence line positioned at 363.6 nm was found. It was attributed to the fluorescence transition of 1D2→3H6 of Tm3+ ion. Sev- eral visible upconversion luminescence lines at 450.7 nm, (477.0 nm, 462.5 nm), 648.5 nm, (680.5 nm, 699.5 nm) and (777.2 nm, 800.7 nm) were also found, which result respectively from the fluorescence transitions of 1D2→3F4, 1G4→3H6, 1G4→3F4, 3F3→3H6 and 3H4→3H6 of Tm3+ ion. The careful measurement and analysis of the variation of upconversion luminescence intensity F as a function of the 975 nm pumping laser power P prove that the upconversion luminescence of 1D2 state is partly a five-photon upconversion luminescence, and the upconversion lumines- cence of 1G4 state and 3H4 state are respectively the three-photon and two-photon upconversion luminescence. The theoretical analysis suggested that the upcon- version mechanism of the 363.6 nm 1D2→3H6 upconversion luminescence is partly the cross energy transfer of {3H4(Tm3+)→3F4(Tm3+), 1G4(Tm3+)→1D2(Tm3+)} and {1G4(Tm3+)→3F4(Tm3+), 3H4(Tm3+)→1D2(Tm3+)} between Tm3+ ions. In addition, the upconversion luminescence of 1G4 and 3H4 state results respectively from the se- quential energy transfer {2F5/2(Yb3+)→2F7/2(Yb3+), 3H4(Tm3+)→1G4(Tm3+)} and {2F5/2(Yb3+) →2F7/2(Yb3+), 3F4(Tm3+)→3F2(Tm3+)} from Yb3+ ions to Tm3+ ions.

The Notch-Hes pathway in mammalian neural development

A wide variety of neurons and gIial cells differentiatefrom common precursor cells in the developing nervoussystem. During this process, Notch-mediated cell-cell in-teraction is essential for maintenance of dividing cells andsubsequellt generation of cell type diversity. Activation ofNotch inhibits cellular differentiation, and abnormality ofthe Notch pathway leads to premature neuronal differenti-ation, the lack of some cell types, and severe defects of tis-sue morphogenesis. Recent data demonstrate that Notchfails to inhibit cellular differentiation in the absence of thebHLH genes Hes1 and He5, which functionally afitago-nize the neuronal bHLH genes such as theh1- These re-sults indicate that the two Hes genes are essential effecorsfor the Notch pathway and that neuronal differentiation iscontrolled by the pathway "Notch-+He1/Hs5,Mish1".

Helicobacter pylori infection and gastropathy:A comparison between Indonesian and Japanese patients

AIM： To compare the effects of He/icobacter py/ori （Hpylon） infection on gastropathy between Indonesian and Japanese patients. METHODS： Biopsy specimens were obtained during upper gastrointestinal endoscopy from 167 subjects （125 Indonesians and 42 Japanese） with uninvestigated symptoms of dyspepsia. The specimens were analyzed for the presence of H pylori using urease analysis, histopathology, and cell culture. The grade and activity of gastritis was assessed using the updated Sydney system. RESULTS： The percentages of Indonesian and Japanese patients who were H py/ori-positive at the antrum or body of the stomach were similar （68% and 59.5%, respectively; P = 0.316）. Of those who were Hpylori-positive, more Japanese patients than Indonesian patients had high levels of polymorphonuclear cells （P = 0.001）, mononuclear cells （P = 0.013）, glandular atrophy （P = 0.000）, and intestinal metaplasia （P = 0.011） in both the antrum and body of the stomach. CONCLUSION： The grade of gastritis and prevalence of mucosal atrophy and intestinal metaplasia were higher in Japanese patients. The difference between Indonesian and Japanese patients was significant.

Improved anti-Stokes energy transfer between rare earth ions in Er(0.5)Yb(9.5):FOV oxyfluoride vitroceramics explains the strong color reversal

The widely used energy transfer theory is a foundation of luminescence, in which the rates of Stokes and anti-Stokes processes have the same calculation formula. An improvement on the anti-Stokes energy transfer to explain the fluorescence intensity reversal between the red and green fluorescence of Er（0.5）Yb（9.5）：FOV is reported in the present article. The range of the intensity reversal Z was measured to be 877. Dynamic processes for 16 levels were simulated. A coefficient, the improvement factor of the intensity ratio of Stokes to anti-Stokes processes in quantum Raman theory compared to classical Raman theory, is introduced to successfully describe the anti-Stokes energy transfer. A new method to calculate the distance between the rare earth ions, which is critical for the energy transfer calculation, is proposed. The validity of these important improvements is also proved by experiment.
