Production of Ergot Alkaloids by the Japanese Isolate Claviceps purpurea var. agropyri on Rice Medium

Ergot alkaloids (EAs) are secondary metabolites produced by ergot fungi (e.g., Claviceps purpurea), which are parasites of Gramineae grasses. EAs and their analogs are used to treat migraine, postpartum uterine bleeding, and Parkinson's syndrome. Recent studies have reported additional new bioactive activities of EAs and their analogs, making them essential compounds for drug development, drug repositioning, and clinical applications. EAs are produced industrially by field cultivation of ergot or liquid fermentation in the mycelial phase, but there are few published studies of the production of EAs by cereal culture and thus this approach is poorly understood. This study searched for Claviceps strains that produce EAs cultured artificially in the mycelial phase, then the selected strains were cultured on cereal media (white rice, brown rice, and rye) to examine their ability to produce EAs on each medium. C. purpurea var. agropyri produced the Clavine-type EAs pyroclavine (1), festuclavine (2), and agroclavine (3) in the mycelial phase. When cultured with white rice, brown rice, or rye, C. purpurea var. agropyri produced 1 - 3 on all cereal media. The total amount of 1 - 3 in each cereal medium (150 g of cereal per Roux flask) was 2220.5 ± 564.1 μg for white rice, 920.0 ± 463.6 μg for brown rice, and 595.4 ± 52.1 μg for rye. The white rice medium supported the highest production of 1 - 3, with the total amount of EAs (150 g of white rice per Roux flask) being about 34 times higher than that in the T25 liquid medium (190 mL per 1 L Erlenmeyer flask) (equivalent amount per flask).

Antifungal Activity of Compounds Isolated from Bamboo Vermicompost against <i>Rhizoctonia solani</i>AG1-IB

Vermicompost has become a promising alternative to chemical pesticide because of its disease suppression effect during these decades. However, the mechanisms by which vermicompost suppress plant disease are not well-understood. Antifungal compounds, which are released by beneficial organisms in the vermicompost, may play an important role in inhibiting plant pathogens;however, these mechanisms have not been widely examined. In this study, Sterilized Water Extracts of bamboo vermicompost (SWE), using a 0.22-μm cellulose acetate filter, significantly (P < 0.05) inhibited the mycelium growth of Rhizoctonia solani AG1-IB on a Potato Dextrose Agar (PDA) plate. This suggests that antifungal compounds are present in bamboo vermicompost. The ethanol acetate (EtOAc) crude extracts of bamboo vermicompost showing antifungal activity were further separated. Two compounds were isolated from the EtOAc fraction of bamboo vermicompost and characterized as ergosterol peroxide (1) and (22E, 24R)-5α,8α-epidioxyergosta-6,9(11),22-trien-3β-ol (2). Their chemical structures and mass spectra were determined by nuclear magnetic resonance and mass spectrometry analyses. Ergosterol peroxide tested at 150, 300, 600, and 900 μg showed 13%, 22%, 34%, and 53% mycelial growth inhibition against R. solani AG1-IB, respectively. Because EtOAc crude extracts of the initial substrate materials of bamboo vermicompost did not inhibit mycelium growth of R. solani AG1-IB, antifungal compounds in the vermicompost may be released by microbes but not from the original substrates during vermicomposting.

Secalonic Acid and Benzoic Acid Analogues Exhibiting Cyto toxicity against Cancer Cells Isolated from Claviceps yanagawaensis

The genus Claviceps (Clavicipitaceae) is noted for producing ergot alkaloids that cause ergotism. Claviceps yanagawaensis, a parasite of Zoysia japonica (family: Poaceae), has been isolated in Japan. Bioactivity screening showed that a methanol extract from a rice culture of C. yanagawaensis was cytotoxic to cancer cells. In our search for active substances, the new secalonic acid analogues (-)-5-epi-F-7 (1) and ergochrysin C (2) and a new benzoic acid analogue, dimethyl bigutol (3), were isolated along with the known compounds 3,4-dihydroxy-5-(3-methyl-2-buten-1-yl)benzoic acid (4) and methyl veratrate (5). The structures of 1 - 3 were elucidated by NMR, MS, and circular dichroism spectroscopy. MTT assays of 1 - 5 using cancer cell lines (HepG2, HL60, HT29, PANC-1, and T98G) showed that 1 - 4 exhibited cytotoxicity against cancer cells.

