The disbalance of LRP1 and SIRPα by psychological stress dampens the clearance of tumor cells by macrophages

The relationship between chronic psychological stress and tumorigenesis has been well defined in epidemiological studies;however, the underlying mechanism remains underexplored. In this study, we discovered that impaired macrophage phagocytosis contributed to the psychological stressevoked tumor susceptibility, and the stress hormone glucocorticoid(GC) was identified as a principal detrimental factor. Mechanistically, GC disturbed the balance of the "eat me" signal receptor(low-density lipoprotein receptor-related protein-1, LRP1) and the "don’t eat me" signal receptor(signal regulatory protein alpha, SIRPa). Further analysis revealed that GC led to a direct, glucocorticoid receptor(GR)-dependent trans-repression of LRP1 expression, and the repressed LRP1, in turn, resulted in the elevatedgene level of SIRPa by down-regulating mi RNA-4695-3 p. These data collectively demonstrate that stress induces the imbalance of the LRP1/SIRPa axis and entails the disturbance of tumor cell clearance by macrophages. Our findings provide the mechanistic insight into psychological stress-evoked tumor susceptibility and indicate that the balance of LRP1/SIRPa axis may serve as a potential therapeutic strategy for tumor treatment.

目的:探讨推拿配合针刺长强穴治疗小儿腹泻的临床疗效。方法:选择2014年1月-2019年1月我院门诊收治的84例腹泻患儿,采用随机数字表法将患儿分为A组(42例)与B组(42例),B组患儿接受蒙脱石散治疗,A组患者在此基础上予以推拿配合针刺长强穴治疗,比较两组疗效。结果:A组患儿治疗3 d、7 d时临床证候积分明显比B组患儿更低,腹泻次数明显比B组患儿更少(P <0.05)。A组患儿临床恢复指标均明显比B组患儿更优(P <0.05)。A组患儿治疗后的血D-乳酸、二胺氧化酶(Diamine Oxidase,DAO)水平及血清肿瘤坏死因子-α(Tumor Necrosis Factor,TNF-α)、白细胞介素(Interleukin,IL)-6、IL-10水平明显比B组患儿更低(P <0.05)。结论:推拿配合针刺长强穴治疗小儿腹泻疗效确切,方法简单,无不良反应,应进一步在临床上推广使用。

Dear Editor,Bread wheat(Triticum aestivum)is one of the most important food crops and provides approximately 20%of the food calories for human consumption.A 70%increase in wheat production is needed by 2050 to keep pace with the growing global population(International Wheat Genome Sequencing,2014).Developing superior cultivars is an efficient way to improve yield.Nevertheless,conventional breeding is time consuming,as more than eight generations are needed to develop new plant varieties.Using doubled haploid(DH)technology,homozygous lines can be produced in only two generations,dramatically accelerating the breeding process.In wheat,haploids can be obtained by cross pollination with corn pollen followed by embryo rescue(Laurie and Bennett,1988).In maize,haploids can be induced by haploid inducer lines derived from Stock6(liu et al.,2022).The cloning of two genes that control haploid induction(HI)in maize,MATL/ZmPLA1/NLD and ZmDMP,paved the way for DH breeding in more crop species(Jacquier et al.,2020).Further studies have shown that loss of function of TaPLAs triggers wheat HI with an efficiency of 5.88%to 31.6%(liu et al.,2020a,2020b);this would be a promising approach for establishing a new,simple,and more efficient DH breeding method in wheat.

目的观察针刺配合康复训练治疗小儿脑瘫的临床效果。方法选取2015年1月到2018年12月徐州市贾汪区人民医院收治的40例小儿脑瘫患儿,按照不同治疗方法分为对照组(单纯的康复训练治疗)和观察组(联合进行中医针刺治疗),各20例。比较患儿治疗前后的肢体功能GMFM量表和生活质量量表Barthel的评分水平。结果治疗前,患者的GMFM和Barthel的评分水平比较差异无统计学意义(P>0.05);治疗后,患者的GMFM和Barthel评分均升高,观察组患者的评分显著高于对照组(P<0.05)。结论针对小儿脑瘫患儿实施针刺配合康复训练能明显提升患儿的肢体功能,改善患儿的生活质量。

Early Stages of CeO_(2) Thin-film Nucleation and Growth with Photo Irradiation

In this paper,the early stages of nucleation and photoirradiation growth of CeO_(2) thin films have been studied.Cyclic voltammetry,chronoamperometry and scanning electron microscopy were used to analyze the nucleation process of CeO_(2) thin films deposited on the anode with photo irradiation.Experimental results show that the anodic deposition process with photo illumination is controlled by diffusion.Compared with the dark state,photo illumination mainly contributed to increase the current density of the three-dimensional nucleation process,because photo illumination is helpful to create active sites and accelerate the nucleation progress on the surface that a thin ceria film has been formed.Two-dimensional nucleation process mainly exists within the initial 2 s,and then only three-dimensional instantaneous nucleation process continues,which may be the main reason why the thickness of the CeO_(2) film can continue to grow with photo illumination but not in the dark state.Increasing the deposition overpotential can promote two-dimensional nucleation and growth rate,whilst when the potential exceeds 0.65 V,three-dimensional current density decreases.The limiting factor at that time may be the diffusion rate of cerium ions in the solution towards the electrode substrate.

