Epstein–Barr virus(EBV)-associated lymphoproliferative diseases(EBV-LPDs)are common complications that occur after solid organ transplantation or allogeneic hematopoietic stem-cell transplantation(HSCT).However,their occurrence and treatment post-chimeric antigen receptor-modified T(CAR-T)cell therapy has not been *** patients had been diagnosed with EBV-positive aggressive B-cell lymphoma and experienced relapses after multiple lines of *** receiving CAR-T cell therapy in tandem with autologous HSCT,the patients achieved complete ***,with a median time of 38.5 months after CAR-T cell therapy,B-cell-derived EBV-LPDs were diagnosed,and they were relieved through the administration of immune checkpoint inhibitor or B-cell-depleting ***,our report suggests that EBV-LPDs may represent a long-term adverse event after CAR-T cell therapy,especially in patients who previously had EBV-positive disorders,and they can be resolved by immune normalization strategy or B-cell depleting therapy.

17种中草药体外抗炎作用研究

采用超声提取法利用水和乙醇分别获得17种中草药的水和醇提物,将其作用于脂多糖(LPS)诱导的RAW264.7细胞模型,通过检测提取物对细胞存活率和炎症介质一氧化氮(NO)、白介素-6(IL-6)、白介素-1β(1L-1β)、肿瘤坏死因子-α(TNF-α)的影响,综合评价其抗炎效果,并研究其作用机制。结果表明:黄花蒿、冷蒿、白头翁、龙蒿、白鲜果皮、多叶棘豆、斜茎黄耆种皮、灵芝菌棒、苍术根须、山韭醇提物和黄花蒿、冷蒿、广布野豌豆、东北珍珠梅、苍耳、茜草水提物无细胞毒性;通过评估无细胞毒性醇和水提物抗炎活性,最终证明冷蒿水提物具有更好的抗炎作用,可显著降低IKKβ、p65和IKKα表达量,揭示了其可通过调控NF-κB通路发挥抗炎作用。本试验结果为冷蒿防治畜禽细菌性肠炎的研发及应用提供理论依据。

芦台春酱香型白酒酿造过程中真菌菌群多样性及代谢分析

该研究采用高通量测序技术对天津芦台春酱香型白酒发酵过程中不同轮次出窖酒醅的真菌群落结构进行研究,并对其与理化因子、挥发性风味成分相关性分别进行典范对应分析(CCA)及偏最小二乘判别分析(PLS-DA)。结果表明,随着轮次的增加,真菌菌群的多样性升高,而丰度先升高后降低;酵母菌是白酒发酵过程中的主要真菌,但相对丰度逐步降低,在属水平上以伊萨酵母属(Issatchenkia)为主,其相对丰度由第一轮次的97.48%逐渐下降至第七轮次的3.53%。相关性分析结果表明,总酸、还原糖和酒精度是窖池发酵微生物演替的重要理化因子;伊萨酵母属(Issatchenkia)与乙酸乙酯、乙酸异戊酯、异戊醇等相关性较大;哈萨克斯坦酵母属(Kazachstania)、丝衣霉属(Byssochlamys)、罗萨氏菌属(Rasamsonia)、复膜孢酵母属(Saccharomycopsis)等与正己酸乙酯、庚酸乙酯、3-羟基丁酸乙酯、丁二酸二乙酯、苯乙酸乙酯、油酸乙酯、乙醇等相关性较大;而Leiothecium、未分类的酵母目(unclassified_o__Saccharomycetales)、Apiotrichum、Cutaneotrichosporon等则对壬酸乙酯、乙酸苯乙酯、正己醇、2,3-丁二醇、糠醇、糠醛等的形成起到重要作用。

靶向CD99的CAR-T细胞扩增优化研究

背景与目的:嵌合抗原受体T(chimeric antigen receptor T,CAR-T)细胞疗法对血液和淋巴系统肿瘤的治疗效果显著,但对实体瘤效果较差,这与靶点选择的因素有关。针对尤因肉瘤(Ewing sarcoma,ES),CD99可作为CAR-T细胞潜在的靶点。由于T细胞自身表达CD99蛋白,靶向CD99的CAR-T细胞存在体外扩增能力有限的问题。本研究旨在通过增加CD99敲低的短发夹RNA(short hairpin RNA,shRNA)、优化慢病毒转导的感染复数(multiplicity of infection,MOI)、筛选培养CAR-T细胞的培养基及培养容器,以获得CD99 CAR-T细胞制备的最优条件。方法:筛选shRNA序列,提高CD99 CAR-T细胞的扩增能力。采用不同的MOI、培养基和培养容器,分别检测在不同条件下CAR-T细胞的转导效率、细胞存活率、增殖能力、特异性杀伤能力及干扰素-γ(interferon-γ,IFN-γ)释放水平等,筛选出最优的细胞制备条件。结果:通过shRNA敲低得到的KO-CD99 CAR-T细胞扩增水平显著高于CD99 CAR-T细胞[(16.40±0.40)vs(6.33±1.53),P<0.01]。转导MOI为0.25~1.0、培养基为OptiVitro时细胞的扩增效果最优。在透气瓶中培养的CAR-T细胞扩增倍数显著高于在培养袋中培养的细胞[MOI=0.25:(50.23±3.32)vs(13.02±4.82);MOI=0.50:(49.96±0.83)vs(18.25±2.88);MOI=1.00:(48.27±5.08)vs(13.16±6.26);P<0.01],且细胞分型更优、特异性杀伤更高。结论:通过shRNA技术得到的KO-CD99 CAR-T细胞可实现稳定扩增。从扩增条件优化结果看,MOI为0.25~1.00,培养基为OptiVitro,培养容器为透气瓶的条件下,KO-CD99 CAR-T细胞可获得更优的扩增能力、更多比例的记忆T细胞,为后续开展CD99 CAR-T细胞治疗ES的临床试验奠定了坚实基础。

