Effect of Pregabalin and Gabapentin on Nociceptive Behaviors Induced by Spinal Nerve Ligation

Pain is defined as an unpleasant sensory and emotional experience, associated with actual or potential tissue damage. According to its neurobiological mechanism, pain is classified into nociceptive, inflammatory, dysfunctional, and neuropathic. Neuropathic pain (NP) is caused by a lesion or disease of the somatosensory nervous system. Both pregabalin and gabapentin are pharmaceuticals used as validation drugs in experimental models of NP. Pregabalin was shown to produce significant antihyperalgesic and antiallodynic effects. Gabapentin is used as a reference compound for new analgesics and reduces tactile allodynia in rats. The aim of this work is to evaluate pregabalin and gabapentin effects on nociceptive behaviors induced by spinal nerve ligation (SNL). Female Wistar rats of 140 - 160 g were used, divided into five groups: Naive, SHAM, SNL rats treated with saline solution, SNL rats treated with pregabalin 30 mg/kg p.o., SNL rats treated with gabapentin 300 mg/kg p.o. Nociceptive behaviors were determined by the up and down method. In the establishment of SNL-induced allodynic behavior, a reduction in paw withdrawal threshold was observed in the time course, which was present from day 1 and it was maintained for 28 days post-ligation. With the administration of pregabalin and gabapentin, anti-allodynic behavior was observed in the time course and in the areas under the curve (AUC) of the time course of anti-allodynic behavior, significant difference was observed between pregabalin, and gabapentin groups compared to vehicle with a value of p jected to SNL.

Background: Despite a rapid evolution in the treatment of Hepatitis C, response to therapy among different racial and ethnic groups is poorly characterized. Study: Three hundred and thirty HCV infected patients naive to previous therapy received induction therapy followed by every other day dosing with consensus interferon. Greater than 30% of treated patients were not white, allowing comparison of response among different races/ethnicities and genotypes. Results: An overall sustained virologic response (SVR) was achieved in 24% of white, 12% of Hispanic, and 4% of AA patients (P = 0.003 white vs. non-white). 15% of white and 13% of Hispanic Genotype 1 patients achieved an SVR; 2% of AA patients achieved an SVR(P = 0.001 AA vs. non AA). Surprisingly, an SVR of 50% and 40% was achieved by AA and White Genotype 2 patients, compared with 10% in Hispanic patients (P = 0.001). Conclusion:Significant differences in response rates to induction therapy followed by every other day dosing with consensus. Interferon was observed when comparing white to non-white patients, particularly when comparing response rates by genotype. These observations reinforce the requirement that prospective studies that enroll a significant percentage of non-whites are needed to adequately characterize response rates to anti-HCV directed therapy.

Characterization of Topsoil Samples and Analysis of the Distribution of Heavy Metals in Parral Chihuahua, Mexico

During the 17th Century, Parral City in the Chihuahua State in Mexico was one of the most important miner zones in the whole world. After more than three centuries important amounts of residuals, known as tailings, with high contents of heavy metals were generated. These residuals are a potential risk for the environment and human health. Given to that, it decided to carry out the analysis of the concentrations of cadmium, lead, chromium, zinc and arsenics, in topsoil samples susceptible of airborne transport and accumulation in risky zones （populated areas）. A 120 kin： area was selected, this include Parral City and its surroundings. From this area 30 samples were obtained. For this purpose, Atomic Absorption Spectroscopy technique was used, expecting high concentrations of heavy metals, above the permit limits, since several studies carried out in the same region, as in San Francisco del Oro Chihuahua, show that the concentrations of all the elements sampled, in topsoil, were above the limits. The analysis in the space distribution of the heavy metal detected will allow us to set the points with the highest susceptibility to the accumulation of those pollutants and to propose mitigation measures and control.