Background: The aim of the present study was to evaluate CCL8 and CXCL10 expression and its regulatory mechanism in peripheral blood leukocytes(PBLs) at the time of maternal recognition in cows. Blood samples were collected on 14, 15, 16, 17 and 18 d after artificial insemination(AI). Based on the day of return of estrus, cows were divided into three groups, pregnant(n = 5), early embryonic mortality(EEM; n = 5) and late embryonic mortality(LEM; n = 5). The gene expression levels in PBLs were assessed with quantitative real-time reverse transcription ***: The expression of CCL8 and CXCL10 mRNA in PBLs gradually increased from 14 to 18 d of pregnant cows and significant differences were observed on 18 d(P < 0.05), whereas no significant changes were observed both in EEM and LEM cows. Interferon-stimulated protein 15 k Da(ISG15), myxovirus-resistance gene(MX) 1 and MX2 mRNA expression in PBLs increased from 14 to 18 d which was significant on 18 d of pregnant cows as well as in LEM cows(P < 0.05), but no changes were observed in EEM cows. To determine whether the expression of CCL8 and CXCL10 in PBLs was regulated by pregnancy-related substances or not, expression level was assessed after exposure to interferon-τ(IFNT) and CCL16. Monocytes, granulocytes and lymphocytes were obtained using density-gradient centrifugation and flow cytometry. The addition of IFNT(100 ng/mL) and CCL16(100 ng/mL) to cultured PBLs increased the expression of CCL8 and CXCL10 mRNA(P < 0.05). The expression of ISG15, MX1 and MX2 mRNA in PBLs was also stimulated by IFNT and CCL16(P < 0.05).Conclusions: The expression of CCL8 and CXCL10 genes increased in PBLs during early pregnancy. AIM To evaluate the morphology of the colon in patients with irritable bowel syndrome(IBS) by using computed tomography colonography(CTC).METHODS Twelve patients with diarrhea type IBS(IBS-D), 13 patients with constipation type IBS(IBS-C), 12 patients with functional constipation(FC) and 14 control patients underwent colonoscopy following CTC. The lengths of the rectosigmoid colon, transverse colon and the total colon were measured. The diameters of the rectum, sigmoid colon, descending colon, transverse colon, and ascending colon were *** The mean length of the total colon was 156.5 cm in group C, 158.9 cm in group IBS-D, 172.0 cm in group IBS-C, and 188.8 cm in group FC. The total colon in group FC was significantly longer than that in group C(P < 0.05). The mean length of the rectosigmoid colon was 56.2 cm, 55.9 cm, 63.6cm, and 77.4 cm(NS). The mean length of the transverse colon was 49.9 cm, 43.1 cm, 57.0 cm, and 55.0 cm. The transverse colonin group IBS-D was significantly shorter than that in group IBS-C(P < 0.01) and that in group FC(P = 0.02). The mean diameter of the sigmoid colon was 4.0 cm, 3.3 cm, 4.2 cm, and 4.3 cm(NS). The mean diameter of the descending colon was 3.6 cm, 3.1 cm, 3.8 cm, and 4.3 cm. The descending colon diameter in group IBS-D was significantly less than that in group IBS-C(P = 0.03) and that in group FC(P < 0.001). The descending colon diameter in group FC was significantly greater than that in group C(P = 0.04). The mean diameter of the transverse colon was 4.4 cm, 3.3 cm, 4.2 cm, and 5.0 cm(NS).CONCLUSION CT colonography might contribute the clarification of subtypes of IBS.

＜正＞Control of interaction between a robot manipulator and the environment is crucial for successful execution of the practical tasks. In this paper, extended state observer （ESO）, which can estimate the dynamic process of a kind of nonlinear systems, is introduced for non-model based compensation for the nonlinear dynamics in force/motion control. The proposed strategy is illustrated via the simulation research on force/motion control of a multi-fingered robot hand.

Investigation of VEGF and PDGF signals in vascular formation by 3D culture models using mouse ES cells

Vascular formation in vivo involves several processes and signal cascades subsequently occurring in the embryo. Several models by ES cells have been reported for analysis in vitro. We show here a 3D culture system using collagen gel (AteloCell) as a simple and useful system for investigating vascular formations and analyzing the roles of factors in vivo. Although VEGF and PDGF are growth factors with multi-potentials for vascular formation, their sequential roles have not been elucidated. We investigated the effects of VEGF and PDGF B signals for vascular formation by a 3D culture system that embedded embryoid bodies (EBs) from ES cells into a collagen gel. After embedding EBs in the collagen gel with a medium containing VEGF, EBs gave off CD105 immunopositive vessels as the initial step of vasculogenesis. When the factor in the culture medium for EBs was switched from VEGF to PDGF B after 5 days of culture, the morphological features of vessels varied, suggesting the occurrence of vascular-type differentiation. After 11 days of 3D culture, vessels in both groups cultured with VEGF alone and switching to VEGF B at day 5 showed Flk-1 immunoreactivity. Some blood vessels cultured with PDGF B after day 5 expressed either EphrinB2 (arteriole marker) or Flt-4 (lymphatic marker) immunoreactivity, but vessels cultured with VEGF alone exhibited neither of them. Vessels cultured with these two factors could not differentiate into a venous type. The present study indicates that VEGF is the initial signal for vasculogenesis, and that PDGF B is probably involved in vascular diversification.
