Bioactive Hydrogels with Pro-coagulation Effect for Hemostasis

Hemostatic hydrogels are widely applied for wound management of damaged tissues,traumatic wounds,and surgical incisions.Some hydrogels composed of bioactive components,including fibrin and thrombin,showed great promise in the clinic due to their good pro-coagulation effect.With the expanding knowledge of cascade reaction of blood coagulation and emerging bioactive substances,massive bioactive hydrogels consisting of peptides,hemocoagulase,polyphosphate(polyP),etc.,have been developed as effective hemostatic materials.Based on the coagulation process and mechanism,we summarize the role of reported bioactive hydrogels in hemostasis in this review.We conclude the key points in the coagulation process,including activation of coagulation factors,fibrinogen polymerization,etc.,then discuss how to design bioactive hydrogels to accelerate coagulation targeted to these points.Finally,we conclude the progress and propose a perspective of bioactive hydrogels with a pro-coagulation effect for hemostasis.

Multiple myeloma(MM)remains a challenging hematologic malignancy despite advancements in chimeric antigen receptor T-cell(CAR-T)therapy.Current targets of CAR-T cells used in MM immunotherapy have limitations,with a subset of patients experiencing antigen loss resulting in relapse.Therefore,novel targets for enhancing CAR-T cell therapy in MM remain needed.Fc receptor-like 5(FCRL5)is a protein marker with considerably upregulated expression in MM and has emerged as a promising target for CAR-T cell therapeutic interventions,offering an alternative treatment for MM.To further explore this option,we designed FCRL5-directed CAR-T cells and assessed their cytotoxicity in vitro using a co-culture system and in vivo using MM cell-derived xenograft models,specifically focusing on MM with gain of chromosome 1q21.Given the challenges in CAR-T therapies arising from limited T cell persistence,our approach incorporates interleukin-15(IL-15),which enhances the functionality of central memory T(TCM)cells,into the design of FCRL5-directed CAR-T cells,to improve cytotoxicity and reduce T-cell dysfunction,thereby promoting greater CAR-T cell survival and efficacy.Both in vitro and xenograft models displayed that FCRL5 CAR-T cells incorporating IL-15 exhibited potent antitumor efficacy,effectively inhibiting the proliferation of MM cells and leading to remarkable tumor suppression.Our results highlight the capacity of FCRL5-specific CAR-T cells with the integration of IL-15 to improve the therapeutic potency,suggesting a potential novel immunotherapeutic strategy for MM treatment.

It is particularly important to comprehensively assess the biotoxicity variation of industrial wastewater along the treatment process for ensuring the water environment security.However,intensive studies on the biotoxicity reduction of industrial wastewater are still limited.In this study,the toxic organics removal and biotoxicity reduction of coal chemical wastewater(CCW)along a novel full-scale treatment process based on the pretreatment process-anaerobic process-biological enhanced(BE)process-anoxic/oxic(A/O)process-advanced treatment process was evaluated.This process performed great removal efficiency of COD,total phenol,NH_(4)^(+)-N and total nitrogen.And the biotoxicity variation along the treatment units was analyzed from the perspective of acute biotoxicity,genotixicity and oxidative damage.The results indicated that the effluent of pretreatment process presented relatively high acute biotoxicity to Tetrahymena thermophila.But the acute biotoxicity was significantly reduced in BE-A/O process.And the genotoxicity and oxidative damage to Tetrahymena thermophila were significantly decreased after advanced treatment.The polar organics in CCW were identified as the main biotoxicity contributors.Phenols were positively correlated with acute biotoxicity,while the nitrogenous heterocyclic compounds and polycyclic aromatic hydrocarbons were positively correlated with genotoxicity.Although the biotoxicity was effectively reduced in the novel full-scale treatment process,the effluent still performed potential biotoxicity,which need to be further explored in order to reduce environmental risk.

Real-world passive radiative cooling requires highly emissive,selective,and omnidirectional thermal emitters to maintain the radiative cooler at a certain temperature below the ambient temperature while maximizing the net cooling power.Despite various selective thermal emitters have been demonstrated,it is still challenging to achieve these conditions sim-ultaneously because of the extreme difficulty in controlling thermal emission of photonic structures in multidimension.Here we demonstrated hybrid polar dielectric metasurface thermal emitters with machine learning inverse design,en-abling a high emissivity of~0.92 within the atmospheric transparency window 8-13μm,a large spectral selectivity of~1.8 and a wide emission angle up to 80 degrees,simultaneously.This selective and omnidirectional thermal emitter has led to a new record of temperature reduction as large as~15.4°C under strong solar irradiation of~800 W/m2,signific-antly surpassing the state-of-the-art results.The designed structures also show great potential in tackling the urban heat island effect,with modelling results suggesting a large energy saving and deployment area reduction.This research will make significant impact on passive radiative cooling,thermal energy photonics and tackling global climate change.

