Involvement of miR-214 and miR-375 in Malignant Features of Non-Small-Cell Lung Cancer by Down-Regulating CADM1

A tumor suppressor gene, CADM1, encoding an immunoglobulin superfamily cell adhesion molecule, is inactivated in various cancers, including non-small-cell lung cancer (NSCLC). Although promoter methylation is one of the mechanisms to suppress CADM1 expression, about half of tumors lacking CADM1 expression do not show methylation of the gene promoter. We herein investigated the possible involvement of microRNA (miRNA) in the down-regulation of CADM1. Using computational algorithms, miR-214 and miR-375 were identified as candidate miRNAs targeting CADM1. A luciferase reporter assay demonstrated that miR-214 and miR-375 repressed the promoter activity through 3’-UTR of CADM1. Quantitative RT-PCR analysis demonstrated that miR-214 and miR-375 was highly expressed in 21 (62%) and 17 cases (50%) of 34 primary NSCLCs. Notably, increased expression of miR-214 was preferentially observed in tumors with advanced pathological stages and in those lacking CADM1 expression but were not associated with the promoter methylation, suggesting that miR-214-mediated silencing would be another mechanism to suppress CADM1 expression. On the other hand, introduction of miR-214 or miR-375 into NSCLC cells decreased CADM1 protein expression. Furthermore, overexpression of miR-214 enhanced anchorage-independent growth of NSCLC cells, A549, whereas transfection of miRNA inhibitor, miR-214 or miR-375, significantly suppressed the in vitro wound healing activity of HCC827 cells. These findings suggest that overexpression of miR-214 and miR-375 could participate in the malignant features of NSCLC through down-regulating CADM1 and would provide a potential target for the treatment of a subset of NSCLC.

Biological features and biomarkers in hepatocellular carcinoma

Similar to other cancers, a multistep process of carcinogenesis is observed in hepatocellular carcinoma(HCC). Although the mechanisms underlying the development of HCC have been investigated in terms of oncology, virology, and stem cell biology, the whole picture of hepatocarcinogenesis remains to be elucidated. Recent progress in molecular biology has provided clues to the underlying cause of various diseases. In particular, sequencing technologies, such as whole genome and exome sequencing analyses, have made an impact on genomic research on a variety of cancers including HCC. Comprehensive genomic analyses have detected numerous abnormal genetic alterations, such as mutations and copy number alterations. Based on these findings, signaling pathways and cancer-related genes involved in hepatocarcinogenesis could be analyzed in detail. Simultaneously, a number of novel biomarkers, both from tissue and blood samples, have been recently reported. These biomarkers have been successfully applied to early diagnosis and prognostic prediction of patients with HCC. In this review, we focus on the recent developments in molecular cancer research on HCC and explain the biological features and novel biomarkers.

Diabetes mellitus and metformin in hepatocellular carcinoma

Hepatocellular carcinoma(HCC) is the leading cause of cancer-related death worldwide. Diabetes mellitus, a risk factor for cancer, is also globally endemic. The clinical link between these two diseases has been the subject of investigation for a century, and diabetes mellitus has been established as a risk factor for HCC. Accordingly, metformin, a first-line oral anti-diabetic, was first proposed as a candidate anti-cancer agent in 2005 in a cohort study in Scotland. Several subsequent large cohort studies and randomized controlled trials have not demonstrated significant efficacy for metformin in suppressing HCC incidence and mortality in diabetic patients; however, two recent randomized controlled trials have reported positive data for the tumor-preventive potential of metformin in non-diabetic subjects. The search for biological links between cancer and diabetes has revealed intracellular pathways that are shared by cancer and diabetes. The signal transduction mechanisms by which metformin suppresses carcinogenesis in cell lines or xenograft tissues and improves chemoresistance in cancer stem cells have also been elucidated. This review addresses the clinical and biological links between HCC and diabetes mellitus and the anti-cancer activity of metformin in clinical studies and basic experiments.