Canine Cutaneous Mast Cell Tumor: Biologic Behavior and Its Correlation with Prognostic Indicators

Cutaneous mast cell tumor (MCT) shows a variable biological behavior in dogs and may present either as solitary masse that can be treated and cured with surgical removal or as a systemic metastatic and fatal disease. Histological grade, KIT pattern and proliferative index are typically prognostic factors in MCTs. In the present study, we have investigated correlation between clinical data (breed, age, gender, tumour location, tumor size, time before surgery, number of tumours, occurrence of metastasis and recurrence), cellular proliferation (Ki-67, mitotic index), intratumoural microvessel density (IMVD) and apoptotic index with the histological grade and KIT pattern. 28 tumors, from 20 dogs with cutaneous MCT were evaluated. There was association between histological grade, IMVD, tumor ulceration and number of tumors. A significant increase of Ki-67 expression and mitotic index was observed in MCTs with cytoplasmic KIT staining pattern. Patnaik histological grade system was related to mitotic index. Histological grade in canine cutaneous MCT should not be assessed as the only prognostic factor, but associated with KIT pattern, IMDV, cellular proliferation, presence of tumour ulceration, number of tumours, recurrences and metastases.

Enhancement of the Photodynamic Therapy Effect on Streptococcus Mutans Biofilm

Biofilm is a community of bacteria, less susceptible to traditions treatments. Although photodynamic therapy （PDT） is a very effective way to microorganism inactivation, in biofilm it is not as efficient as it is in planktonic bacteria cultures. The increment of an element to increase the effectiveness of PDT was our aim. Therefore, this in vitro study evaluates the susceptibility ofa biofilm formed by Streptococcus mutans on metallic surface of orthodontic accessories under the application of PDT with a surfactant. Samples obtained from blades of orthodontic bands （NiCr）, where used as adhesion surface for the biofilm. They were treated with 1 mg/ml of curcumin, with 0.1% of sodium dodecyl sulfate and exposed to 30 j/cm^2 of light （455 nm）. Eight experimental groups were studied, including the positive and negative controls. The results show that the group with PDT and surfactant had a significant decrease （p 〈 0.001） in viability. In this case, the reduction observed was of 5.6 log10 （CFU/ml） in comparison to the control group. We have shown that, even though the biofilm is very tough and complex structure, we are able to promote almost the complete inactivation ofS. mutans in systems similar to an orthodontic treated patient＇s mouth.

Chronic diarrhea and colitis are common in patients positive for human immunodeficiency virus (HIV) under highly active antiretroviral treatment (HAART). This prospective doubleblind study explores the effect of mesalamine vs. placebo in HIV-positive patients. Thirteen HIV-infected patients with noninfectious chronic diarrhea and >250CD4+ cells/mm3 were randomized to mesalamine (2.4 g/day; n = 9) or placebo (n = 4) for 6 weeks. Colonoscopy was performed at baseline and week 6, and biopsies were obtained to calculate the Biopsy Activity Index (BAI). Diarrhea was assessed at baseline and end of treatment using the Disease Activity Index (DAI). Patients and clinicians completed Patient Global Improvement index (PGI) and Clinical Global Improvement index (CGI) at weeks 2 and 6. Comparisons at week 6 were statistically significant between mesalamine and placebo groups for BAI (P = 0.03), DAI (P = 0.007), PGI (P = 0.008), and CGI (P = 0.008). Furthermore, major improvements were documented in the mesalamine group at week 6 compared to baseline for all variables, whereas the placebo group did not have any. Mesalamine was effective for treatment of chronic diarrhea and moderate nonspecific colitis in HIV patients.

Study of the second harmonic emission of glycine-sodium nitrate crystals at different pH

This work shows an optical and structural study of glycine sodium nitrate crystals. This study was supported with the respective X ray diffracttion and Second-Harmonic Generation signal detection by using a little variant to the Kurtz- Perry method. The goal of this work is to obtain the right pH that modifies the charge of glycine sodium nitrate system in order to obtain the best second harmonic emission. Furthermore with the change on the charge on the aminoacid, it is observed how it modifies the optical properties in the glycine sodium nitrate complex.
