Inverse relationship between platelet Akt activity and hippocampal atrophy:A pilot case-control study in patients with diabetes mellitus

BACKGROUND Akt plays diverse roles in humans.It is involved in the pathogenesis of type 2 diabetes mellitus(T2DM),which is caused by insulin resistance.Akt also plays a vital role in human platelet activation.Furthermore,the hippocampus is closely associated with memory and learning,and a decrease in hippocampal volume is reportedly associated with an insulin-resistant phenotype in T2DM patients without dementia.AIM To investigate the relationship between Akt phosphorylation in unstimulated platelets and the hippocampal volume in T2DM patients.METHODS Platelet-rich plasma(PRP)was prepared from the venous blood of patients with T2DM or age-matched controls.The pellet lysate of the centrifuged PRP was subjected to western blotting to analyse the phosphorylation of Akt,p38 mitogen-activated protein(MAP)kinase and glyceraldehyde 3-phosphate dehydrogenase(GAPDH).Phosphorylation levels were quantified by densitometric analysis.Hippocampal volume was analysed using a voxel-based specific regional analysis system for Alzheimer’s disease on magnetic resonance imaging,which proposes the Z-score as a parameter that reflects hippocampal volume.RESULTS The levels of phosphorylated Akt corrected with phosphorylated p38 MAP kinase were inversely correlated with the Z-scores in the T2DM subjects,whereas the levels of phosphorylated Akt corrected with GAPDH were not.However,this relationship was not observed in the control patients.CONCLUSION These results suggest that an inverse relationship may exist between platelet Akt activation and hippocampal atrophy in T2DM patients.Our findings provide insight into the molecular mechanisms underlying T2DM hippocampal atrophy.

BACKGROUND Neuropathy may occur at some sites after catheterization for close examination of cardiac disease.Although the radial artery is considered a relatively uncomplicated site for catheterization,the radial artery and median nerve are in relatively close proximity,with the risk of median nerve injury depending on the angle of puncture.The purpose of this study was to report the outcomes of surgery performed for conservative therapy-resistant median neuropathy following forearm catheterization.CASE SUMMARY A 50-year-old woman experienced palsy from the right thumb to the radial side of the ring finger after catheterization from the radial artery of the right forearm.Paresthesia developed at the same site and a positive tinel-like sign was seen for the median nerve area at the high level of the puncture site.Nerve conduction study showed reduced compound muscle action potentials and loss of sensory nerve action potentials.Symptoms did not improve despite pharmacotherapy and the patient gradually developed flexion restrictions of the index and middle fingers.Median nerve injury and associated flexor tendon adhesion was diagnosed,and the patient was referred for surgery at 3 mo after injury.When the same area was opened,no injury to the median nerve epithelium was obvious,but the area of the positive tinel-like sign was highly adherent to surrounding tissue and to the flexor digitorum superficialis of the index and middle fingers.The surgery was terminated with adequate adhesion release.Rehabilitation was initiated postoperatively,improving neurological symptoms and range of motion of the fingers.Six months after surgery,the patient returned to daily activities without discomfort.CONCLUSION This case provides the appropriate diagnosis and treatment for a suspected peripheral nerve injury.

After field cooling(FC)alternating current poling(ACP),we investigated the dielectric and piezoelectric properties of[001]_(pc)-oriented 0.24Pb(In_(1/2)Nb_(1/2))O_(3)(PIN)-0.46Pb(Mg_(1/3)Nb_(2/3))O_(3)(PMN)-0.30PbTiO3(PT)(PIMN-0.30PT)single crystals(SCs),which were manufactured by continuous-feeding Bridgman(CF BM)within morphotropic phase boundary(MPB)region.By ACP with 4 kVrms/cm from 100 to 70℃,the PIMN-0.30PT SC attained high dielectric permittivity of 8330,piezoelectric coefficient(d_(33))of 2750 pC/N,bar mode electromechanical coupling factorκ_(33)of 0.96 with higher phase change temperature(T_(pc))of 103℃,and high Curie temperature(7c)of 180℃.These values are the highest ever reported as PIMN-xPT SC system with Tpc>100℃.The enhancement of these properties is attributed to the induced low symmetry multi-phase supported by phase analysis.This work indicates that FC ACP is a smart and promising method to enhance piezoelectric properties of relaxor-PT ferroelectric SCs including PIMN-xPT,and provides a route to a wide range of piezoelectric device applications.

The effects of alternating current poling(ACP)at 80℃on electrical properties of[001]-oriented 0.72 Pb(Mg_(1/3)Nb_(2/3))O_(3)-0.28PbTiO_(3)(PMN-28PT)single crystals(SCs)have been investigated.The square-wave ACP SCs poled at high voltage(HV,5 kV_(rms)/cm)occasionally showed large fluctuations and low opposite values of piezoelectric coefficient(d_(33)=±1370 pC/N)in one plate.This revealed spuriousmode vibrations(SMV)of impedance spectrum.However,after depolarizing and repolarizing the sample with a sine-wave ACP at low voltage(LV,3.5 kV_(rms)/cm),the d_(33) enhanced to be 1720 pC/N(+26%)and did not exhibit large fluctuation or opposite values in one plate any more.The impedance spectrum became clean and the abnormal SMV disappeared.We proposed four possible mechanisms of the SMV,and speculate that the main cause maybe by macro-scale sub-domain structure and/or phase change in the main domain structure and/or phase in the SC plate due to the specific poling conditions not eternal mechanical damage of PMN-PT SCs.This study will be useful to realize a high d_(33) and improve other properties of PMN-PT ACP SC ultrasonic transducers without any SMV for high-frequency medical imaging equipment.