TP53 gene alteration confers inferior prognosis in refractory/relapse aggressive B-cell non-Hodgkin lymphoma(r/r B-NHL).From September 2016 to September 2020,257 r/r B-NHL patients were assessed for eligibility for two trials in our center,assessing anti-CD19 and anti-CD22 chimeric antigen receptor(CAR19/22)T-cell cocktail treatment alone or in combination with autologous stem cell transplantation(ASCT).TP53 alterations were screened in 123 enrolled patients and confirmed in ***19/22 T-cell administration resulted in best objective(ORR)and complete(CRR)response rate of 87.1%and 45.2%in patients with TP53 alterations.

RNA-dependent protein kinase(PKR) is crucial for the innate immune response,cell growth,proliferation,signal transduction and *** activation process of PKR has been studied for many years and is still under *** obtain new insight into the mechanism of PKR activation,we solved the crystal structure of a latent mutant of the PKR kinase domain(PKR-KD) in the apo form at a resolution of *** overall structure of PKR-KD is similar to previously reported *** analysis revealed a classical back-to-back dimer and a newly defined face-to-face *** ultracentrifugation experiments,electrostatic surface maps and the model of PKR-KD in complex with the eIF2 substrate all support that the face-to-face dimer is more reflective of PKR in *** results provide new information on PKR dimerization and its activation mechanism.

Plant cytochrome P450s play key roles in the diversification and functional modification of plant natural *** over 200,000 plant P450 gene sequences have been recorded,only seven crystalized P450 genes severely hampered the functional characterization,gene mining and engineering of important ***,we combined Rosetta homologous modeling and MD-based refinement to construct a high-resolution P450 structure prediction process(PCPCM),which was applied to 181 plant P450s with identified ***,we constructed a ligand docking process(PCPLD)that can be applied for plant P450s virtual screening.10 examples of virtual screening indicated the process can reduce about 80%screening space for next experimental ***,we constructed a plant P450 database(PCPD:http://***/),which includes the sequences,structures and functions of the 181 plant P450s,and a web service based on PCPCM and *** study not only developed methods for the P450-specific structure analysis,but also introduced a universal approach that can assist the mining and functional analysis of P450 enzymes.

维生素B_(12)是唯一含有金属离子的维生素,其合成途径尤为复杂。大肠杆菌工程菌是生成维生素B_(12)的新菌种。氢咕啉酸作为维生素B_(12)合成途径重要的稳定中间体,通过代谢工程提高大肠杆菌氢咕啉酸的产量具有重要意义。经过不同宿主的对比发现BW25113(DE3)是生产氢咕啉酸的合适宿主。利用CRISPR/Cas9介导的基因组编辑技术将氢咕啉酸生物合成基因在染色体上表达,氢咕啉酸产量提高12倍,达到6.38 mg/L。增加前体尿卟啉原Ⅲ供应,氢咕啉酸产量再次提高,达到11.61 mg/L。敲除ackA-pta对氢咕啉酸产量提高没有效果。经过引入运动发酵假单胞菌来源的Entner-Doudoroff途径提高还原型烟酰胺腺嘌呤二核苷酸磷酸(nicotinamide adenine dinucleotide phosphate,NADPH)水平,1株无抗菌株的氢咕啉酸产量提高到14.60 mg/L。该研究为大肠杆菌高效生产维生素B_(12)奠定了基础。

整合素integrinβ4调控VEGF诱导的骨髓间充质干细胞向内皮细胞分化

血管内皮生长因子(vascular endothelial growth factor,VEGF)可以诱导间充质干细胞(mesenchymal stem cells,MSCs)向内皮细胞(endothelial cells,ECs)分化,但分化机制尚不清楚.本研究鉴定了整合素integrin β4在VEGF诱导的MSCs向ECs分化过程中表达明显上调,同时利用亚硫酸氢盐测序法(EZ DNA Methylation)分析了5-aza-dc和VEGF处理MSCs后的integrin β4启动子Cp G岛发生部分的去甲基化.在分化过程中持续加入integrin β4的功能阻断性抗体,可显著降低内皮分化标志基因KDR(endothelial gene kinase insert domain containing receptor,KDR)、Flt-1的表达.以上结果表明,integrin β4可调控VEGF诱导的MSCs向ECs分化.
