Lung cancer,highly prevalent and the leading cause of cancer-related death globally,persists as a significant challenge due to the lack of definitive tumor markers for early diagnosis and personalized therapeutic ***,extracellular vesicles(EVs),functioning as natural carriers for intercellular communication,have received increasing attention due to their ability to traverse biological barriers and deliver diverse biological cargoes,including cytosolic proteins,cell surface proteins,microRNA,lncRNA,circRNA,DNA,and *** are increasingly recognized as a valuable resource for non-invasive liquid biopsy,as well as drug delivery platforms,and anticancer vaccines for precision medicine in lung ***,given the diagnostic and therapeutic potential of tumor-associated EVs for lung cancer,we discuss this topic from a translational *** delve into the specific roles that EVs play in lung cancer carcinogenesis and offer a particular perspective on how advanced engineering technologies can overcome the current challenges and expedite and/or enhance the translation of EVs from laboratory research to clinical settings.

An LC-MS/MS method for simultaneous analysis of the cystic fibrosis therapeutic drugs colistin,ivacaftor and ciprofloxacin

Inhaled antibiotics such as colistin and ciprofloxacin are increasingly used to treat bacterial lung infections in cystic fibrosis *** this study,we established and validated a new HPLC-MS/MS method that could simultaneously detect drug concentrations of ciprofloxacin,colistin and ivacaftor in rat plasma,human epithelial cell lysate,cell culture medium,and drug transport *** aliquot of200 μL drug-containing rat plasma or cell culture medium was treated with 600 μL of extraction solution(acetonitrile containing 0.1% formic acid and 0.2% trifluoroacetic acid(TFA)).The addition of 0.2% TFA helped to break the drug-protein ***,the addition of 0.1% formic acid to the transport medium and cell lysate samples could significantly improve the response and *** vortexing and centrifuging,the sample components were analyzed by HPLC-MS/*** multiple reaction monitoring mode was used to detect the following transitions:585.5-101.1(colistin A),578.5-101.1(colistin B),393.2-337.2(ivacaftor),332.2-314.2(ciprofloxacin),602.3-101.1(polymyxin B1 as internal standard(IS)) and 595.4-101.1(polymyxin B2 as IS).The running time of a single sample was only 6 min,making this a time-efficient *** correlations were found for colistin A at 0.029-5.82 μg/m L,colistin B at 0.016-3.14 μg/m L,ivacaftor at 0.05-10.0 μg/m L,and ciprofloxacin at 0.043-8.58 μg/m ***,precision,and stability of the method were within the acceptable *** method would be highly useful for research on cytotoxicity,animal pharmacokinetics,and in vitro drug delivery.

Mayall Ⅱ = G1 is one of the most luminous globular clusters （GCs） in M31. Here, we determine its age and mass by comparing multicolor photometry with theoretical stellar population synthesis models. Based on far- and near-ultraviolet GALEx photometry, broad-band UBVRI, and infrared JHKs 2MASS data, we construct the most extensive spectral energy distribution of G 1 to date, spanning the wavelength range from 1538 to 20 000A. A quantitative comparison with a variety of simple stellar population （SSP） models yields a mean age which is consistent with G1 being among the oldest building blocks of M31 and having formed within ～1.7 Gyr after the Big Bang. Irrespective of the SSP model or stellar initial mass function adopted, the resulting mass estimates （of order 10^7M⊙） indicate that GI is one of the most massive GCs in the Local Group. However, we speculate that the cluster＇s exceptionally high mass suggests that it may not be a genuine GC. Our results also suggest that G1 may contain, on average, （1.65±0.63） × 10^2L⊙ far-ultraviolet-bright, hot, extreme horizontal-branch stars, depending on the adopted SSP model. In addition, we demonstrate that extensive multi-passband photometry coupled with SSP analysis enables one to obtain age estimates for old SSPs that have similar accuracies as those from integrated spectroscopy or resolved stellar photometry, provided that some of the free parameters can be constrained independently.

Reptile sex determination is attracting much attention because the great diversity of sex-determination and dosage compensation mechanisms permits us to approach fundamental questions about mechanisms of sex chromosome turnover. Recent studies have made significant progress in better understanding diversity and conservation of reptile sex chromosomes, with however no reptile master sex determination genes identified. Here we describe an integrated genomics and cytogenetics pipeline,combining probes generated from the microdissected sex chromosomes with transcriptome and genome sequencing to explore the sex chromosome diversity in non-model Australian reptiles. We tested our pipeline on a turtle, two species of geckos, and a monitor lizard. Genes identified on sex chromosomes were compared to the chicken genome to identify homologous regions among the four species. We identified candidate sex determining genes within these regions, including conserved vertebrate sex-determining genes pdgfa,pdgfra amh and wt1, and demonstrated their testis or ovary-specific expression. All four species showed gene-by-gene rather than chromosome-wide dosage compensation. Our results imply that reptile sex chromosomes originated by independent acquisition of sex-determining genes on different autosomes,as well as translocations between different ancestral macro-and *** discuss the evolutionary drivers of the slow differentiation and turnover of reptile sex chromosomes.

When tony Blair and New Labour came to office in Britain late in the 20th century,John Atkinson Hobson( 1858-1940) was rediscovered and republished. In spite of his prolific work( over 50 books),he had sunk into oblivion. Yet John Maynard Keynes paid tribute to him( T he General T heory of Employment,Interest and Money,1936) and recognized his debt to this"heretic"of economics. Hobson's contribution to the history of economic thought is a major one,thanks to his stimulating analysis of under-consumption. His book Imperialism,A Study( 1902) was Lenin's bedside reading. But Hobson was no socialist: he w as a"N ew Liberal",belonging to a group of theoreticians who thought that classical liberalism had to be changed radically if it was to have any future.