Gene expression analyses suggest that more than 1000–2000 genes are expressed predominantly in mouse and human testes.Although functional analyses of hundreds of these genes have been performed,there are still many testis-enriched genes whose functions remain unexplored.Analyzing gene function using knockout(KO)mice is a powerful tool to discern if the gene of interest is essential for sperm formation,function,and male fertility in vivo.In this study,we generated KO mice for 12 testis-enriched genes,1700057G04Rik,4921539E11Rik,4930558C23Rik,Cby2,Ldhal6b,Rasef,Slc25a2,Slc25a41,Smim8,Smim9,Tmem210,and Tomm20l,using the clustered regularly interspaced short palindromic repeats/CRISPR-associated protein 9(CRISPR/Cas9)system.We designed two gRNAs for each gene to excise almost all the protein-coding regions to ensure that the deletions in these genes result in a null mutation.Mating tests of KO mice reveal that these 12 genes are not essential for male fertility,at least when individually ablated,and not together with other potentially compensatory paralogous genes.Our results could prevent other laboratories from expending duplicative effort generating KO mice,for which no apparent phenotype exists.

Operations research(OR)is a scientific approach for solving various types of societal problems and making decisions to deal with these problems appropriately.First,we briefly describe the history of OR,focusing on applying OR to public sectors,and then provide its characterization as a scientific method for decision making.Then,OR activities in Japan are introduced,emphasizing three major roles:(i)quantitative data analysis,(ii)mathematical modeling analysis,and(iii)theory building analysis.We provide an example for each of these three types of major roles.Based upon the analyses,we seek an Asian OR applicable to policy making in the public sector.Finally,we provide a summary and discuss future perspectives for OR.

There is a need for the prevention of upper extremity injuries that affect a large number of competitive baseball players.Currently available evidence alludes to three possible ways to prevent these injuries:1) regulation of unsafe participation factors,2) implementation of exercise intervention to modify suboptimal physical characteristics,and 3) instructional intervention to correct improper pitching techniques.Of these three strategies,instruction of proper pitching technique is under-explored as a method of injury prevention.Therefore,the purpose of this review was to explore the utility of pitching technique instruction in prevention of pitching-related upper extremity injuries by presenting evidence linking pitching technique and pitching-related upper extremity injuries,as well as identifying considerations and potential barriers in pursuing this approach to prevent injuries.Various kinematic parameters measured using laboratory-based motion capture system have been linked to excessive joint loading,and thus pitching-related upper extremity injuries.As we gain more knowledge about the influence of pitching kinematics on joint loading and injury risk,it is important to start exploring ways to modify pitching technique through instruction and feedback while considering the specific skill components to address,mode of instruction,target population,duration of program,and ways to effectively collaborate with coaches and parents.

Strict intraoperative glycemic control can significantly decrease the incidence of postoperative infection; however, anesthesiologists must carefully control blood glucose levels as well as properly manage the respiratory and cardiovascular systems. However, standard blood glucose measurement systems and insulin dosing algorithms, which are necessary for achieving strict glycemic control, have not yet been developed. An artificial pancreas (STG-22TM; Nikkiso Co., Tokyo, Japan) is considered a highly accurate blood glucose monitoring system capable of closed-loop control of blood glucose. The device has, however, many problems to be addressed since it is a large and expensive system with little versatility, and it requires a large amount of blood to be collected. Therefore, the development of less invasive and inexpensive systems with future technological progress is greatly anticipated.

Ocean thermal energy conversion(OTEC)is a process of generating electricity by exploiting the temperature difference between warm surface seawater and cold deep seawater.Due to the high static and dynamic pressures that are caused by seawater circulation,the stiffened panel that constitutes a seawater tank may undergo a reduction in ultimate strength.The current paper investigates the design of stiffening systems for OTEC seawater tanks by examining the effects of stiffening parameters such as stiffener sizes and span-over-bay ratio for the applied combined loadings of lateral and transverse pressure by fluid motion and axial compression due to global bending moment.The ultimate strength calculation was conducted by using the non-linear finite element method via the commercial software known as ABAQUS.The stress and deformation distribution due to pressure loads was computed in the first step and then brought to the second step,in which the axial compression was applied.The effects of pressure on the ultimate strength of the stiffener were investigated for representative stiffened panels,and the significance of the stiffener parameters was assessed by using the sensitivity analysis method.As a result,the ultimate strength was reduced by approximately 1.5%for the span-over-bay ratio of 3 and by 7%for the span-over-bay ratio of 6.