胰腺导管癌（PDAC）是预后极差的恶性实体肿瘤，5年生存率仅为5％。尽管近20年来研究者对胰腺癌发病机制的了解有了很大提高，但胰腺癌临床治疗方面尚无突破性进展．因此亟待寻找有效的新疗法。KRAS是胰腺癌中突变最早且几率最大的基因，并且在癌症起始、进展和转移中发挥着显著的作用。这预示着KRAS是抗胰腺癌治疗的良好靶点。但是，目前临床上仍然缺乏特异性靶向KRAS的有效药物。直接靶向KRAS对于治疗胰腺癌具有良好前景。另外，通过影响KRAS翻译后修饰以阻断KRAS的细胞膜定位和活性、联合抑制RAS下游通路也是寻找以KRAS为靶标治疗胰腺癌的良好途径。

Although stress has been known to increase the susceptibility of pathogen infection,the underlying mechanism remains elusive.In this study,we reported that restraint stress dramatically enhanced the morbidity and mortality of mice infected with the influenza virus(H1N1)and obviously aggravated lung inflammation.Corticosterone(CORT),a main type of glucocorticoids in rodents,was secreted in the plasma of stressed mice.We further found that this stress hormone significantly boosted virus replication by restricting mitochondrial antiviral signaling(MAVS)protein-transduced IFN-βproduction without affecting its mRNA level,while the deficiency of MAVS abrogated stress/CORT-induced viral susceptibility in mice.Mechanistically,the effect of CORT was mediated by proteasome-dependent degradation of MAVS,thereby resulting in the impediment of MAVS-transduced IFN-βgeneration in vivo and in vitro.Furthermore,RNA-seq assay results indicated the involvement of Mitofusin 2(Mfn2)in this process.Gain-and loss-offunction experiments indicated that Mfn2 interacted with MAVS and recruited E3 ligase SYVN1 to promote the polyubiquitination of MAVS.Co-immunoprecipitation experiments clarified an interaction between any two regions of Mfn2(HR1),MAVS(C-terminal/TM)and SYVN1(TM).Collectively,our findings define the Mfn2-SYVN1 axis as a new signaling cascade for proteasome-dependent degradation of MAVS and a‘fine tuning’of antiviral innate immunity in response to influenza infection under stress.

Electroreduction of nitrate(NO_(3)-)to ammonia(NH_(3))is an environmentally friendly route for NH_(3)production,serving as an appealing alternative to the Haber-Bosch process.Recently,various noble metal-based electrocatalysts have been reported for electroreduction of NO_(3)-.However,the application of pure metal electrocatalysts is still limited by unsatisfactory performance,owing to the weak adsorption of nitrogen-containing intermediates on the surface of pure metal electrocatalysts.In this work,we report thiol ligand-modified Au nanoparticles as the effective electrocatalysts toward electroreduction of NO_(3)-.Specifically,three mercaptobenzoic acid(MBA)isomers,thiosalicylic acid(ortho-MBA),3-mercaptobenzoic acid(meta-MBA),and 4-mercaptobenzoic acid(para-MBA),were employed to modify the surface of the Au nanocatalyst.During the NO_(3)-electroreduction,para-MBA modified Au(denoted as para-Au/C)displayed the highest catalytic activity among these Au-based catalysts.At-1.0 V versus reversible hydrogen electrode(vs RHE),para-Au/C exhibited a partial current density for NH_(3)of 472.2 mA cm^(-2),which was 1.7 times that of the pristine Au catalyst.Meanwhile,the Faradaic efficiency(FE)for NH_(3)reached 98.7%at-1.0 V vs RHE for para-Au/C.The modification of para-MBA significantly improved the intrinsic activity of the Au/C catalyst,thus accelerating the kinetics of NO_(3)-reduction and giving rise to a high NH_(3)yield rate of para-Au/C.

矿井通风系统现状模拟中的双约束最优化风阻修正方法

针对使用网络解算方法进行矿井通风系统现状模拟时需要人工反复调整风阻、模拟结果与实测结果吻合度不高的问题,建立了矿井通风系统现状模拟的双约束最优化风阻修正模型;该最优化模型以分支实测风量、初始风阻为原始数据,以对初始风阻的修正量最小为目标函数,以回路风压平衡方程和节点流量平衡方程为等式约束条件,以井巷风阻的上、下限值为不等式约束条件;采用拉格朗日乘数法先求解等式约束条件下风阻修改量的最优解,再使用迭代法求得不等式约束条件下风阻修改量的最终解。开发了相应的软件,通过实例应用表明:针对不等式约束条件的迭代求解流程在应用实例中经过12次迭代后得到收敛结果,经本模型修正后的风阻能使网络解算的风量、风机风压与实测值完全吻合;仅考虑等式约束条件下的最优化模型有可能得到极小、甚至为负的无意义风阻值,考虑风阻限值的双约束模型可以将风阻限定在合理取值范围之内;开发的软件将风网解算与最优化方法同时用于矿井通风系统现状模拟,减少了人工反复调整风阻的繁琐操作。

从花溪区黔陶乡赵司村的茶树叶片上分离到2株拟盘多毛孢属真菌,菌株典型形态学特征为分生孢子具4~6根普遍分枝的顶生附属丝,这与近似种存在较明显的差别。同时,基于ITS,β-tubulin以及tef1的分子系统学研究结果表明,这2个菌株属于同一个种并聚集成一个相对独立的进化枝,此外,该分枝还与山茶拟盘多毛孢和顶生分枝拟盘多毛孢保持了比较近的遗传进化关系。综合形态学观察及DNA序列分析,所征集的真菌属于拟盘多毛孢属的一个新种,将该种命名为中国茶拟盘多毛孢(Pestalotiopsis camellia-sinensis)。
