Use of Cryopreserved Osteogenic Matrix Cell Sheets for Bone Reconstruction

Skeletal diseases, such as nonunion and osteonecrosis, are now treatable with tissue engineering techniques. Single cell sheets called osteogenic matrix cell sheets (OMCSs) grown from cultured bone marrow-derived mesenchymal stem cells show high osteogenic potential;however, long preparation times currently limit their clinical application. Here, we report a cryopreservation OMCS transplantation method that shortens OMCS preparation time. Cryopreserved rat OMCSs were prepared using slow- and rapid-freezing methods, thawed, and subsequently injected scaffold-free into subcutaneous sites. Rapid- and slow-frozen OMCSs were also transplanted directly to the femur bone at sites of injury. Slow-freezing resulted in higher cell viability than rapid freezing, yet all two cryopreservation methods yielded OMCSs that survived and formed bone tissue. In the rapid- and slow-freezing groups, cortical gaps were repaired and bone continuity was observed within 6 weeks of OMCS transplantation. Moreover, while no significant difference was found in osteocalcin expression between the three experimental groups, the biomechanical strength of femurs treated with slow-frozen OMCSs was significantly greater than those of non-transplant at 6 weeks post-injury. Collectively, these data suggest that slow-frozen OMCSs have superior osteogenic potential and are better suited to produce a mineralized matrix and repair sites of bone injury.

A Minimalist Account of Wh-Movement in Fulfulde

Prediction of CO<sub>2</sub>and H<sub>2</sub>S Emissions from Wastewater Wet Wells

The transportation of wastewater through sewer networks can cause potential problems due to the formation of lethal gases such as ammonia, carbon dioxide, hydrogen sulphide and methane. As a result, they can cause irritating, toxic, and microbial induced corrosion. The true depth of these problems can be measured by measuring the emission of such lethal gasses in the sewer atmosphere. The amount of gases released from such sewer networks has to be controlled. In this paper, we present a simple experimental methodology used to determine the emissions of Carbon Dioxide (CO2) and Hydrogen Sulphide (H2S) using Landtec GEM-2000 plus multi-gas analyser from wastewater wet wells.

Growing Public Health Concerns from Poor Urban Air Quality: Strategies for Sustainable Urban Living

Urban areas around the world, particularly in emerging nations such as China, India, and Brazil are experiencing high levels of air pollution due to increased population, economy, spending, and consumption, all of which contribute to deterioration in environmental and public health conditions in urban areas. This paper briefly discusses important sources of air pollution, air pollutants of concern, public health impacts, and proposed strategies to combat urban air pollution and promote sustainable urban living. A team of researchers under the mentorship of the main author is working on a number of air quality projects that involve air quality monitoring (sources, ambient, indoor, and occupational), emissions modeling, atmospheric dispersion modeling, air pollution control, and development of knowledge-based systems to manage air quality.? This paper presents potential strategies that could help address the growing public health concerns in urban areas and promote sustainable and healthy living.

针对窄带相机拍摄的作物冠层图像,本文开发了一种图像处理算法,可以去除背景干扰,更加真实地获取作物冠层的NDVI值,实际使用时在标准工业RGB相机的镜头上加装窄带滤光片以获取窄带图像。在自然光照条件下,拍摄了青椒苗在窄带红光(660nm±10nm)和窄带近红外(770nm±15nm)两个波段的照片。为了使两幅窄带图像的像素位置能够精确对应,以简化图像处理过程,实验过程中用单个相机切换滤光片的方式,拍摄不同波段的窄带图像(植物保持在相同的位置),从而使后继图像处理更加高效。在本研究中,通过对窄带图像中冠层与背景的强烈对比特性,开发了一种背景去除方法,通过对获取的窄带图像进行处理,识别背景中不需要的噪声和不必要的信息被有效滤除,获取了感兴趣区域(ROI),并最终计算出作物的NDVI。在该研究中发现窄带波长770nm与660nm的白板成像值之比为定值(定义为Cw),且该值与光照强度和拍摄位置无关,从而构建了一种计算NDVI的新方法。在实验过程中,在不同的自然光照条件下,拍摄了从30cm到70cm不同高度的图像,根据窄带图像中冠层与背景的强烈对比特征,该算法有效地实现了作物冠层与土壤背景的分离。本文建立一套实验装置,对生长三周的辣椒幼苗冠层进行了 NDVI测试。实验结果表明,在室外自然光下,测量高度变化时可以得到稳定的NDVI值。该算法在没有背景干扰的情况下,获得了 NDVI值,为作物冠层NDVI的研究提供了新的思路。通过对所获得的数据进行了比较处理,证明了 NDVI的稳定性,并说明了所开发模型的相关性、可靠性以及它的实用性。从实验结果可以看出,Cw的平均标准偏差为0.1979%,表明在测量高度变化时Cw值非常稳定,这证明Cw基本接近常数。相应地,当测量高度变化时,NDVI相对误差的平均标准偏差为2.339%,表明NDVI的计算模型是可靠的。实验过程中,当相机镜头与植物冠层高度差为30-60cm时,NDVI计算模型的值相对较为稳定和可靠,这是由于当高度差为70cm时,相机的视野中背景的比例较大,使得该高度下图像比其他高度的图像有更多的干扰,使得该模型的偏差最高。

Promoting Sustainability at the Ports through Knowledge Sharing

Ports are a major source of economic activity and environmental pollution. They use a variety of equipment for loading/unloading and movement of cargo, such as marine vessels, cranes, trucks and railroads. These activities can cause large amounts of pollution to the air, water and soil media. Despite the pollution generated, it also serves as a medium for tourism with its cruise liners. This paper discusses port activities and how it effects the environment. The main focus of this paper is Knowledge Sharing for Sustainability of Ports. Sustainability indicators should help in evaluating port conditions and this knowledge sharing among the proposed 10 major world ports should accelerate economic growth and use of latest technologies for pollution reduction.

Hypo-Anxious Phenotype of Adolescent Offspring Prenatally Exposed to LPS Is Associated with Reduced mGluR5 Expression in Hippocampus

Many studies have reported long-term modulation of metabotropic glutamate receptor 5 (mGluR5) by inflammatory processes and a pharmacological modulation of mGluR5 is known to regulate anxiety level. However, it is not known if non-pharmacological modulation of mGluR5 by inflammation impaired the unconditional level of anxiety. In this study, we investigated this relation in LPS prenatal immune challenge (120 μg/kg, 3x i.p. injection in late gestation), a developmental model of neuroinflammation in which some studies have reported hypo-anxious phenotype. Using positron emission tomographic imaging (PET) approaches, we have demonstrated a decrease in the binding potential of [18F]fluoro-5-(2-pyridinylethynyl)benzonitrile([18F]FPEB, a radioligand for mGluR5) in hippocampus of adolescent offspring prenatally exposed to LPS, without significant change in the binding of [11C]peripheral benzodiazepine receptor 28 ([11C]PBR28), an inflammatory marker. In addition, dark-light box emergence test revealed a lower level of anxiety in LPS-exposed offspring and this behavioural phenotype was associated with the binding potential of [18F]FPEB in hippocampus. These results confirm that neuroinflammation during developmental phase modulates the physiology of mGluR5 and this alteration can be associated with behavioural phenotype related to anxiety. In addition, this study supports a hypotheses that mGluR5 could be used as a diagnostic target in anxiety.