目前在土木工程,建筑,汽车等领域里使用的复合材料中,玻璃纤维是用量最大的增强材料。由于保护地球环境的呼声日趋高涨,天然纤维被期待着代替源于石油而且再利用困难的玻璃纤维,成为绿色复合材料的必要材料之一。本文通过注射成型工艺制作了玻璃短纤维,大麻短纤维以及混杂型纤维增强复合材料,并在拉伸实验中应用两种不同频率的声发射技术检测了拉伸断裂特性。实验发现,随着大麻纤维的加入和混杂复合材料绿色度的增加,复合材料的拉伸弹性模量随之线性增大,而拉伸强度基本保持不变。当大麻纤维的含量超过20wt%的时候,拉伸强度开始降低。在不同频率的声发射实验中,混杂型复合材料的声发射事件的产生都比单一纤维增强型复合材料要来的晚,也就是说,纤维的混杂有助于推迟微裂纹的产生。

Long-term follow-up of ulcerative colitis patients treated on the basis of their cytomegalovirus antigen status

AIM:To clarify the impact of cytomegalovirus(CMV)activation and antiviral therapy based on CMV antigen status on the long-term clinical course of ulcerative colitis(UC)***:UC patients with flare-up were divided into CMV-positive and-negative groups according to the CMV antigenemia *** main treatment strategy provided for the patients in the CMV-positive group comprised a dose reduction of corticosteroids and administration of ***:The median number of days to initial remission was significantly greater for the patients in the CMV-positive group(21 d vs 16 d,P=0.009).However,the relapse rate after remission and colectomy rate during more than 30 mo of observation did not differ between the two *** analysis revealed that administration of ganciclovir was the only independent factor for avoiding colectomy in patients of the CMV-positive ***:CMV antigen status did not significantly affect the long-term prognosis in UC patients under treatment with appropriate antiviral therapy.

Risk factors for the recurrence of hepatocellular carcinoma after radiofrequency ablation of hepatocellular carcinoma in patients with hepatitis C

AIM: To analyze the risk factors of hepatocellular carcinoma (HCC) recurrence after radiofrequency ablation (RFA) treatment with HCV-associated hepatitis. METHODS: Twenty-six patients with HCV-associated HCC who were followed-up for more than 12 mo were selected for this study. Risk factors for distant intrahepatic recurrences of HCC were evaluated for patients in whom complete coagulation was achieved without recurrence in the same subsegment as the primary nodule. Twelve clinical and tumoral factors were examined: Age, gender, nodule diameter, number of primary HCC nodule, Child-Pugh classification, serum platelet, serum albumin, serum AST, post RFA AST, serum ALT, post RFA ALT, post RFA ***: Distant recurrences of HCC in remnant liver after RFA were observed in 14 cases and in the number of primary HCC nodules (P = 0.047), and the serum platelets (P = 0.030), the clear difference came out by the recurrence group and the non-recurrence group. The cumulative recurrence rates after 1 and 2 years were30.8% and 86.8%, respectively for primary multinodular HCC, and 15.4% and 29.5% respectively, for primary uninodular HCC. In addition the 1-year recurrence rates for patients with serum albumin more than 3.4 g/dL and less than 3.4 g/dL were 23.1% for both, but the 2-years recurrence rates were 89.0% and 23.1%, respectively. The number of primary HCC nodules (relative risk, 6.970; P = 0.016) were found to be a statistically significant predictor for poor distant intrahepatic recurrence by univariate ***: Patients who have multiple HCC nodules, low serum platelets and low serum albumin accompanied by HCV infection, should be carefully followed because of the high incidence of new HCC lesions in the remnant liver, even if coagulation RFA is complete.