Dispersion fuels,knowned for their excellent safety performance,are widely used in advanced reactors,such as hightemperature gas-cooled reactors.Compared with deterministic methods,the Monte Carlo method has more advantages in the geometric modeling of stochastic media.The explicit modeling method has high computational accuracy and high computational cost.The chord length sampling(CLS)method can improve computational efficiency by sampling the chord length during neutron transport using the matrix chord length?s probability density function.This study shows that the excluded-volume effect in realistic stochastic media can introduce certain deviations into the CLS.A chord length correction approach is proposed to obtain the chord length correction factor by developing the Particle code based on equivalent transmission probability.Through numerical analysis against reference solutions from explicit modeling in the RMC code,it was demonstrated that CLS with the proposed correction method provides good accuracy for addressing the excludedvolume effect in realistic infinite stochastic media.

The application of photothermal therapy(PTT)is greatly limited by the low accumulation of photothermal agents,uneven photothermal distribution,and heat endurance of cancer cells.Worse still,despite PTT enhances immunogenicity,the anti-tumor immune efficacy is still unsatisfactory due to the inefficient immunogenic cell death(ICD)induction and poor infiltration of immune cells.To solve the above problems of PTT,we developed hyaluronic acid(HA)modified hollow copper sulfide nanoparticles encapsulating diethyldithiocarbamate(DDTC)to construct a breast tumor targeting and near infrared(NIR)photo-responsive drug delivery system(D-HCuS@HA),which further combined with losartan to improve the accumulation and penetration in the tumor site.Upon irradiation,D-HCuS@HA realized enhanced PTT and released cytotoxic Cu(DDTC)_(2)to eliminate heat endurance tumor cells,thereby enhancing antitumor effect and inducing effective ICD.Moreover,the combination with losartan could remodel the tumor microenvironment,allowing more T cells to infiltrate into the tumor,and significantly inhibiting the occurrence and development of metastatic tumors.In vitro/vivo results revealed the great potential of D-HCuS@HA combined with losartan,which provides a new paradigm for anti-tumor and anti-metastases.

Coal chemical wastewater(CCW)is a type of refractory industrial wastewater,and its treatment has become the main bottleneck restricting the sustainable development of novel coal chemical industry.Biological treatment is considered as an economical,effective and environmentally friendly technology for CCW treatment.However,conventional biological process is difficult to achieve the efficient removal of refractory organics because of CCW with the characteristics of composition complexity and high toxicity.Therefore,seeking the novel enhancement strategy appears to be a favorable solution for enhancing biological treatment efficiency of CCW.This review focuses on presenting a comprehensive picture about the exogenous enhancement strategies for CCW biological treatment.The performance and potential application of exogenous enhancement strategies,including co-metabolic substrate enhancement,biofilm filler enhancement,adsorption material enhancement and conductive mediator enhancement,were expounded.Meanwhile,the enhancing mechanisms of different strategies were comprehensively discussed from a biological perspective.Furthermore,the prospects of enhancement strategies based on the engineering performance,economic cost and environmental impact(3E)evaluation were introduced.And novel enhancement strategy based on“low carbon emissions”,“resource recycling”and“water environment security”in the context of carbon neutrality was proposed.Taken together,this review provides technical reference and new direction to facilitate the regulation and optimization of typical industrial wastewater biological treatment.

By analyzing the influence of time and safety factors on the behavior strategies of vehicles on the road,a network game evolution model between drivers that considers the behavior strategies of the driving vehicle itself and its neighbors is constructed,and the competition relationship between different types of cars is studied.The influence of the proportion of driving vehicle types on the potential risk of the road is also discussed.This paper presents a guidance algorithm for vehicle dynamic behavior preference information.The correctness of the algorithm is verified by an example.Research shows:The choice of behavior strategies,such as speeding and lane changing,is related to the expected benefits of time,safety,and neighboring vehicle strategies,and the critical value of payable benefits is obtained.The higher the proportion of aggressive vehicles on the road,the greater the potential risk on the road.Whether there is a vehicle in the adjacent lane of the driving vehicle will affect the type of driving vehicle.Information guidance helps to stabilize the state of vehicles on the road,and the policy transition probability also helps stabilize the form of vehicles cars on the road.Still,information guidance has a more significant impact on the transition of vehicle types.Finally,the guidance strategy of managers is given when the road is smooth and congested.