Implementation and Evaluation of a Postpartum Care Program for Mothers Raising Infants: A Pilot Study

Background: The aims of this pilot study were to implement and evaluate a postpartum care program for mothers raising children younger than one year to provide physical relaxation, and to reduce mothers’ parenting problems. Methods: A day-service and group-type postpartum care program was implemented with the aim of interacting with peers, promoting local communication, and relaxing mothers. Results: Forty-five pairs of mothers and infants participated in the study. Most of the mothers experienced relaxation, communicated with each other, resolved physical problems, reduced childcare concerns, and developed parenting friends. Conclusions: In addition, feasibility was assured as they were very satisfied with the content of programs such as baby massage and aroma treatment.

Hypertensive cardiomyopathy:A clinical approach and literature review

Hypertensive cardiomyopathy(HTN-CM) is a structural cardiac disorder generally accompanied by concentric left ventricular hypertrophy(LVH) associated with diastolic or systolic dysfunction in patients with persistent systemic *** occurs in the absence of other cardiac diseases capable of causing myocardial hypertrophy or cardiac *** systemic hypertension leads to structural and functional myocardial abnormalities resulting in myocardial ischemia,fibrosis,and ***-CM is predominantly a disease of impaired relaxation rather than impaired contractility,so patients are usually asymptomatic during resting ***,their stiff left ventricles become incapable of handling increased blood volume and cannot produce appropriate cardiac output with the slight change of circulating volume that may occur during ***,the accompanying LVH is itself a risk factor for mortality and ***,early detection of LVH development in patients with hypertension(referred to as HTN-CM) is critical for optimal *** addition to pathological findings,echocardiography and cardiac magnetic resonance imaging are ideal tools for the diagnosis of *** diagnosis of this condition and utilization of appropriate treatment are required to improve morbidity and mortality in hypertensive *** review article presents an overview of the multidimensional impact of myocardial disorder in patients with *** literature is highlighted and the effects of hypertension on cardiac hypertrophy and heart failure development are discussed,including possible therapeutic options.

How Do Weekly Magazines Provide Information on Urogenital Cancer to the Public in Aged Societies?

Little information on cancer coverage rather than newspaper and television is available. Japanese weekly magazines have a circulation of over 2,700,000 per week. To examine how they delivered urogenital cancer information to the public, cancer-related articles and advertisements in six major Japanese weekly magazines from 2009 to 2010 was analyzed. 1.8% of total articles and advertisements were cancer-related. Prostate cancer (n = 119) was the second-most common topic, following lung cancer (n = 145), whereas only three articles were published on kidney or bladder cancer. The 53 articles on therapies for prostate cancer comprised radiotherapy (n = 29), surgery (n = 16), chemotherapy (n = 4), and others (n = 4). All 42 comments or interviews were cited in the article on prostate cancer, while 26 of them were attributed to only two famous doctors. Although cancer coverage in weekly magazines could be useful to spread information on prostate cancer, we should recognize their considerable bias based on a disproportionate emphasis.

Inhibitory Activity of Paenibacillussp. Isolated from Soil in Gotsu City, Shimane Prefecture, Against Xanthomas oryzaepv. oryzae, the Causal Agent of Rice Bacterial Leaf Blight

Microorganism isolates (n = 49) were obtained from the soil samples collected from field in Gotsu city (Kawahira), Shimane. Isolate GT2-E culture inhibited the growth of Xanthomonas oryzae pv. oryzae in disk diffusion method. Rice bacterial leaf blight was suppressed by GT2-E culture in the pre- and post-treated rice leaves. Sequence analysis of 16S rDNA region of the GT2-E isolate indicated that it shared 99% similarity with Paenibacillus polymyxa. The growth of GT2-E on LB medium was observed at 15°C, 28°C, 37°C, and 45°C, but not at 4°C. GT2-E isolate could be grown even in the presence of agrochemicals (Amister, Blasin and Kasumin). Furthermore, the growth of X. oryzae pv. oryzae was inhibited by the culture filtrate of GT2-E isolate in disk diffusion method. However, the inhibitory activity of the culture filtrate was heat-unstable. This result suggested that GT2-E isolate can produce heat-unstable inhibitory compound(s). In conclusion, GT2-E isolate might contribute to the development of a new bactericide and biological agent against rice bacterial leaf blight.
